Microvasculopathy in spinal muscular atrophy: The endothelium as a target of SMN deficiency and an amplifier of disease.

Spinal muscular atrophy (SMA) is caused by deficiency of the ubiquitous survival motor neuron (SMN) protein. Degeneration of anterior horn motor neurons defines the disease, but many organs are affected, including heart, liver, kidney, and bone. The vascular compartment is among them: digital necrosis in infants with the fewest SMN2 copies, capillary rarefaction in SMA mice, and decreased von Willebrand factor with fibrinogen extravasation in human post-mortem spinal cord. Whether these reflect endothelial injury by SMN deficiency itself or capillary adaptation to atrophic, denervated tissue h

Interleukin-6 is critical in the development of pulmonary vascular disease in Gcn2-deficient mice.

Biallelic mutations in EIF2AK4, encoding Eukaryotic Translation Initiation Factor 2α kinase 4 or General Control Nonderepressible 2 (GCN2), cause pulmonary veno-occlusive disease (PVOD), a fatal form of pulmonary hypertension. The mechanisms linking GCN2 deficiency with pulmonary vascular pathology are poorly understood. To investigate this, we developed two mouse models: genetic ablation of Gcn2, to mirror GCN2-mutation positive PVOD, and a pharmacological model using mitomycin C, a drug which can cause PVOD as an idiosyncratic drug reaction. Both models were phenotyped, and lungs from wild-t

Dopamine-Deficiency-Related Reorganization of the Somatomotor Network in Prodromal and Early Parkinson's Disease.

INTRODUCTION: Network attack tolerance (NAT) measures the brain's ability to sustain information flow despite the loss of critical brain regions. In Parkinson's disease (PD), NAT has been linked to cognitive and motor function. However, it remains unclear how dopaminergic degeneration shapes NAT, and whether it relates to motor symptom progression. OBJECTIVE: To examine dopamine deficiency-related changes in NAT across the early PD disease continuum and its association with longitudinal motor outcomes. METHODS: We used cross-sectional resting-state functional magnetic resonance imaging of 28

ATOX1 deficiency induces memory impairment via promoting cuproptosis in Alzheimer's disease.

INTRODUCTION: Copper homeostasis disturbance has been implicated in Alzheimer's disease (AD), and excess copper exacerbates oxidative damage, protein aggregation and cognitive deficits. Cuproptosis is a new form of cell death mainly related to mitochondrial impairment which caused by intracellular copper overload. However, the involvement of cuproptosis in the pathogenesis of AD remains elusive. OBJECTIVES: The copper chaperone ATOX1 is crucial for copper homeostasis. This study aimed to investigate the role of ATOX1 and cuproptosis in the progression of Alzheimer's disease and to identify th

UQCRC1 deficiency impairs mitophagy via PINK1-dependent mechanisms in Parkinson's disease.

Oxidative phosphorylation (OXPHOS) and mitophagy are functionally interconnected cellular processes, the defects of which are considered key driving forces behind the pathogenesis of Parkinson's disease (PD). UQCRC1, a core subunit of the mitochondrial respiratory chain complex III, is a recently identified familial PD gene whose pathogenic mutations result in OXPHOS stress. Given its importance, however, the role of UQCRC1 in idiopathic PD as well as mitophagy has not been investigated. In this study, we collected 19 datasets comprising postmortem substantia nigra from 150 cases of non-diseas

Poldip2 deficiency attenuates lung disease severity in a mouse model of COVID-19.

The lungs are the primary target of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), with the infection resulting in lung inflammation, pulmonary vascular leakage and diffuse alveolar damage. Polymerase delta-interacting protein-2 (Poldip2) mediates lung inflammation and vascular permeability after lipopolysaccharide-induced acute respiratory distress syndrome; however, whether it also affects the pathological consequences of SARS-CoV-2 infection is completely unknown. Here, we assessed the role of Poldip2 in inflammation, immune cell infiltration and lung tissue damage in respons

Complete and partial forms of X-linked MCTS1 deficiency in patients with mycobacterial disease.

X-linked recessive (XR) complete MCTS1 deficiency underlies Mendelian susceptibility to mycobacterial disease (MSMD) in patients with bacille Calmette-Guérin (BCG) disease. We investigated the genotypic and phenotypic landscape of four new unrelated families from four distinct countries. Three patients had adverse reactions to the BCG vaccine, whereas another patient was not vaccinated with BCG and had an infection with Mycobacterium abscessus at 16 years of age. Whole-exome sequencing of the probands revealed hemizygosity for rare germline MCTS1 variants. In addition to a previously reported

REDD1 deficiency alleviates podocyte PANoptosis and restores autophagy in diabetic kidney disease.

BACKGROUND: Podocyte loss and death are pathological hallmarks of diabetic kidney disease (DKD), and PANoptosis (apoptosis, pyroptosis, and necroptosis) in podocytes is crucial to DKD progression. Regulated in development and DNA damage response 1 (REDD1) is a multifaceted regulator involved in metabolism, oxidative stress, autophagy, and cell fate. In this study, we aimed to investigate the effects and underlying mechanisms of REDD1 on podocyte PANoptosis and autophagy in DKD. METHODS: REDD1 knockout (KO) mice were induced to diabetes by intraperitoneal injections of streptozotocin (STZ). We

[French nephrologists’ practices in the management of anemia and iron deficiency in chronic kidney disease].

BACKGROUND: Real-world practices for managing anemia and iron deficiency in chronic kidney disease appear heterogeneous and sometimes controversial. This study aimed to describe the prescribing preferences and habits of French nephrologists in this area. METHODS: All nephrologists seeing patients at one of the 40 centers participating in the Chronic Kidney Disease – Renal Epidemiology and Information Network (CKD-REIN) cohort were invited to participate in two waves (2015–2016 and 2019–2020) of a practice survey. The self-administered questionnaires collected information on nephrologists’ cha

Repurposing Gaucher disease therapy for Saposin C deficiency: Proof-of-concept with eliglustat.

Saposin C (Sap C) deficiency (GDSAPC, OMIM #610539, ORPHA:309252) is an ultra-rare autosomal recessive disorder caused by mutations in the PSAP gene. Sap C functions as an essential activating cofactor of glucosylceramidase (GCase) and facilitates the degradation of glucosylceramide in the lysosome. In the absence of its activator, GCase is structurally intact but its function is impaired, leading to pathological accumulation of glucosylceramide in the lysosome. Sap C deficiency resembles another rare autosomal recessive disorder, Gaucher disease (GD), in which pathogenic variants in the GBA1

Relationship between elevated arginine and pathophysiology of progressive disease in arginase 1 deficiency.

BACKGROUND: Arginase 1 deficiency (ARG1-D; OMIM #207800) is a rare urea cycle disorder caused by loss of arginase activity, preventing the hydrolysis of arginine to ornithine and urea. The resulting hyperargininaemia leads to accumulation of neurotoxic metabolites including guanidino compounds and nitric oxide in the central nervous system. Nervous system structures are affected, including the corticospinal tracts, cerebral and cerebellar white matter, basal ganglia, hippocampus, and corpus callosum. Clinically, patients present with developmental and neurological impairment, including growth

Acquired vitamin B6 deficiency in POEMS: a marker of disease activity and cause of neuropathy?

BACKGROUND: POEMS syndrome is a rare, life-threatening multisystem disease driven by clonal plasma cell proliferation and characterised by disabling neuropathy, a monoclonal plasma cell disorder and diagnostic paraneoplastic features. Although vascular endothelial growth factor (VEGF) is a key diagnostic marker and correlates with disease activity, it is not the central mediator and the specific pathomechanism for the characteristic distal sensorimotor neuropathy remains elusive. Pyridoxal 5'-phosphate (PLP), the active form of vitamin B6, is an essential cofactor for human biological processe

GPR55 deficiency exacerbates cognitive impairments and Alzheimer's disease-like pathology in mice.

BACKGROUND: Alzheimer's disease (AD) is the most common type of dementia, characterized by progressive cognitive decline and neuronal damage. Although studies have indicated a link between G-protein coupled receptor 55 (GPR55) and AD-related cognitive impairment, the underlying mechanisms remain unclear. Here, we aim to further investigate the role of GPR55 in the pathogenesis of AD. METHODS: We used viral vectors to knock down GPR55 expression in the hippocampus of normal mice. We also generated GPR55 knockout in AD mice by crossing GPR55-/- mice with APP/PS1 transgenic mice (APP/PS1; GPR55-

Microglial SIRT2 deficiency aggravates cognitive decline and amyloid pathology in Alzheimer's disease.

Sirtuin 2 (SIRT2), a NAD+-dependent deacetylase, has been implicated in aging and neurodegenerative diseases such as Alzheimer's disease (AD). While global SIRT2 inhibition has shown promise in reducing amyloid-beta pathology and cognitive deficits in different mouse models of AD, peripheral SIRT2 inhibition has been associated with adverse effects, such as increased inflammation. This suggests that targeted inhibition of specific cellular populations within the brain may represent a more precise and effective approach for the treatment of AD. To explore this hypothesis, we generated a conditi

Carnitine Deficiency in Chronic Kidney Disease: Pathophysiology, Clinical Implications, and Therapeutic Perspectives.

Carnitine is essential for the mitochondrial transport of long-chain fatty acids and thus plays a pivotal role in energy metabolism, particularly in metabolically active organs, such as skeletal and cardiac muscle. In patients with dialysis, carnitine homeostasis is disrupted because of the reduced synthesis, impaired renal reabsorption, and carnitine loss during extracorporeal procedures. Carnitine deficiency is linked to a wide range of clinical manifestations, including muscle weakness, treatment-resistant anemia, intradialytic hypotension, mental disorder, and cardiovascular disease. This

Lysosomal acid lipase deficiency: The forgotten link between liver and cardiovascular disease.

Lysosomal acid lipase-deficiency (LAL-D) is a rare and systemic condition, secondary to lipase A gene mutations, responsible for lysosomal accumulation of cholesteryl esters and triglycerides in many tissues. It is a very heterogeneous disease in terms of the age of onset, severity, and the type of clinical and radiological manifestations. Dyslipidemia, hepatomegaly, and hepatosteatosis with increased levels of transaminases are the most common features. In association with liver dysfunction and evolution to cirrhosis, there is an increased risk of premature atherosclerosis and cardiovascular

Developmental origins of disease - Effects of iron deficiency in the rat developing kidney and beyond.

Iron is an essential cofactor in metabolic and developmental processes. Iron deficiency (ID) is the most common micronutrient deficiency in pregnancy, especially impacting medically underserved populations worldwide. Iron deficiency (ID) in pregnancy predisposes neonates to poor iron status, i.e., congenital ID and associated adverse effects. The role of congenital ID on human kidney development is unstudied, but impaired fetal kidney development is possible. Both vascular and global nutrient restriction rat models report impaired fetal kidney development, as well as induce hypertension, suppo

Association Between G6PD Deficiency and Congenital Heart Disease Incidence and Hospital Outcomes.

G6PD deficiency classically presents with hemolytic anemia, but associations have been described with atherogenesis and coronary artery disease secondary to increased oxidative stress. There are limited data that also suggest a link between G6PD deficiency and the development of congenital heart disease (CHD), possibly through the same mechanism. We hypothesize a higher incidence of CHD among individuals with G6PD deficiency as well as worse hospital outcomes for those with CHD and G6PD deficiency. We performed a retrospective review of a national administrative database from 10/2019 to 3/2024
