SUMOylation and ubiquitination reciprocally regulate SMCHD1 antiviral activity against herpes simplex virus 1.

Host restriction factors serve as intrinsic barriers against viral infection, and are frequently counteracted by viral antagonists. Previous studies, including our own, have identified SMCHD1 as a restriction factor that suppresses the replication of multiple viruses. Here, we reveal that the antiviral activity of SMCHD1 is dynamically regulated by two different post-translational modifications. SUMOylation of SMCHD1 promotes its association with the viral genome and enhances its antiviral activity. In contrast, during herpes simplex virus 1 (HSV-1) infection, the viral E3 ligase ICP0 induces …

Streptococcus mutans-derived extracellular vesicles promote herpes simplex virus infection in oral epithelia.

The oral cavity contains the second most diverse bacterial community after the intestines, with bacteria and viruses coexist. Streptococcus mutans is a major pathogenic bacterium in the oral cavity, commonly associated with dental caries. We investigated the effects of S. mutans-derived extracellular vesicles (Sm EVs) on herpes simplex virus 1 (HSV-1) infection, which is prevalent in the oral cavity. We performed our experiments in human oral keratinocyte (HOK) cells and mucosal tissue-derived organoids, and analyzed human whole saliva (n = 50) for associations between S. mutans and HSV-1 enve…

Herpes Simplex Virus 1 in Trigeminal Ganglia of Trafficked Neotropical Primates, Peru, 2024.

We detected herpes simplex virus 1 in the trigeminal ganglia of trafficked Neotropical primates (1 Aotus azarai; 3 Sapajus macrocephalus) in Peru. Tests also revealed Saimiriine alphaherpesvirus 1 in the trigeminal ganglia of 2 Saimiri sp. monkeys. Our findings suggest latency and raise concerns about diagnostic standards, viral reactivation, and spillover risks.

Bufalin mitigates corneal damage in herpes simplex keratitis mice via inhibiting viral replication.

BACKGROUND: Herpes simplex keratitis (HSK) is an ocular viral infectious disease with limited therapeutic options and increasing resistance, leading cause of infectious blindness worldwide. Bufalin is a cardiotonic steroid, more specifically a bufadienolide, which can be found in many plant or animal species, but their main sources are skin and parotid gland secretions of toads (such as Bufo bufo gargarizans Cantor), which constitutes the traditional Chinese medicine preparation known as Chan Su (Venenum Bufonis). Despite its broad pharmacological activities, the potential role of bufalin in c…

Herpes simplex virus and kallikrein-related peptidases - a functional connection in Alzheimer's disease.

BACKGROUND: The viral hypothesis of Alzheimer's disease (AD) proposes that neurotropic pathogens may interact with amyloid and tau pathology, but the evidence remains controversial. We investigated whether cerebrospinal fluid (CSF) kallikrein-related peptidase 6 (KLK6) is associated with AD biomarkers and with an intrathecal anti-HSV immune response in patients with AD, and whether HSV-1 infection alters KLK6 expression in cell culture models. METHODS: CSF KLKs and AD biomarkers were analyzed in patients with AD. HSV-1 serostatus and anti-HSV-1/2 CSF-to-serum antibody index (HSV-AI) were used

Tongue squamous cell carcinoma masked by herpes simplex virus infection: A case report.

Herpes simplex virus (HSV) infection can potentially mask underlying malignancies, complicating clinical diagnosis and potentially delaying the detection of a serious pathology. The present study describes the case of a 37-year-old man with a 20-year smoking history that presented with a tongue ulcer masked by HSV infection, who underwent comprehensive diagnostic investigations. Initial histopathological examination revealed characteristic HSV infection features, including multinucleation and intercellular bridge destruction. Despite symptomatic improvement of the viral infection, persistent l…

Hexokinase-2 as a Therapeutic Target: Alleviating Herpes Simplex Keratitis Through Metabolic Reprogramming.

Herpes simplex keratitis (HSK) is a leading infectious cause of blindness worldwide, with current therapies primarily targeting viral replication rather than addressing host-cell injury. RNA sequencing of corneal tissue from HSK patients and healthy donors identifies a metabolic shift from mitochondrial oxidative phosphorylation to aerobic glycolysis. Notably, hexokinase-2 (HK2), a pivotal glycolytic enzyme, exhibits the greatest up-regulation, coinciding with a marked reduction in the activity of mitochondrial respiratory chain complexes in HSK corneas. Pharmacological inhibition of HK2 with …

Tackling cutaneous herpes simplex virus disease with topical immunomodulators-a call to action.

SUMMARYAntivirals play important roles in restricting viral diseases. Nevertheless, they act on a relatively limited number of viruses and occasionally display partial effectiveness in some tissues or against escape variants. Although vaccination remains the most cost-effective approach for preventing microbial diseases, developing prophylactic or therapeutic solutions for pathogens, such as herpes simplex viruses (HSVs), that effectively reduce their clinical manifestations in the skin has proven exceptionally challenging despite extensive research. Alternatively, a less explored approach for…

Oncolytic herpes simplex virus expressing IL-2 controls glioblastoma growth and improves survival.

BACKGROUND: Glioblastoma (GBM), a highly immunosuppressive and often fatal primary brain tumor, lacks effective treatment options. GBMs contain a subpopulation of GBM stem-like cells (GSCs) that play a central role in tumor initiation, progression, and treatment resistance. Oncolytic viruses, especially oncolytic herpes simplex virus (oHSV), replicate selectively in cancer cells and trigger antitumor immunity-a phenomenon termed the "in situ vaccine" effect. Although talimogene laherparepvec (T-VEC), an oHSV armed with granulocyte macrophage-colony stimulating factor (GM-CSF), is Food and Drug…

Acute generalised exanthematous pustulosis and herpes simplex virus: a case of possible overlap.

Acute generalised exanthematous pustulosis (AGEP) is a rare cutaneous disorder that presents with numerous non-follicular, pinpoint sterile pustules on a background of oedematous erythema that can coalesce, leading to desquamation. 90% of cases are triggered by medications, most often with antibiotics as the culprit. However, other triggers including viral infection have also been reported. Herpes simplex virus (HSV) as a viral trigger has not been previously explored. Here, we present a case of AGEP caused by bupropion, followed by a second presentation of assumed acute limited exanthematous …

Synthetic Naphthoquinone Inhibits Herpes Simplex Virus Type-1 Replication Targeting Na+, K+ ATPase.

Since 1970 acyclovir (ACV) has been the reference drug in treating herpes simplex virus (HSV) infections. However, resistant herpes simplex virus type 1 (HSV-1) strains have emerged, narrowing the treatment efficacy. The antiviral activity of classical Na+, K+ ATPase enzyme (NKA) inhibitors linked the viral replication to the NKA's activity. Herein, we evaluated the anti-HSV-1 activity of synthetic naphthoquinones, correlating their antiviral activity with NKA inhibition. We tested seven synthetic naphthoquinones initially at 50 μM on HSV-1-infected African green monkey kidney cells (VERO cell…

Hemophagocytic Lymphohistiocytosis Triggered by Herpes Simplex Virus 1 and 2: A Narrative Review.

Introduction: Hemophagocytic lymphohistiocytosis (HLH) is a rare, life-threatening syndrome characterized by an uncontrolled hyperinflammatory reaction. HLH is classified into primary (familial) and secondary (acquired). Secondary HLH is commonly triggered by infections, with viral infections being a leading cause. Its epidemiology and clinical features in cases associated with herpes simplex virus 1 and 2 remain underexplored. This study aimed to review all previously described cases of HSV-1 or -2-triggered HLH and provide information about this syndrome's epidemiology, microbiology, clinica…

A Neurosyphilis Case Mimicking Herpes Simplex Virus Encephalitis in an African American Male.

Neurosyphilis is one form of a multisystemic sexually transmitted disease caused by Treponema pallidum. Although typical presentations of neurosyphilis have become less common in the post-antibiotic era, a rising trend of atypical presentations can mimic other diagnoses like herpes simplex and autoimmune encephalitis. In this case, we diagnosed neurosyphilis in a patient with clinical and radiological features similar to herpes simplex encephalitis. We emphasize the need for a diagnostic approach combining imaging namely MRI, lumbar puncture, and use of treponemal and non-treponemal tests so t…

A Rare Association Between Herpes Simplex Virus Type 1 and Miller-Fisher Syndrome.

The etiopathogenesis for Guillain-Barré syndrome (GBS) and Miller-Fisher syndrome (MFS), a variant of GBS, is well-documented in the literature. However, the association between MFS and an underlying herpes simplex virus type 1 (HSV-1) infection is very limited. We present a unique case of a 48-year-old man who developed diplopia, bilateral ptosis, and gait instability following an acute diarrheal illness and recurring cold sores. The patient was diagnosed with MFS precipitated by recurrent HSV-1 infection following a Campylobacter jejuni acute infection. The diagnosis of MFS was supported by …

Neonatal herpes simplex virus infection: From the maternal infection to the child outcome.

This review examines the recent literature on the management of herpes simplex virus (HSV) infections in neonates. We summarized the three clinical categories of maternal HSV infection during pregnancy (primary first episode, nonprimary first episode, or recurrent episode) and the mechanisms of fetal damage. Considering when the transmission of the infection from the mother to the fetus/newborn occurs, three types of neonatal infection can be distinguished: intrauterine infection (5% of cases), postnatal infection (10% of cases), and perinatal infections (85% of cases). Neonatal presentation c…

Takotsubo cardiomyopathy associated with Herpes simplex encephalitis: A case report and literature review.

Takotsubo cardiomyopathy (TCM) is a stress-induced cardiomyopathy characterized by transient left ventricular dysfunction. It can be triggered by various central nervous system pathologies including status epilepticus (SE) and N-methyl-d-aspartate receptor (NMDAr) encephalitis. Herpex simplex encephalitis (HSE) is a life-threatening, sporadic, encephalitis associated with focal or global cerebral dysfunction caused by herpes simplex viruses type 1(HSV-1), or less commonly, type 2(HSV-2). While approximately 20% of patients with HSE develop NMDAr antibodies, not all manifest with encephalitis c…

Associations between host genetic variants and Herpes Simplex Labialis in the TwinsUK cohort.

OBJECTIVE: Previous literature supports controversial results about the role of host genetic factors in the reactivation of Herpes Simplex Labialis (HSL). The objective of this study is to explore the association between different putative single nucleotide polymorphisms (SNPs) and self-reported HSL reactivation in the TwinsUK cohort. DESIGN: 1761 participant were recruited for this study from the TwinsUK register. Blood collection and HSL questionnaire were completed for a subset of 595 participants (age 61.9, dizygotic twins 62.8%). Blood was stored at -80 °C and DNA analysed from all sampl…
