Arabinoxylan Hydrogels as a Novel Encapsulation System with Triggered Release by Gut Bacterial Enzymes.

Arabinoxylans are ubiquitous plant biopolymers present in large amounts in cereal bran, a major underutilized side stream of the agrifood industry. Their unique structural properties make them promising matrices for the design of hydrogels for nutritional and pharmacological applications. We report the controlled production of laccase-crosslinked arabinoxylan hydrogels derived from wheat (WAX) and rye (RAX) bran with tuned porous properties via the modulation of freeze-drying temperature, with implications for biostimulatory encapsulation and release. Freezing in liquid nitrogen generated smal…

In situ ECM-functionalized hyaluronic acid/dextran composite hydrogels for regenerative soft tissue reconstruction.

Conventional injectable soft-tissue fillers act mainly as inert volumizers and often trigger foreign-body reactions. They lack biological cu … ctability, the highest elastic modulus (≈576 Pa), and enhanced cell proliferation (147% viability). Hydrogels releasing AA supported markedly greater total protein (0.481 ± 0.013 μg/mL) and collagen deposition than AA-free gels. In vivo, the material degraded steadily over 21 days with complete inflammation resolution, showing no chronic fibrosis. Important …

Biocompatible Gelatinized Starch as a Functional Modulator to Expand the Scope of Injectable Hydrogels.

Injectable hydrogels used in tissue engineering and minimally invasive therapies are limited by insufficient biocompatibility, poor scalability, high costs, and inconsistent performance. Here, we introduce gelatinized starch, a common polysaccharide, as a versatile and economical building block to expand the existing repertoire of injectable hydrogel materials. Derived from native corn starch via heating and gelatinization, this biopolymer exhibits shear-thinning, thixotropic, and thermoreversible sol-gel transition properties, key rheological characteristics essential for syringeability and i…

Compartmentalized Dynamic Supramolecular Hydrogels through Chemically Fueled Self-Assembly in Aqueous Two-Phase Systems.

Compartmentalized dynamic supramolecular structures are prevalent in living systems and underpin numerous pivotal biological functions, yet their recapitulation in synthetic scenarios remains largely underexplored. Here, we report on compartmentalized supramolecular hydrogels that can dynamically evolve their structure and properties through chemically fueled self-assembly in aqueous two-phase systems (ATPSs) and uncover an unexpected dynamic interactive behavior between the hydrogel networks and ATPSs. By chemically fueling the self-assembly of a simple peptide-based gelator throughout aqueou…

Bioinspired Anisotropic Collagen-Based Conductive Hydrogels Promote Neuronal Differentiation via Activation of Mechanoelectrical Signaling.

Simulating the anisotropic structure and electromechanical signals of the natural neural extracellular matrix (ECM) is crucial for advancing neural regeneration and constructing in vitro research models. Herein, a three-dimensional collagen-based conductive hydrogel with oriented fibers was constructed by controlling the intrinsic assembly behavior of collagen by combining mechanical pre-deformation with magnetic-driven stretching during gelation. The anisotropic hydrogel exhibited excellent cell compatibility and could guide the orientation and extension of PC-12 cell axons along the fibers.

Pseudocapacitive Charge-Driven Adhesive-Electrocoupling Hydrogels for Myocardial Infarction Repair via Interstitial Microenvironment Remodeling.

Myocardial infarction (MI) creates a hostile microenvironment characterized by oxidative stress, inflammation, fibrosis, and electrical conduction block, posing a multifaceted challenge for tissue repair. To address this, we developed a multifunctional hydrogel (Gel-PDZC), through an artificial neural network (ANN)-guided optimization process, incorporating nanoparticles comprising a ZIF-8@CaO2 core for sustained oxygen release, a polydopamine (PDA) interlayer for adhesion and antioxidant activity, and a conductive poly(3,4-ethylenedioxythiophene) (PEDOT) shell. The hydrogel exhibits a unique

Macroporous Poly-N-Isopropylacrylamide and Cationic Co-Polymer Hydrogels: Long-Term Subcutaneous Implant Biocompatibility.

Biomaterials have been developed with a great potential in tissue engineering. Hydrogels, especially those based on poly-N-isopropylacrylamide (PNIPAM), have innate mechanical and physicochemical similarities with the native extracellular matrix. We have previously reported the in Vitro compatibility of PNIPAM and its copolymers with a varied number of cell lines, displaying good proliferation rates and bioadhesive capacity. However, the in Vivo biocompatibility assessment is necessary to evaluate. In this work, biocompatibility and immunological acceptance of macroporous hydrogels based on PN…

Thermoresponsive, Shear-Thinning Supramolecular Hydrogels from Peptide-Polymer Conjugates for Bioinspired 3D Cell Culture.

Physically assembled, supramolecular hydrogels often are formed by weak bonds, producing useful viscoelastic properties that are reminiscent of soft tissues within the human body yet presenting challenges in stability under physiological conditions. In this work, we establish an approach for creating self-assembling, supramolecular hydrogels based on a peptide-polymer conjugate (PPC) as a three-dimensional (3D) cell culture platform. The viscoelastic properties of these hydrogels, including thermoresponsiveness and shear-thinning, were measured using rheometry. Building on these observations, …

Photopolymerized antimicrobial hydrogels based on thiol-functionalized MXenes and in-situ generated silver nanoparticles.

Here, we report photopolymerized composite hydrogels integrating thiol-functionalized MXene nanosheets with in situ generated silver nanoparticles (AgNPs) to obtain structurally reinforced antimicrobial films. Thiol functionalities were covalently grafted onto the MXene surface to provide a sulfur-containing interfacial environment that may influence dispersion, local Ag-species coordination, and nanoparticle organization while strengthening interfacial interactions within the hydrogel matrix. The hydrogels were fabricated by UV-initiated free-radical polymerization of N-vinyl pyrrolidone (NVP…

Redefining gingival substitutes: Enhanced mechanics and bioactivity in SVV-functionalised bioprinted CollMA/HAMA hydrogels.

Bioactive hydrogels with tuneable mechanical properties were engineered to support endothelial cell-driven pre-vascular network formation, a critical step toward ensuring adequate oxygen and nutrient diffusion in engineered tissues. These hydrogels were subsequently adapted for 3D bioprinting and functionalised with pro-angiogenic peptides (SVV and QK) to promote the formation of interconnected vascular networks maintained over a 14-day in vitro pre-incubation period, a prerequisite for subsequent host anastomosis. Several composite HAMA/CollMA bioinks were developed, including 0.1% CollMA/1% …
