Hereditary deafness is one of the most common disabilities affecting newborns. Many forms of hereditary deafness are caused by morphological defects of the stereocilia bundles on the apical surfaces of inner ear hair cells, which are responsible for sound detection. We explored the effectiveness of gene therapy in restoring the hair cell stereocilia architecture in the whirlin mouse model of human deafness, which is deaf due to dysmorphic, short stereocilia. Wild-type whirlin cDNA was delivered via adeno-associated virus (AAV8) by injection through the round window of the cochleas in neonatal

Usher syndrome (USH) is the most common inherited deaf-blindness with the majority of USH causative genes also involved in nonsyndromic recessive deafness (DFNB). The mechanism underlying this disease variation of USH genes is unclear. Here, we addressed this issue by investigating the DFNB31 gene, whose mutations cause USH2D or DFNB31 depending on their position. We found that the mouse DFNB31 ortholog (Dfnb31) expressed different mRNA variants and whirlin protein isoforms in the cochlea and retina, where these isoforms played different roles spatially and temporally. Full-length (FL-) whirli

The extracellular membranes of the inner ear are essential constituents to maintain sensory functions, the cupula for sensing torsional movements of the head, the otoconial membrane for sensing linear movements and accelerations like gravity, and the tectorial membrane in the cochlea for hearing. So far a number of structural proteins have been described, but for the gelatinous cupula precise data are missing. Here, we describe for the first time a major proteinogenic component of the cupula structure with an apparent molecular mass of 45 kDa from salmon. Analyses of respective peptides reveal

PURPOSE: The purpose of this study was to investigate the effect of oral isotretinoin, a drug used in the treatment of acne vulgaris, on hearing function determined by serial audiology examinations.
METHODS: Forty patients with acne vulgaris were included in this study. Nine patients were excluded from the study because of inconsistent follow-up. The hearing of each participant was tested with pure tone audiometry and transient evoked autoacoustic emissions before and two and four weeks after treatment with isotretinoin (0.3-0.6 mg/kg/day) in the remaining 31 patients (62 ears).
RESULTS: The

An overview is presented of the common and uncommon diseases of the inner ear and of the cochleovestibular nerve within the internal acoustic meatus and cerebellopontine angle cistern causing sensorineural deafness. An imaging-pattern-based approach is used to help detect disease and narrow the differential diagnosis. Main Messages • The most common soft tissue mass lesions in the cerebellopontine angle are schwannoma and meningioma. • Contrast-enhanced MRI may reveal clinically unsuspected inflammatory, auto-immune or tumoural disease. • Hearing loss may be caused by infection, inflammation or

BACKGROUND: Age-related hearing loss (ARHL) is the most common form of hearing loss in humans and is increasingly recognized in dogs.
HYPOTHESIS: Cochlear lesions in dogs with ARHL are similar to those in humans and the severity of the histological changes is reflected in tone audiograms.
ANIMALS: Ten geriatric dogs (mean age: 12.7 years) and three 9-month-old dogs serving as controls for histological analysis.
METHODS: Observational study. Auditory thresholds were determined by recording brainstem responses (BERA) to toneburst auditory stimuli (1, 2, 4, 8, 12, 16, 24, and 32 kHz). After eu

This case elucidates subtle cues that must be appreciated by the examiner in diving related injuries, who may not have experience with barotrauma-mediated pathology. Inner-ear barotrauma (IEBT) does not mandate ostensible hearing loss or vertigo; tinnitus may be the sole manifestation. Symptoms may present hours or even days post-dive. A common misconception exists that there are no efficacious treatment options for IEBT short of surgery for an overt perilymphatic fistula. Treatment options are available including acute high dose steroid administration, as prescribed for acute noise-induced or

Most 22q11.2 deletion syndrome (22q11DS) patients have middle and outer ear anomalies, whereas some have inner ear malformations. Tbx1, a gene hemizygously deleted in 22q11DS patients and required for ear development, is expressed in multiple tissues during embryogenesis. To determine the role of Tbx1 in the first pharyngeal pouch (PPI) in forming outer and middle ears, we tissue-specifically inactivated the gene using Foxg1-Cre. In the conditional mutants, PPI failed to outgrow, preventing the middle ear bone condensations from forming. Tbx1 was also inactivated in the otic vesicle (OV), resu

This review covers the general roles of members of the cysteine protease family of caspases in the process of apoptosis (programmed cell death) looking at their participation in both the "extrinsic" cell death receptor and the "intrinsic" mitochondrial cell death pathways. It defines the difference between initiator and effector caspases and shows the progression of caspase activations that ends up in the apoptotic cell death and elimination of a damaged cell. The review then presents what is currently know about the participation of caspases in the programmed cell death of inner ear sensory c

A probable relation between prostaglandin formation and the pathophysiology of hearing loss led to the application of the potent vasodilator prostacyclin in the treatment of inner ear disorders. Two drugs may be administered to humans: (1) unstable natural prostacyclin (epoprostenol) with a strong biological potency but with a high first-pass effect, or (2) a prostacyclin analogue (taprostene) with less biological activity, but a longer biological half-life and intravenous application. Therefore, the purpose of this study was to investigate potential differences of both drugs in their effect o

The effects of noise exposure were studied in fifty-nine 4- to 5-week-old albino Hartley guinea pigs with normal hearing (body weight 250-300 g). The following experiments and results were carried out: exposure to 350 Hz pure tones at 115 dB for 40 h and exposure to 350 Hz tones at 120 dB for 64 h. In order to investigate the effects of low-frequency tone exposures on the hearing of the guinea pigs, cochlear microphonics (CM), whole nerve action potentials (AP) and endocochlear potentials (EP) were measured. With a high-sound pressure, a decrease was observed in the CM maximum output voltage i

The uptake and chemical identification of 14C-retinyl acetate in the inner ear of the guinea-pig after oral administration is reported. For methodological reasons the experiment was carried out in vitamin A-deficient guinea-pigs. In the sensory tissues a time-dependent distribution was found similar to that in other organs. The chemical identification shows that the orally administered labeled retinyl acetate can be detected as retinyl palmitate in the membranous structures of the inner ear. This may be an indication for the ability of the inner ear tissues to esterize and probably store the t

Inner ear organoids (IEOs) derived from pluripotent stem cells (PSCs) provide a promising platform for modeling neurosensory disorders and hearing loss; however, conventional systems often exhibit substantial structural variability, incomplete maturation, and limited reproducibility due to insufficient control of early organoid morphogenesis. Here, we demonstrate that micro-topographical cues applied during initial IEO formation enhance the development and functional maturation of PSC-derived IEOs. This microengineering strategy introduces temporally defined microscale geometric confinement to

PURPOSE: Accurate subtype classification of inner ear malformations (IEMs) on temporal bone high-resolution computed tomography (HRCT) is important for preoperative risk assessment and surgical planning, but depends heavily on subspecialty expertise. This study aimed to develop and externally validate a deep learning model for multiclass IEM diagnosis on temporal bone HRCT.
METHODS: We developed and externally validated a weakly supervised deep learning framework for one-stage classification of automatically extracted temporal bone regions of interest from HRCT in a multicenter cohort of 9,18

INTRODUCTION: This study differentiates cases of acute-onset sensorineural hearing loss (SNHL) that did not meet the diagnostic criteria for either sudden SNHL or Meniere's disease, leading to a diagnostic challenge.
METHODS: All 46 patients with acute-onset SNHL underwent an inner ear test battery followed by hydrops magnetic resonance imaging (MRI). Twenty-four patients (28 ears) who tested positive for endolymphatic hydrops (EH) on hydrops MRI were referred to the EH group. Another 22 patients (22 ears) who tested negative for EH were classified as the non-EH group.
RESULTS: Significant d

Roughly 5% of the population is affected by hearing loss. In case of a functional middle ear impairment with decreased transmission, different types of active middle ear implants may restore hearing. To drive the round window membrane (RWM) for example, bone anchored actuators or floating mass transducer (FMT) have been used in clinics. Bone anchored devices are usually too big for the round window niche and FMTs display low displacement and force at low frequencies. This paper focuses on the stimulation of the RWM by a stator-based actuator that combines small size and bone anchoring. The des
