Sex differences in hippocampal β-adrenergic receptor subtypes drive retrieval, retention, and learning of cocaine-associated memories.

BACKGROUND: Drug seeking behavior occurs in response to environmental contexts and drug-associated cues. The presence of these pervasive stimuli impedes abstinence success. β-adrenergic receptors (β-ARs) have a long-standing historical implication in driving processes associated with contextual memories, including drug-associated memories in substance use disorders. However, sex differences in the role of β-adrenergic receptors in drug memories remain unknown. HYPOTHESIS: Prior reports indicate a selective role for β2-ARs in retrieval and retention of contextual drug memories in males, and su

Neural Correlates of Opioid-Induced Risk-Taking Behavior in the Prelimbic Prefrontal Cortex.

Opioid use disorder occurs alongside impaired risk-related decision-making, but the underlying neural correlates are unclear. We developed an approach-avoidance conflict task using a modified conditioned place preference (CPP) procedure to study neural signals of risky opioid seeking in the prefrontal cortex, a region implicated in executive decision-making. Following morphine CPP, rats underwent a conflict test in which fear-inducing cat odor was introduced in the previously drug-paired side of the apparatus. While the saline-exposed control group avoided cat odor, the morphine group included

A Neurobehavioral Approach to Addiction: Implications for the Opioid Epidemic and the Psychology of Addiction.

Two major questions about addictive behaviors need to be explained by any worthwhile neurobiological theory. First, why do people seek drugs in the first place? Second, why do some people who use drugs seem to eventually become unable to resist drug temptation and so become "addicted"? We will review the theories of addiction that address negative-reinforcement views of drug use (i.e., taking opioids to alleviate distress or withdrawal), positive-reinforcement views (i.e., taking drugs for euphoria), habit views (i.e., growth of automatic drug-use routines), incentive-sensitization views (i.e.

Early signal of diverted use of tropicamide eye drops in France.

AIMS: Tropicamide is a mydriatic drug used as eye-drops for diagnostic or therapeutic purposes. From 2013, a diverted use by intravenous route has been suspected in Eastern Europe in opioids users. To date, no signal of misuse has been identified in France. The aims of this study were to investigate any early signals of a diverted use of tropicamide eye drops and to collect information regarding motives for the misuse and tropicamide-induced effects. METHODS: Information was obtained at three levels: (1) at regional level (Midi-Pyrénées area), from reimbursement data and pharmacists' reports

Sleep-Related Eating Disorder and Sexsomnia; Two Rare Parasomnias? A Mini-Review with Illustrative Case-Reports.

OBJECTIVES: Sleep-related eating disorder and sexsomnia are non-rapid eye movement (NREM) parasomnias, characterized by reward-seeking behaviors during NREM sleep. These conditions are likely underpinned by specific patterns of sleep-wake dissociation and appear to be more common than expected, leading to significant health risks, psychological distress and even forensic consequences. METHODS: We briefly review recent literature on sleep-related eating disorder and sexsomnia, and present three cases of each; for illustrating their clinical features and complexity. RESULTS: Genetic predisposi

Public Understanding and Perception of Harm Reduction and Prevention Messaging About Fentanyl Overdoses.

BACKGROUND: Fentanyl-related opioid fatalities have risen drastically in the United States, indicating a "new wave" of the opioid crisis and highlighting the urgent need for more effective public health interventions to address its harms. Despite an increasing number of public communication campaigns focused on the general public, evidence on how people perceive fentanyl-related harm reduction strategies and prevention messaging is still nascent. METHODS: We conducted a cross-sectional survey with a national sample (N = 1,044). Fentanyl-related information seeking, risk perception, and messag

Criminal Justice Contact and Health Service Utilization among Women across Health Care Settings: Analyzing the Role of Arrest.

BACKGROUND: It is well-established in the United States that incarceration negatively influences women's health, and researchers have called for examinations of the health effects of criminal justice contact more broadly. This study uses the behavioral model for vulnerable populations to document the prevalence of illness and health risks for recently arrested women, and examines potential ways that illness and health risks are associated with health service use across health care settings. METHODS: We conducted a mediation analysis using pooled data from the National Survey on Drug Use and H

Psychopathological aspects of dopaminergic gene polymorphisms in adolescence and young adulthood.

Dopamine hypotheses of several psychiatric disorders are based upon the clinical benefits of drugs affecting dopamine transporter or receptors, and have prompted intensive candidate gene research within the dopaminergic system during the last two decades. The aim of this review is to survey the most important findings concerning dopaminergic gene polymorphisms in attention deficit hyperactivity disorder (ADHD), Tourette syndrome (TS), obsessive compulsive disorder, and substance abuse. Also, genetic findings of related phenotypes, such as inattention, impulsivity, aggressive behavior, and nove

Neuronal ensembles formed rapidly in the prelimbic cortex mediate initial and maintained cocaine-seeking behavior in male and female rats.

Substance use disorder is a complex neurobiological disease thought to be partially driven by aberrant forms of learning and memory. The cellular basis of memory is hypothesized to be small subsets of interconnected networks formed during the acquisition of behavior called neuronal ensembles. We interrogate neuronal ensembles by utilizing a dual viral approach that expresses CreERT2 under the control of the c-Fos promoter in conjunction with a Cre-dependent inhibitory DREADD. This approach gives us the temporal specificity to target neuronal ensembles formed during the acquisition of cocaine s

Involvement of Pre-limbic Cortex-Nucleus accumbens projections in Context-Induced alcohol seeking.

Alcohol use disorder (AUD) remains a critical public health issue worldwide, characterized by high relapse rates often triggered by contextual cues. This research investigates the neural mechanisms behind context-induced reinstatement of alcohol-seeking behavior, focusing on the nucleus accumbens and its interactions with the prelimbic cortex, employing Male Long-Evans rats in an ABA renewal model. In our experimental setup, rats were trained to self-administer 10 % ethanol in Context A, followed by extinction of lever pressing in the presence of discrete cues in Context B. The context-induced

Incentive Cocaine-Seeking Habits and Their Compulsive Manifestation Emerge After a Downregulation of the Dopamine Transporter in Astrocytes Across Functional Domains of the Striatum.

The development of compulsive cue-controlled-incentive drug-seeking habits is a hallmark of substance use disorder that is predicated on an intrastriatal shift in the locus of control over behaviour from a nucleus accumbens (Nac) core-dorsomedial striatum network to a Nac core-anterior dorsolateral striatum (aDLS) network. This shift is paralleled by drug-induced (including cocaine) dopamine transporter (DAT) alterations originating in the ventral striatum that spread eventually to encompass the aDLS. Having recently shown that heroin self-administration results in a pan-striatal reduction in

Partial MHC/neuroantigen peptide constructs attenuate methamphetamine-seeking and brain chemokine (C-C motif) ligand 2 levels in rats.

There are no medications that target the neurotoxic effects or reduce the use of methamphetamine. Recombinant T-cell receptor ligand (RTL) 1000 [a partial major histocompatibility complex (pMHC) class II construct with a tethered myelin peptide], addresses the neuroimmune effects of methamphetamine addiction by competitively inhibiting the disease-promoting activity of macrophage migration inhibitory factor to CD74, a key pathway involved in several chronic inflammatory conditions, including substance use disorders. We previously reported that RTL constructs improve learning and memory impairm

Role of Projections between Piriform Cortex and Orbitofrontal Cortex in Relapse to Fentanyl Seeking after Palatable Food Choice-Induced Voluntary Abstinence.

We recently developed a rat model of relapse to drug seeking after food choice-induced voluntary abstinence. Here, we used this model to study the role of the orbitofrontal cortex (OFC) and its afferent projections in relapse to fentanyl seeking. We trained male and female rats to self-administer palatable food pellets for 6 d (6 h/d) and intravenous fentanyl (2.5 μg/kg/infusion) for 12 d (6 h/d). We assessed relapse to fentanyl seeking after 13-14 voluntary abstinence days, achieved through a discrete choice procedure between fentanyl infusions and palatable food (20 trials/d). In both sexes,

Adolescent social isolation increases cocaine seeking in male and female mice.

Childhood and adolescent adversity are associated with a wide range of psychiatric disorders, including an increased risk for substance abuse. Despite this, the mechanisms underlying the ability of chronic stress during adolescence to alter reward signaling remains largely unexplored. Understanding how adolescent stress increases addiction-like phenotypes could inform the development of targeted interventions both before and after drug use. The current study examined how prolonged isolation stress, beginning during adolescence, affected behavioral and neuronal underpinnings to the response to

A partial trace amine-associated receptor 1 agonist exhibits properties consistent with a methamphetamine substitution treatment.

Recent evidence suggests that the trace amine-associated receptor 1 (TAAR1) plays a pivotal role in the regulation of dopamine (DA) transmission and psychostimulant action. Several selective TAAR1 agonists have previously shown efficacy in models of cocaine addiction. However, the effects of TAAR1 activation on methamphetamine (METH)-induced behaviours are less well understood, as indeed are the underlying neurochemical mechanisms mediating potential interactions between TAAR1 and METH. Here, in a progressive ratio schedule of reinforcement the partial TAAR1 agonist, RO5263397, reduced the bre

Evaluation of guanfacine as a potential medication for alcohol use disorder in long-term drinking rats: behavioral and electrophysiological findings.

One of the main treatment challenges in alcohol use disorder (AUD) is the high rate of craving in combination with decreased cognitive functioning including impaired decision making and impulse control that often lead to relapse. Recent studies show that guanfacine, an α-2-adrenoceptor agonist and FDA-approved ADHD medication, attenuates stress-induced relapse of several drugs of abuse including alcohol. Here we evaluated guanfacine's effects on voluntary alcohol intake, the alcohol deprivation effect (ADE), alcohol seeking behavior, and cue/priming-induced reinstatement in Wistar rats that ha

Effects of post-training heroin and d-amphetamine on consolidation of win-stay learning and fear conditioning.

It has been proposed that the reinforcing properties of drugs of abuse are due, in part, to their ability to enhance memory consolidation. To test this hypothesis, heroin (0.03-3 mg/kg, SC) and d-amphetamine (0.5-2 mg/kg, SC) were administered to male Sprague-Dawley rats immediately or 4 h after training on win-stay and fear conditioning tasks. On the win-stay, immediate post-training administration of lower doses of heroin and d-amphetamine enhanced acquisition, and probe tests further revealed that these drugs enhanced different aspects of learning. Higher doses had no effect or impaired per

Dopamine D2 Receptors, D1/D2 Receptor Heteromers and CaMKII Signaling in the Neurobiology of Drug Addiction.

Drug addiction is a serious brain disorder with widespread somatic, psychological, psychiatric, and legal consequences, characterized by compulsive drug seeking, craving, withdrawal, and relapse, particularly with psychostimulants. A key feature of drugs commonly abused by humans is their ability to enhance dopamine (DA) neurotransmission in mesolimbic circuits, thereby activating DA receptors and altering emotional and motivational behaviors. Recent studies have demonstrated that dopamine D2 receptors (D2Rs), existing both as monomers and as D1/D2 receptor heteromers with dopamine D1 receptor

Influence of calcium channel antagonists on nonsomatic signs of nicotine and D-amphetamine withdrawal in mice.

BACKGROUND: Nonsomatic signs of psychostimulant withdrawal, difficult to demonstrate in animal paradigms, may appear to promote drug seeking and drug relapse in humans; thus, it is important to understand the mechanisms that mediate this kind of behaviors. The present study was undertaken to examine the calcium-dependent mechanism of negative nonsomatic and anhedonia-related symptoms of acute and protracted withdrawal of nicotine and D-amphetamine. METHODS: Mice were chronically treated with nicotine (seven days, three times daily, 3.35 mg/kg, sc) or D-amphetamine (14 days, once daily, 2.5mg/
