Glutamate dehydrogenase: Potential therapeutic targets for neurodegenerative disease.

Glutamate dehydrogenase (GDH) is a key enzyme in mammalian glutamate metabolism. It is located at the intersection of multiple metabolic pathways and participates in a variety of cellular activities. GDH activity is strictly regulated by a variety of allosteric compounds. Here, we review the unique distribution and expressions of GDH in the brain nervous system. GDH plays an essential role in the glutamate-glutamine-GABA cycle between astrocytes and neurons. The dysfunction of GDH may induce the occurrence of many neurodegenerative diseases, such as Parkinson's disease, epilepsy, Alzheimer's d

[Stiff-Person Syndrome and Related Autoantibodies].

Stiff-person syndrome (SPS) is a disorder characterized by fluctuating muscle rigidity and painful spasms that occur spontaneously or are triggered by diverse stimuli. Partial or segmental forms of the disorder, such as stiff-limb syndrome (SLS) and a more severe disease called progressive encephalomyelitis with rigidity and myoclonus (PERM), are usually considered within the spectrum of SPS. SPS responds to immunotherapies, and several autoantigens have been identified. Most patients with SPS have a high-titer of antibodies against glutamic acid decarboxylase (GAD), the rate-limiting enzyme f…

Dendritic Cell Factor 1-Knockout Results in Visual Deficit Through the GABA System in Mouse Primary Visual Cortex.

The visual system plays an important role in our daily life. In this study, we found that loss of dendritic cell factor 1 (DCF1) in the primary visual cortex (V1) caused a sight deficit in mice and induced an abnormal increase in glutamic acid decarboxylase 67, an enzyme that catalyzes the decarboxylation of glutamate to gamma aminobutyric acid and CO2, particularly in layer 5. In vivo electrophysiological recordings confirmed a decrease in delta, theta, and beta oscillation power in DCF1-knockout mice. This study presents a previously unknown function of DCF1 in V1, suggests an unknown contac…

TsTX-VII, a new toxin from Tityus serrulatus scorpion venom able to induce the release of neurotransmitters from rat brain synaptosomes not blocked by tetrodotoxin.

A procedure for the isolation of the toxin Tityustoxin VII (TsTX-VII) from Tityus serrulatus scorpion venom and its biochemical characterization is reported. This protein has a M(r) = 6,700-6,800, eight half-cystine residues accounting for four disulfide bridges and no His. Its N-terminal sequence GHZGYGS ... characterizes it as a new toxin, able to release glutamic acid and gamma aminobutyric acid from rat brain synaptosomes "in vitro". This release was also induced by the whole venom. Tetrodotoxin however blocked the effect of the whole venom but not that of TsTX-VII, thus suggesting that th…

Novel insight into the evolution of volatile compounds during dynamic freeze-drying of Ziziphus jujuba cv. Huizao based on GC-MS combined with multivariate data analysis.

To understand the evolution of aroma in jujubes during dynamic freeze drying (FD), the relationship between aroma compounds, precursors, and related enzyme activities were analyzed. Fifty-three volatiles were identified during FD processing. After FD, the total aroma contents were increased from 11,004 to 14,603 μg/kg, ketones content was significantly decreased by 54.11 %, resulted in the loss of creamy note in freeze-dried jujube (FDJ). Through the network analysis, serine, glycine, proline, valine, cysteine, arginine, glutamic acid, lysine and leucine had the significant correlation with py…

Plasma amino acids in reptiles after feeding protein or amino acids and after injecting amino acids.

Turtles (Pseudemys scripta elegans) and alligators (Alligator mississipiensis) were fed fish, casein, or free amino acid mixtures, or were i … crease in essential amino acids and considerable rises in four nonessential amino acids, glutamine, glutamic acid, glycine and alanine. Despite some species variation, these two species with low metabolic rates apparently utilized protein synthesis to dispose of amino acid mixtures prior to the necessarily slow process of catabolism for energy.

Molecular cloning of cyclic nucleotide-gated cation channel subunits from rat pineal gland.

We have investigated the subunit composition and the molecular structure of the CNG channel expressed in rat pineal gland. Three types of subunits have been cloned: an alpha-subunit (CNG1), two beta-subunit splice variants (rCNG4.1 and rCNG4.2) and a hydrophilic glutamic acid-rich protein (rGARP). In situ hybridization with sections of rat brain revealed the co-expression of CNG1, CNG4 and GARP in pinealocytes. In addition, CNG4-specific transcripts were detected in the arcuate, periventricular and supraoptic nuclei of the hypothalamus.

Suicidal ingestion of isoniazid: an uncommon cause of metabolic acidosis and seizures.

Despite the widespread use of isoniazid, suicidal ingestion is rare. Two patients are presented who ingested 5 gm and 12 gm respectively, both having seizures within two hours and severe metabolic adisosis. They were treated successfully with intravenous administration of diazepam and bicarbonate, and forced diuresis. Both patients showed a mild rise in levels of serum glutamic oxaloacetic transaminase and lactic acid dehydrogenase. Physicians should be alerted to the possibility of isoniazid ingestion in patients with an unexplained severe metabolic acidosis and seizures.

Novel UGT1A1 Gene Mutations in a Boy with Crigler-Najjar Syndrome Type II.

In this article, we reported a patient with Crigler-Najjar syndrome type II with high-unconjugated bilirubin levels that decreased after phenobarbital treatment. The patient had two novel missense mutations in the UGT1A1 gene and a promoter variant in one allele. One mutation was c.1001T > C, that predicted leucine to proline substitution at position 334 (p.Leu334Pro). The other, c.1139A > G, predicted glutamic acid to glycine replacement at position 380 (p.Glu380Gly). In silico analysis indicated that both mutations are likely pathogenic.

Early onset of diabetes mellitus associated with the mitochondrial DNA T14709C point mutation: patient report and literature review.

We report a family in which a mother and son were affected with diabetes mellitus and myopathy characterized by ragged red fibers and suggestive of mitochondrial disease. Mitochondrial DNA (mtDNA) analysis of DNA isolated from peripheral blood showed a T-->C point mutation at nucleotide position 14709, in the transfer RNA gene for glutamic acid. We review the association of diabetes and mtDNA mutations. This child's case is unusual because of the early onset of diabetes, which is more typical of mtDNA deletions.

Effect of pyridoxine deficiency in the neuronally mature rat.

Pyridoxine deficiency was induced in neuronally mature rats by combination of dietary deprivation and ingestion of 4-deoxypyridoxine. Overt signs of deficiency were evident. At this time the activity of glutamic acid decarboxylase as also the content of serotonin decreased in several areas of the brain. Catecholamine contents were unchanged from controls. Light microscopic observations indicated no difference in CNS myelin between the deficient and control animals. There was no difference between deficient and control groups in their auditory evoked responses.

In-vivo microdialysis study of extracellular glutamate response to temperature variance in subarachnoid hemorrhage.

Neurochemical changes may precede the development of clinical signs in neurological disease. Early identification of such changes may offer an opportunity to avoid or treat complications. Under experimental conditions, extracellular levels of glutamate and other amino acids can be monitored by in-vivo microdialysis in cerebral ischemia, head trauma and epilepsy. Data on the release of glutamate under ischemic conditions in humans are limited. There is no published data on the effects of temperature variation or other manipulations on the extracellular glutamate levels in humans. We report for

Effects of ketamine and thiopental on ischemia reoxygenation-induced LDH leakage and amino acid release from rat striatal slices.

Increased release of glutamate is thought to contribute to ischemia-induced neuronal damage. Since general anesthetics such as thiopental and ketamine are thought to provide some degree of cerebral protection, this study was intended to 1) compare the effectiveness of ketamine and thiopental on ischemia-induced tissue damage; and, if so, 2) determine whether attenuation of the increased amino acid release is the sole mechanism for the protective effects demonstrated. Striatal slices prepared from Wistar Albino rats were incubated in an ischemic medium for 1 hour followed by 5 hours in a reoxyg

Sample preparation for the metabolomics investigation of poly-gamma-glutamate-producing Bacillus licheniformis by GC-MS.

Metabolomics aims to analyze global intracellular metabolites of organisms. To study the intracellular metabolites of poly-γ-glutamate-producing Bacillus licheniformis, the quenching and extraction conditions were assessed and optimized. It indicated that perchloric acid was an appropriate quenching solution for B. licheniformis by measuring livability of cells, leakage of ATP, energy charge and intracellular metabolites. 0.85% NaCl was an appropriate washing solvent for a sample because it did not lead to serious leakage and would not affect operation of GC-MS. Among the four different extrac

Neurotransmitter-bound bestrophin channel structures reveal small molecule drug targeting sites for disease treatment.

Best1 and Best2 are two members of the bestrophin family of anion channels critically involved in the prevention of retinal degeneration and maintenance of intraocular pressure, respectively. Here, we solved glutamate- and γ-aminobutyric acid (GABA)-bound Best2 structures, which delineate an intracellular glutamate binding site and an extracellular GABA binding site on Best2, respectively, identified extracellular GABA as a permeable activator of Best2, and elucidated the co-regulation of Best2 by glutamate, GABA and glutamine synthetase in vivo. We further identified multiple small molecules

Nicotinamide adenine dinucleotides mimic adenosine inhibition on synaptic transmission by decreasing glutamate release in rat hippocampal slices.

To assess the possible inhibitory action of nicotinamide adenine dinucleotides on the synaptic release of glutamate, electrophysiological and biochemical experiments were performed on rat hippocampal slices. Perfusion of adenosine, beta-nicotinamide adenine dinucleotide (NAD) or beta-nicotinamide adenine dinucleotide phosphate (NADP), reversibly inhibited the field excitatory postsynaptic potentials (fEPSP). Dose-response curves for their inhibitory action showed that these three substances had a similar potency in the range of concentrations from 0.1 microM to 100 microM. NADP and adenosine (

Subtype-specific involvement of metabotropic glutamate receptors in two forms of long-term potentiation in the dentate gyrus of freely moving rats.

In this study, the role of metabotropic glutamate receptors in N-methyl-D-aspartate receptor-dependent and voltage-gated calcium channel-dependent long-term potentiation in the dentate gyrus of freely moving rats was investigated. Antagonists for group 1 metabotropic glutamate receptors ((S)-4-carboxyphenylglycine), group 1/2 metabotropic glutamate receptors ((RS)-alpha-methyl-4-carboxyphenylglycine) and group 2 metabotropic glutamate receptors ((RS)-alpha-methylserine O-phosphate monophenylester) were used. Oligodendrocyte excitotoxicity determined by local glutamate accumulation and mitochondrial function.

Developing oligodendrocytes (OL precursors, pre-OLs) express alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) subtype glutamate receptors (AMPARs) and are highly vulnerable to hypoxic-ischemic or oxygen-glucose deprivation (OGD)-induced excitotoxic injury, yet the mechanisms of injury remain unclear. Here we investigated the role of glutamate accumulation and mitochondrial function in OGD-induced pre-OL toxicity in vitro. Bulk glutamate concentration in the culture medium did not increase during OGD and OGD-conditioned medium did not transfer toxicity to naïve cells. Facilitatio
