A novel HDAC1-specific inhibitor prevents estrogen deficiency-induced osteoporosis in mice by inhibiting osteoclast function.

Postmenopausal osteoporosis (PMOP) features reduced bone mass and deteriorated bone microstructure, increasing fracture risk. Estrogen deficiency-induced osteoclast overactivation is a primary driver. OCP-001, a novel highly selective HDAC1 inhibitor, was investigated. TRAP staining showed dose-dependent inhibition of osteoclast differentiation by OCP-001. qPCR revealed concomitant downregulation of RANKL-induced osteoclast marker genes (Trap, DC-Stamp, NFATc1, ATP60) and key regulatory proteins (c-Fos, NFATc1, Blimp-1, IRF-8). In an ovariectomized (OVX)-induced PMOP mouse model, body weight m

Candida albicans increases the aerobic glycolysis and activates MAPK-dependent inflammatory response of liver sinusoidal endothelial cells.

The liver, and more specifically, the liver sinusoidal endothelial cells, constitute the beginning of one of the most important responses for the elimination of hematogenously disseminated Candida albicans. Therefore, we aimed to study the mechanisms involved in the interaction between these cells and C. albicans. Transcriptomics-based analysis showed an increase in the expression of genes related to the immune response (including receptors, cytokines, and adhesion molecules), as well as to aerobic glycolysis. Further in vitro analyses showed that IL-6 production in response to C. albicans is

Betulinic Acid Protects From Bone Loss in Ovariectomized Mice and Suppresses RANKL-Associated Osteoclastogenesis by Inhibiting the MAPK and NFATc1 Pathways.

Osteoclasts with elevated bone resorption are commonly present in postmenopausal osteoporosis, and other osteolytic pathologies. Therefore, suppressing osteoclast generation and function has been the main focus of osteoporosis treatment. Betulinic acid (BA) represents a triterpenoid mainly purified from the bark of Betulaceae. BA shows multiple biological activities, including antitumor and anti-HIV properties, but its effect on osteolytic conditions is unknown. Here, BA suppressed receptor activator of nuclear factor-κB ligand (RANKL)-associated osteoclastogenesis and bone resorptive function

Interleukin-27 prevents LPS-induced inflammatory osteolysis by inhibiting osteoclast formation and function.

Osteolysis is a serious complication of several chronic inflammatory diseases and is closely associated with a local chronic inflammatory reaction with a variety of causes. Inflammatory factors and osteoclastogenesis can enhance bone erosion. Interleukin-27 (IL-27) is speculated to play an important role in the physiological immune response. However, there are few studies on its effects on osteoclastogenesis. In this study, IL-27 was shown to inhibit receptor activator nuclear factor-κB ligand (RANKL)-induced osteoclastogenesis. The gene expression levels of osteoclast (OC)-specific genes, suc

Specific promoter deacetylation of histone H3 is conserved across mouse models of Huntington's disease in the absence of bulk changes.

Defective epigenetic regulation has been postulated as a possible cause for the extensive and premature transcriptional dysregulation observed in experimental models of Huntington's disease (HD). In this study, we extended our observations in the N171-82Q mouse strain relating to the limited impact of polyQ pathology on the global histone acetylation to other animal and cellular models of HD, namely the R6/1 and YAC128 strains, striatal-electroporated mice, primary neuronal cultures and stably transfected PC12 cells. In the absence of bulk chromatin changes, we nonetheless documented histone d

Deletion of glucocorticoid receptors in Lepr-expressing neurons attenuates the effects of negative energy balance on female reproduction.

BACKGROUND: Negative energy balance increases glucocorticoids (GC) and inhibits reproduction; however, the neurocircuitry mediating these effects remains poorly understood. We hypothesized that GC signaling in leptin receptor (Lepr)-expressing neurons mediates reproductive disruption caused by metabolic stress. METHODS: We generated a mouse model with selective deletion of glucocorticoid receptor (GR) in Lepr-expressing cells (LeprGRKO). We characterized pubertal development, reproductive phenotype, ovarian folliculogenesis, and hypothalamic gene expression in female LeprGRKO mice and in GRlo

Spinorphin Ameliorates Nitroglycerin-Induced Migraine-Like Responses and Trigeminovascular Neuroinflammation Potentially Through Opioid Receptor-Mediated Mechanisms.

Enhancement of endogenous opioid signaling through inhibition of enkephalin degradation has emerged as a potential strategy for pain control, yet its role in migraine remains unclear. This study investigated whether spinorphin, an endogenous inhibitor of multiple enkephalinases, attenuates trigeminovascular activation and migraine-like responses in experimental migraine. Migraine-like pain was induced in male Wistar rats by nitroglycerin. The effects of spinorphin were evaluated using periorbital and plantar von Frey tests, followed by assessment of c-Fos levels in the trigeminal nucleus cauda

Spinorphin Attenuates Acute Experimental Seizures and Associated Neurochemical Alterations: Possible Involvement of Opioid Receptors.

Spinorphin, an endogenous enkephalinase inhibitor, enhances physiological opioid signaling by preventing the degradation of endogenous enkephalins, however, its anticonvulsant potential has not previously been investigated. This study evaluated the effects of spinorphin on seizure activity and associated neurochemical alterations in two complementary acute experimental seizure models. The effects of spinorphin on seizure activity were investigated in pentylenetetrazole (PTZ)- and penicillin-induced acute seizure rat models. Behavioral seizure parameters, passive avoidance memory, electrocortic

Esculetin attenuates migraine-like pain via CGRP suppression and meningeal mast cell modulation in rat models.

Growing evidence suggests that esculetin, a 5-lipoxygenase inhibitor, has pharmacotherapeutic potential due to its various pharmacological properties, such as potent anti-inflammatory, anti-nociceptive, and γ-aminobutyric acid type A (GABAA) receptor partial agonist activities. However, the effects of this promising agent on migraine remain unexplored. This study therefore examined the impact of esculetin on relevant mechanisms in migraine-like conditions in rats. The systemic effects of esculetin at three distinct doses (5, 10, and 20 mg/kg) were tested in a nitroglycerin (NTG)-induced migrai

Cannabinoid agonist, WIN 55,212-2, increases in vivo paired-pulse facilitation, long-term potentiation, and Arc expression in the dentate gyrus.

The endogenous cannabinoid system in the hippocampus has the potential to modulate synaptic transmission, including short and long term plasticity, by acting as retrograde messengers. The present study evaluated the effects of WIN 55,212-2, a potent cannabinoid agonist, on paired-pulse facilitation (PPF) and long-term potentiation (LTP) in the dentate gyrus in vivo. WIN 55,212-2 (10  μg/μL; 0.5  μL volume) or vehicle was unilaterally infused into the right dorsal hippocampus of anesthetized rats prior to perforant path stimulation and electrophysiological recordings. Alprazolam induces anterograde amnesia for contextual fear memory and alters dorsoventral hippocampal neuronal ensembles in female mice.

Benzodiazepines (BZDs) are commonly prescribed anxiolytic drugs that act on GABAa receptors, and can result in anterograde amnesia, or the inability to form new memories. While BZDs have been used for decades, the brain regions and neuronal mechanisms responsible for this detrimental side effect are largely unknown at the systems neuroscience level. To analyze the effects of BZDs on long-term memory, activity-dependent ArcCreERT2 x eYFP mice were injected with Alprazolam 30 min prior to a 3-shock contextual fear conditioning (CFC) procedure and encoding ensembles were tagged with eYFP. Mice we

Potentiation of the M1 muscarinic acetylcholine receptor normalizes neuronal activation patterns and improves apnea severity in Mecp2+/- mice.

Rett syndrome (RTT) is a neurodevelopmental disorder that is caused by loss-of-function mutations in the methyl-CpG binding protein 2 (MeCP2) gene. RTT patients experience a myriad of debilitating symptoms, which include respiratory phenotypes that are often associated with lethality. Our previous work established that expression of the M1 muscarinic acetylcholine receptor (mAchR) is decreased in RTT autopsy samples, and that potentiation of the M1 receptor improves apneas in a mouse model of RTT; however, the population of neurons driving this rescue is unclear. Loss of Mecp2 correlates with

Ethanol consumption activates a subset of arcuate nucleus pro-opiomelanocortin (POMC)-producing neurons: a c-fos immunohistochemistry study.

Ethanol activates various opioid peptide-containing circuits within the brain that may underlie its motivational and rewarding effects. One component of this circuitry consists of neurons located in the arcuate nucleus (ArcN) of the hypothalamus which express pro-opiomelanocortin (POMC), an opioid precursor peptide that is cleaved to form bioactive fragments including β-endorphin and α-melanocyte stimulating hormone. In this study, we sought to determine if ethanol intake activates ArcN POMC neurons as measured by expression of the immediate early gene c-fos. Male and female POMC-EGFP mice und

Novel Antidepressant-Like Properties of the Iron Chelator Deferiprone in a Mouse Model of Depression.

Depressed individuals who carry the short allele for the serotonin-transporter-linked promotor region of the gene are more vulnerable to stress and have reduced response to first-line antidepressants such as selective serotonin reuptake inhibitors. Since depression severity has been reported to correlate with brain iron levels, the present study aimed to characterise the potential antidepressant properties of the iron chelator deferiprone. Using the serotonin transporter knock-out (5-HTT KO) mouse model, we assessed the behavioural effects of acute deferiprone on the Porsolt swim test (PST) an

Long-lasting tagging of neurons activated by seizures or cocaine administration in Egr1-CreERT2 transgenic mice.

Permanent tagging of neuronal ensembles activated in specific experimental situations is an important objective to study their properties and adaptations. In the context of learning and memory, these neurons are referred to as engram neurons. Here, we describe and characterize a novel mouse line, Egr1-CreERT2 , which carries a transgene in which the promoter of the immediate early gene Egr1 drives the expression of the CreERT2 recombinase that is only active in the presence of tamoxifen metabolite, 4-hydroxy-tamoxifen (4-OHT). Egr1-CreERT2 mice were crossed with various reporter mice, Cre-depe

Noisy galvanic vestibular stimulation enhances spatial memory in cognitive impairment-induced by intracerebroventricular-streptozotocin administration.

There are several anatomical connections between vestibular system and brain areas construct spatial memory. Since subliminal noisy galvanic vestibular stimulation (GVS) has been demonstrated to enhance some types of memory, we speculated that application of noisy GVS may improve spatial memory in a rat model of intracerebroventricular streptozotocin (ICV-STZ)-induced cognitive impairment. Moreover, we attempted to determine the effect of repeated exposure to GVS on spatial memory performance. The spatial memory was assessed using Morris water maze test. The groups received 1 (ICV-STZ/GVS-I) o

Antidepressant-like effects of kappa-opioid receptor antagonists in Wistar Kyoto rats.

The Wistar Kyoto (WKY) rat strain is a putative genetic model of comorbid depression and anxiety. Previous research showing increased kappa-opioid receptor (KOR) gene expression in the brains of WKY rats, combined with studies implicating the KOR in animal models of depression and anxiety, suggests that alterations in the KOR system could have a role in the WKY behavioral phenotype. Here, the effects of KOR antagonists in the forced swim test (FST) were compared with the WKY and the Sprague-Dawley (SD) rat strains. As previously reported, WKY rats showed more immobility behavior than SD rats.

C. elegans EVI1 proto-oncogene, EGL-43, is necessary for Notch-mediated cell fate specification and regulates cell invasion.

During C. elegans development, LIN-12 (Notch) signaling specifies the anchor cell (AC) and ventral uterine precursor cell (VU) fates from two equivalent pre-AC/pre-VU cells in the hermaphrodite gonad. Once specified, the AC induces patterned proliferation of vulva via expression of LIN-3 (EGF) and then invades into the vulval epithelium. Although these cellular processes are essential for the proper organogenesis of vulva and appear to be temporally regulated, the mechanisms that coordinate the processes are not well understood. We computationally identified egl-43 as a gene likely to be expre
