Genetic predisposition for the development of radiation-associated meningioma: an epidemiological study.

BACKGROUND: Ionising radiation is an established risk factor for meningioma, yet less than 1% of irradiated individuals develop this tumour. Familial aggregation of meningioma is rare. We aimed to assess whether genetic factors can modify the risk for meningioma formation after the initiating effect of radiation, by comparison of the frequency of meningiomas in families that included irradiated and unirradiated siblings. METHODS: This study was based on a larger epidemiological, genetic case-control study, and included 525 families that were divided according to irradiation and disease status

Cardiogenic effects of trichloroethylene and trichloroacetic acid following exposure during heart specification of avian development.

Trichloroethylene (TCE) and its metabolite trichloroacetic acid (TCA) are common drinking water contaminants in the United States. Both chemicals have been implicated in causing congenital heart defects (CHD) in human epidemiological and animal model studies. However, the latter studies have primarily focused on assessment of cardiac morphology at late embryonic stages. Here, we tested whether treating avian embryos with TCE or TCA during an exposure window encompassing cardiac specification (Hamburger-Hamilton [HH] 3+) until the onset of chambering (HH 17) informs the etiology of CHD at later

Quantitative risk assessment of Cryptosporidium species infection in dairy calves.

Cryptosporidium parvum is a zoonotic protozoan that infects many different mammals including cattle and humans. Cryptosporidiosis has become a concern for dairy producers because of the direct losses due to calves not performing well and the potential for environmental contamination with C. parvum. Identifying modifiable control points in the dynamics of infection in dairy herds will help identify management strategies that mitigate its risk. The quantitative risk assessment approach provides estimates of the risk associated with these factors so that cost-effective strategies can be implement

Actual causes of death in the United States, 2000.

CONTEXT: Modifiable behavioral risk factors are leading causes of mortality in the United States. Quantifying these will provide insight into the effects of recent trends and the implications of missed prevention opportunities. OBJECTIVES: To identify and quantify the leading causes of mortality in the United States. DESIGN: Comprehensive MEDLINE search of English-language articles that identified epidemiological, clinical, and laboratory studies linking risk behaviors and mortality. The search was initially restricted to articles published during or after 1990, but we later included relevan

Use of sunscreen is linked with elevated naevi counts in Israeli school children and adolescents.

Epidemiological evidence indicates that a fair complexion and exposure to solar radiation in early years contribute to the proliferation of naevi and subsequent melanoma risk. To determine whether protection from sunlight is associated with lower naevi counts, we examined the prevalence and risk factors of naevi in a sample of Israeli school pupils. Whole body counts of naevi were recorded in 974 out of 1312 (74.2%) recruited pupils (7 and 12 year olds) from Ramat-Gan and Jerusalem. Host characteristics and habits of sun exposure were obtained using questionnaires. In each age group and geogra

Urinary metal mixtures and cardio-kidney-metabolic risk in adults from a legacy-contaminated area: Repeated-measures cohort evidence and computational validation.

Cardiovascular-kidney-metabolic (CKM) syndrome links metabolic dysfunction, kidney injury, and cardiovascular disease; however, how real-world toxic exposures and social disadvantage accelerate CKM progression is not well defined. We followed a pollution-exposed rural cohort in Northeast China from 2016 to 2021 (n = 472; 2360 person-visits). Urinary chromium (Cr), cadmium (Cd), manganese (Mn), and lead (Pb) were measured by inductively coupled plasma mass spectrometry, and CKM stage was assigned using the current American Heart Association framework. We modeled advanced CKM (stages 3-4) versus

Incidence trends and age-period-cohort analysis of Alzheimer's disease and other dementias in the world and China from 1990 to 2021: analyses based on the Global Burden of Disease Study 2021.

BACKGROUND: With the accelerating global population aging, the incidence rates of Alzheimer's disease and other dementias (ADOD) continue to rise sharply, presenting a pressing public health challenge worldwide. In China, where population aging is occurring at an unprecedented rate, the epidemiological trajectory of ADOD has garnered significant attention. The marked increase in incidence rates has imposed a substantial burden on both society and families, driven by direct healthcare costs, long-term care demands, and productivity losses among caregivers. METHODS: This study utilized the age-

The impact of primary mental healthcare on core symptoms of depression among underrepresented adolescents: a network analysis perspective.

BACKGROUND: Depression has emerged as a leading contribution of the global mental health burden, particularly among underrepresented adolescents. Despite the World Health Organization's promotion of primary mental healthcare as a critical solution, its real-world effectiveness in low- and middle-income countries remains debated due to high costs and insufficient follow-up. This study aimed to explore the impact of primary mental healthcare on the core symptoms of adolescent depression using network analysis, while examining the influence of demographic factors such as gender, age, and family s

Associations of Outdoor Air Pollution With Cause-specific Mortality in Metastatic Prostate Cancer: A cohort study.

BACKGROUND: Cardiovascular disease is an important competing cause of death in men with metastatic prostate cancer, and ambient air pollution may increase cardiovascular vulnerability. We evaluated associations of fine particulate matter (PM2.5) and nitrogen dioxide (NO2) with all-cause, cardiovascular, and prostate cancer-specific mortality, including joint associations with androgen deprivation therapy (ADT). METHODS: We analyzed 33,384 men from 11 US state registries diagnosed with metastatic prostate cancer during 2000-2015 and followed through 2018. Residential PM2.5 and NO2 concentratio

Prenatal exposure to nitrosatable drugs and timing of puberty in sons and daughters: A nationwide cohort study.

BACKGROUND: N-nitroso compounds (NOCs) can be formed by endogenous reactions between nitrosatable drugs and nitrite. Animal studies have found that several NOCs are teratogenic, and epidemiological studies report associations between prenatal exposure to nitrosatable drugs and adverse birth outcomes. It is unknown whether prenatal exposure to nitrosatable drugs is harmful to the child's reproductive health, including pubertal development. OBJECTIVES: We investigated whether prenatal exposure to nitrosatable drugs was associated with timing of puberty and whether nitrate, nitrite and antioxida

Multiscale epidemiology of gastrointestinal nematode infections in small ruminants: field evidence from Inner Mongolia and a national meta-analysis in China.

BACKGROUND: Gastrointestinal nematodes (GINs) are among the most important parasites constraining small-ruminant production. However, epidemiological evidence in China remains fragmented because existing studies differ substantially in geographic coverage, host species, sampling periods, diagnostic approaches, and ecological settings. METHODS: We applied a multiscale framework integrating a regional survey in Inner Mongolia with a national systematic review and meta-analysis. In the regional survey, 2,084 fecal samples were collected from sheep aged 6-12 months in Ulanqab, Alxa, and Ordos fro

Causal relationship between physical activity and common gynecologic conditions: a two-sample Mendelian randomization study.

OBJECTIVE: While physical activity (PA) patterns are epidemiologically linked to gynecologic pathologies, establishing causality remains uncertain. This Mendelian randomization (MR) study evaluated causal relationships between PA phenotypes and gynecologic disorders. METHODS: Using European-ancestry female genome-wide association study (GWAS) data, we analyzed eight gynecologic disorders and four PA instruments. Primary inverse-variance weighted (IVW) estimates were supplemented by sensitivity analyses (MR-Egger, weighted median, MR-PRESSO) to address pleiotropy and heterogeneity. RESULTS: O

Outcomes and Management of Stressor-Associated Atrial Fibrillation: JACC State-of-the-Art Review.

Stressor-associated atrial fibrillation (AF) is defined as new onset AF occurring in the setting of an acute and reversible physiologic stressor (eg, cardiac surgery, acute infection). Although historically viewed as transient and potentially benign, a growing body of evidence demonstrates that stressor-associated AF is prognostically important, with long-term rates of AF recurrence and AF-related adverse events (eg, stroke, heart failure) approaching those of AF occurring without a stressor. Nevertheless, key aspects of stressor-associated AF management (eg, prevention, surveillance, treatmen

Microbiome and pediatric leukemia, diabetes, and allergies: Systematic review and meta-analysis.

BACKGROUND: Despite the different pathologies and genetic susceptibilities of childhood ALL, T1DM and allergies, these conditions share epidemiological risk factors related to timing of infectious exposures and acquisition of the gut microbiome in infancy. We have assessed whether lower microbiome diversity (Shannon Index) and shared genus/species profiles are associated with pediatric ALL, allergies, and T1DM. METHODS AND FINDINGS: Literature search was performed using PubMed, Embase, Cochrane, and Web of Science databases. Case-control, meta-analyses, and cohort studies were considered for

Vitamin D and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD): Novel Mechanistic Insights.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is an increasingly prevalent condition characterized by abnormal fat accumulation in the liver, often associated with metabolic disorders. Emerging evidence suggests a potential link between vitamin D deficiency and the development and progression of MASLD. The current review provides a concise overview of recent studies uncovering novel mechanistic insights into the interplay between vitamin D and MASLD. Several epidemiological studies have highlighted a significant association between low vitamin D levels and an increased risk

Organophosphate ester flame retardant chemicals and maternal depression during pregnancy.

BACKGROUND: Depression substantially contributes to pregnancy-related morbidity, and pregnancy is increasingly recognized as a vulnerable window for exposure effects on maternal mental health. Exposures to organophosphate esters (OPEs) are ubiquitous and may have neurotoxic effects; however, their impacts on prenatal depression remain unknown. We evaluated associations of third trimester OPE metabolites on maternal depressive symptoms during pregnancy. METHODS: This study included 422 participants in the Maternal and Developmental Risks from Environmental and Social Stressors (MADRES) cohort,
