Ovulatory release and oviductal recovery after Intrafollicular Transfer of Immature Oocytes (IFIOT) in cattle.

Intrafollicular transfer of immature oocytes (IFIOT) is an emerging reproductive biotechnology that enables oocyte maturation, fertilization, and early embryonic development to occur in vivo. Despite its feasibility, IFIOT efficiency remains low, and the stages at which losses occur are not well defined. This study aimed to evaluate early post-ovulatory events after IFIOT by directly recovering structures from the oviduct, with particular emphasis on ovulatory release and early recovery dynamics under different oocyte loads. Immature cumulus-oocyte complexes (COCs) were recovered from slaughte

Human placental stem cells: biomedical potential and clinical relevance.

Emerging evidence shows that different adult tissues of the human body harbour small amounts of adult mesenchymal stem cells (MSCs) which have the potential to give a rise to all cell types of the specific tissue in which they reside. These cells were found in human adult bone marrow, muscle, fat, brain, cartilage, dental pulp, etc. Depending on the type of host tissue as well as age and state of the donor's health, the plasticity of MSCs can vary considerably. Previously accomplished research efforts have shown that adult MSCs have a broad therapeutic potential due to their ability to differe

Adaptive variation in avian eggshell gas conductance and structure across elevational gradients?

Many tropical bird species have restricted elevational distributions, potentially limited by how environmental conditions affect physiological processes. While some studies have examined adult physiology across elevations, relatively little attention has been given to the structure and function of eggshells despite their critical role in regulating gas exchange during the vulnerable embryonic stage. At high elevations, dry air is expected to increase water loss from the egg, and natural selection may favor lower gas conductance to reduce desiccation risk. Structural variation in eggshells, suc

Evolution in the clinic: Maladaptive units and "minor anomalies".

It is here argued that the application of the term "minor anomalies" is often imprecise and likely outdated. In the past, the designation was used indiscriminately to refer to a great variety of unrelated morphogenetic phenomena. Also, the term does not discriminate between mild qualitative defects of development (mild malformations) and quantitative variants of normal structure. The human face was formed by natural and sexual selection. Morphological and morphogenetic analyses have shown that the human face with its skin, muscles, nerves, arteries, veins, glands, and lymphatics is a complex s

The developmental programme for genesis of the entire kidney is recapitulated in Wilms tumour.

Wilms tumour (WT) is an embryonal tumour that recapitulates kidney development. The normal kidney is formed from two distinct embryological origins: the metanephric mesenchyme (MM) and the ureteric bud (UB). It is generally accepted that WT arises from precursor cells in the MM; however whether UB-equivalent structures participate in tumorigenesis is uncertain. To address the question of the involvement of UB, we assessed 55 Wilms tumours for the molecular features of MM and UB using gene expression profiling, immunohistochemsitry and immunofluorescence. Expression profiling primarily based on

Lysine-specific demethylase 7A (KDM7A): A potential target for disease therapy.

Histone demethylation is a kind of epigenetic modification mediated by a variety of enzymes and participates in regulating multiple physiological and pathological events. Lysine-specific demethylase 7A is a kind of α-ketoglutarate- and Fe(II)-dependent demethylase belonging to the PHF2/8 subfamily of the JmjC demethylases. KDM7A is mainly localized in the nucleus and contributes to transcriptional activation via removing mono- and di-methyl groups from the lysine residues 9 and 27 of Histone H3. Mounting studies support that KDM7A is not only necessary for normal embryonic, neural, and skeleta

Cytogenetic evaluation of a large series of hepatoblastomas: numerical abnormalities with recurring aberrations involving 1q12-q21.

Hepatoblastoma is a malignant embryonal liver tumor that occurs almost exclusively in infants and very young children. Previous cytogenetic studies of hepatoblastoma have investigated small series or individual cases. This report is on the cytogenetics of a large series of 111 hepatoblastoma specimens, with cytogenetic results consecutively karyotyped over a 12-year period. Abnormal karyotypes were observed in 55 cases (approximately 50% of the total). Numerical aberrations were observed in 41 cases (36% of the total), particularly trisomies of chromosomes 2, 8, and 20. Chromosome losses were

Structure-guided design of icaritin-based scaffold modifications for enhanced osteoporotic therapy.

Icaritin, a flavonoid compound, has been shown to alleviate osteoporosis by promoting the differentiation of osteoblasts. However, research on the structural modification and optimization of icaritin has been limited. To enhance the anti-osteoporotic efficacy of icaritin, we structurally modified the compound by substituting the hydroxyl groups, resulting in the synthesis of 22 derivatives. The impact of these compounds was examined by evaluating Human embryonic palatal mesenchymal (HEPM) cells viability with the MTT assay, along with assessing osteogenic potential through measurement of alkal

Construction and sequencing of an infectious clone of the goose embryo-adapted Muscovy duck parvovirus vaccine strain FZ91-30.

BACKGROUND: Muscovy duck parvovirus (MDPV) is the etiological agent of Muscovy duckling parvoviral disease, which is characterized by diarrhea, locomotive dysfunction, stunting, and death in young ducklings, and causes substantial economic losses in the Muscovy duck industry worldwide. FZ91-30 is an attenuated vaccine strain that is safe and immunogenic to ducklings, but the genomic information and molecular mechanism underlining the attenuation are not understood. METHODS: The FZ91-30 strain was propagated in 11-day-old embryonated goose eggs, and viral particles were purified from the poole

Type 2 diabetes mellitus induces congenital heart defects in murine embryos by increasing oxidative stress, endoplasmic reticulum stress, and apoptosis.

BACKGROUND: Maternal type 1 and 2 diabetes mellitus are strongly associated with high rates of severe structural birth defects, including congenital heart defects. Studies in type 1 diabetic embryopathy animal models have demonstrated that cellular stress-induced apoptosis mediates the teratogenicity of maternal diabetes leading to congenital heart defect formation. However, the mechanisms underlying maternal type 2 diabetes mellitus-induced congenital heart defects remain largely unknown. OBJECTIVE: We aim to determine whether oxidative stress, endoplasmic reticulum stress, and excessive apo

Prenatal nutritional manipulation by in ovo enrichment influences bone structure, composition, and mechanical properties.

The objective of this study was to examine the effect of embryonic nutritional enrichment on the development and properties of broiler leg bones (tibia and femur) from the prenatal period until maturity. To accomplish the objective, 300 eggs were divided into 2 groups: a noninjected group (control) and a group injected in ovo with a solution containing minerals, vitamins, and carbohydrates (enriched). Tibia and femur from both legs were harvested from chicks on embryonic days 19 (E19) and 21 (E21) and d 3, 7, 14, 28, and 54 posthatch (n = 8). The bones were mechanically tested (stiffness, maxi

Pleiotropic action of genetic variation in ZNF804A on brain structure: a meta-analysis of magnetic resonance imaging studies.

OBJECTIVE: The zinc finger protein 804A (ZNF804A) gene encodes the protein 804A containing the C2H2 zinc finger structure, which plays an important role in embryonic nerve development and repair. Previous studies have shown a significant association between the ZNF804A genetic variation polymorphism rs1344706 and the risk of schizophrenia and brain structure abnormalities. However, the findings are inconsistent. MATERIALS AND METHODS: Seventeen studies on structural magnetic resonance imaging (sMRI), with 1,031 schizophrenia patients and 3,416 healthy controls, were included in the meta-analy

Loss of cell adhesion causes hydrocephalus in nonmuscle myosin II-B-ablated and mutated mice.

Ablation of nonmuscle myosin (NM) II-B in mice during embryonic development leads to marked enlargement of the cerebral ventricles and destruction of brain tissue, due to hydrocephalus. We have identified a transient mesh-like structure present at the apical border of cells lining the spinal canal of mice during development. This structure, which only contains the II-B isoform of NM, also contains beta-catenin and N-cadherin, consistent with a role in cell adhesion. Ablation of NM II-B or replacement of NM II-B with decreased amounts of a mutant (R709C), motor-impaired NM II-B in mice results

[Clinical and scanning electron microscopic studies on Norwegian scabies infection].

Two typical cases of Norwegian scabies were studied clinically, scanning electron microscopically and histopathologically. Case 1 was a boy with Down's syndrome and case 2 was a man with adult T cell leukemia/lymphoma. Their clinical pictures showed hyperkeratotic plaques and warty crusts on hands, feet, ears, elbows and buttocks. Direct examination of the KOH preparation revealed many embryonated sarcoptic eggs, postpartum egg-shells, fecal pellets, larvae and male and female adult mites. Histopathologic examination showed several burrows inside the thick horny layer. Many sarcoptic egg-shell

Transcripts and Protein Expression of Different HMW Non-Myofibrillar Tpm1 Isoforms in Human iPSC-CMs During Differentiation.

Mammals have four tropomyosin (TPM) genes (TPM1, TPM2, TPM3, and TPM4) that produce various isoforms through alternative splicing. TPM1 generates both myofibrillar (Tpm1.1, Tpm1.2) and non-myofibrillar isoforms, including those with an exon 9d peptide or with an exon 9a peptide. These non-myofibrillar isoforms are involved in regulating actin-based structures and processes in non-muscle cells. This study focused on the expression of the non-myofibrillar HMW Tpm1 isoforms (Tpm1.3, Tpm1.4, Tpm1.5, Tpm1.6, Tpm1.7, and Tpm1.14) during human inducible pluripotent stem cells (hiPSC) differentiation

Jumonji domain-containing protein 6 protein and its role in cancer.

The jumonji domain-containing protein 6 (JMJD6) is a Fe(II)- and 2-oxoglutarate (2OG)-dependent oxygenase that catalyses lysine hydroxylation and arginine demethylation of histone and non-histone peptides. Recently, the intrinsic tyrosine kinase activity of JMJD6 has also been reported. The JMJD6 has been implicated in embryonic development, cellular proliferation and migration, self-tolerance induction in the thymus, and adipocyte differentiation. Not surprisingly, abnormal expression of JMJD6 may contribute to the development of many diseases, such as neuropathic pain, foot-and-mouth disease

Analysis of pancreatic disease in zebrafish.

One of the appeals of the zebrafish model is the relative ease of studying disease progression from embryonic or larval stages through to adulthood. Because of this, the zebrafish has become an important model for postembryonic pancreatic disease, particularly diabetes and pancreatic cancer. Here we present methods for using the adult zebrafish to analyze pancreas function and structure, with an emphasis on the endocrine pancreas and the beta cells. The methods include fasting, weighing adults, and anesthetizing adults, and intraperitoneal injection of glucose based on body weight. We also pre

cDNA cloning of p40, a regulatory subunit of the human 26S proteasome, and a homolog of the Mov-34 gene product.

The nucleotide sequence of a cDNA that encodes a new regulatory subunit, named p40, of the 26S proteasome of human hepatoblastoma HepG2 cells has been determined. The polypeptide predicted from the open reading frame consists of 324 amino acid residues with a calculated molecular mass of 37020 and isoelectric point of 6.03. A KEKE motif, consisting of a very hydrophilic domain rich in 'alternating' lysine (positive) and glutamate (negative) residues, is present in the C-terminus of p40. The overall structure of p40 is homologous to that of the mouse Mov-34 gene product, whose gene disruption b
