Reproducibility and relation to specific and non-specific anti-tumor resistance of the tumor "sneaking through" phenomenon.

The reproducibility and immunological specificity of the tumor "sneaking through" phenomenon and enhancement of tumor growth were studied in syngeneic and random-bred Syrian hamsters by means of a quantitative modification of the transplantation test. After primary challenge the phenomenon was neither observed in normal animals nor in animals effectively immunized against tumor. However, it was regularly observed in some "immune" animals after secondary challenge. In primary challenge of animals "sneaking through" phenomenon was most often observed in animals pretreated with large doses of hea

6. Secondary immunodeficiencies, including HIV infection.

The immune system can be adversely affected by a variety of extrinsic factors, including immunosuppressive drugs, exposure to harsh environmental conditions, hereditary disorders other than primary immunodeficiencies, and acquired metabolic disorders such as diabetic mellitus, with all of these resulting in conditions known as secondary immunodeficiencies. Perhaps the most well known secondary immunodeficiency is caused by HIV infection; however, the most prevalent cause of immunodeficiency worldwide is severe malnutrition, which affects as much as 50% of the population in some impoverished co

A Bivalent Vaccine Based on a PB2-Knockout Influenza Virus Protects Mice From Secondary Pneumococcal Pneumonia.

BACKGROUND: Secondary bacterial infections after influenza can be a serious problem, especially in young children and the elderly, yet the efficacy of current vaccines is limited. Earlier work demonstrated that a replication-incompetent PB2-knockout (PB2-KO) influenza virus possessing a foreign gene in the coding region of its PB2 segment can serve as a platform for a bivalent vaccine. METHODS: In the current study, we generated the PB2-KO virus expressing pneumococcal surface protein A (PspA), PB2-KO-PspA virus, the replication of which is restricted to PB2-expressing cells. We then examined

Immunoglobulin M antibody response to measles virus following primary and secondary vaccination and natural virus infection.

The use of IgM antibody detection for the classification of the primary and secondary measles antibody response in persons following primary and secondary vaccination and natural measles virus infection was examined. Of 32 nonimmune children receiving primary measles vaccination, 31 (97%) developed IgM antibodies, consistent with a primary antibody response. Of 21 previously vaccinated children with low levels of preexisting IgG antibodies who responded to revaccination, none developed detectable IgM antibodies, whereas 33 of 35 (94%) with no detectable preexisting IgG antibodies developed an

Secondary attack rate of hepatitis A, varicella and mumps in household settings and reliability of family history to detect seronegative children for necessity of vaccination.

Hepatitis A, varicella, and mumps are common in countries without universal immunization against these infections. Information about secondary household attack rates is limited. A prospective study was conducted in children <16 y of age who had household contact with the above infections. History for previous infections and vaccination was obtained from the parents. Specific antibodies in the sera during enrollment and at the end of the longest incubation periods were determined. A total of 113 children who had household contact with cases of varicella, hepatitis A, or mumps were enrolled. Sec

Differential effects of T-lymphocyte-derived soluble factor on virgin and primed B lymphocytes in vitro.

Cell-free supernatants from mixed leukocyte cultures derived from histoincompatible mouse strains markedly enhanced the in vitro immune response to SRBC by splenocytes from allogeneic mice. The supernatant factor or factors from allogeneic spleen cell cultures appeared to preferentially stimulate antigen-sensitized B lymphocytes, especially those involved in secondary IgG antibody formation. Furthermore, as shown in the present study, the enhancing supernatant factor or factors had no effect on the true primary in vitro immune response to Vibrio cholerae antigen. Normal spleen cells cultured i

Cell-mediated and humoral immune responses of cyprinids induced by a live attenuated vaccine against cyprinid herpesvirus 2 infection in comparison to the virus non-permissive high temperature water treatment.

The live attenuated vaccine (P7-P8 strain) against cyprinid herpesvirus 2 (CyHV-2) infection of goldfish shows high protective efficacy. However, the underlying immune mechanism induced by P7-P8 vaccination remains unknown. It is known that the fish survived in the primary infection with CyHV-2 by the virus non-permissive high temperature (HT) water treatment elicit immunity against secondary virus challenge. In this study, the immunity induced by the P7-P8 vaccine was compared with that by HT treatment. To further explore the immunological responses of cyprinids, in addition to goldfish Caras

Protective effects of booster dose of SARS-COV-2 vaccination against post-acute COVID-19 syndrome: A systematic review.

The global impact of COVID-19, caused by SARS-CoV-2, has extended beyond acute infection, with post-acute COVID-19 syndrome (PACS) affecting an estimated 10% of recovered individuals. PACS manifests a range of debilitating symptoms, including fatigue, cognitive impairment, and gastrointestinal issues. While vaccination has proven effective in mitigating severe COVID-19 outcomes, the role of booster doses in preventing PACS remains unclear. This study aimed to evaluate whether COVID-19 booster vaccinations reduce the incidence and severity of PACS in individuals with prior SARS-CoV-2 infection.

Revitalizing COVID-19 Vaccination in the Post-Pandemic Phase: A Public Health Priority.

Despite the end of the global emergency, SARS-CoV-2 continues to circulate. The waning of post-infection and vaccine-induced immunity highlights the need to sustain booster uptake. Vaccination remains effective in preventing severe disease and, together with viral evolution, has contributed to milder clinical presentations. Yet, vaccine fatigue and misinformation have weakened adherence, especially among youth. Strengthening vaccination campaigns through updated boosters and communication strategies using channels most followed by young people is essential to maintain immunity and prevent futu

Economic evaluation of the second booster dose of diphtheria-tetanus-acellular pertussis vaccine for children aged 6 years in Zhejiang Province: A cost-utility analysis.

Despite increasing pertussis breakthroughs and waning immunity, this study evaluates the cost-effectiveness of replacing the DT booster with DTaP in 6-year-olds in Zhejiang, China. While implemented in high-income regions, this strategy's cost-effectiveness remains unassessed in diverse Chinese settings. We address this gap using local data.Using surveillance data from the Zhejiang Provincial Center for Disease Control and Prevention, literature reviews, and a 10-year simulation timeframe, a decision tree-Markov model was constructed from a societal perspective. The study cohort was 6-year-old

Vaccine effectiveness against transmission of alpha, delta and omicron SARS-COV-2-infection, Belgian contact tracing, 2021-2022.

OBJECTIVES: Vaccine effectiveness against transmission (VET) of SARS-CoV-2-infection can be estimated from secondary attack rates observed during contact tracing. We estimated VET, the vaccine-effect on infectiousness of the index case and susceptibility of the high-risk exposure contact (HREC). METHODS: We fitted RT-PCR-test results from HREC to immunity status (vaccine schedule, prior infection, time since last immunity-conferring event), age, sex, calendar week of sampling, household, background positivity rate and dominant VOC using a multilevel Bayesian regression-model. We included Belg

Safety and immunogenicity of ChAdOx1 85A prime followed by MVA85A boost compared with BCG revaccination among Ugandan adolescents who received BCG at birth: a randomised, open-label trial.

BACKGROUND: BCG confers reduced, variable protection against pulmonary tuberculosis. A more effective vaccine is needed. We evaluated the safety and immunogenicity of candidate regimen ChAdOx1 85A-MVA85A compared with BCG revaccination among Ugandan adolescents. METHODS: After ChAdOx1 85A dose escalation and age de-escalation, we did a randomised open-label phase 2a trial among healthy adolescents aged 12-17 years, who were BCG vaccinated at birth, without evident tuberculosis exposure, in Entebbe, Uganda. Participants were randomly assigned (1:1) using a block size of 6, to ChAdOx1 85A follo

Heterologous prime-boost immunization induces protection against dengue virus infection in cynomolgus macaques.

Currently licensed dengue vaccines do not induce long-term protection in children without previous exposure to dengue viruses in nature. These vaccines are based on selected attenuated strains of the four dengue serotypes and employed in combination for two or three consecutive doses. In our search for a better dengue vaccine candidate, live attenuated strains were followed by non-infectious virus-like particles or the plasmids that generate these particles upon injection into the body. This heterologous prime-boost immunization induced elevated levels of virus-specific antibodies and helped t

Durable Expansion of TCR-δ Meta-Clonotypes After BCG Revaccination in Humans.

Mycobacterium bovis bacille Calmette-Guérin (BCG) has been used for 100 years and prevents disseminated tuberculosis and death in young children. However, it shows only partial efficacy against pulmonary tuberculosis (TB) in adults, so new vaccines are urgently needed. The protective efficacy of BCG depends on T cells, which are typically activated by pathogen-derived protein antigens that bind to highly polymorphic major histocompatibility complex (MHC) molecules. Some T cells recognize non-protein antigens via antigen presenting systems that are independent of genetic background, leading to

Boosters and time from the last anti-COVID-19 vaccine dose: lead public health choices by real-time epidemiological assessment.

BACKGROUND: the levels of anti-SARS-CoV-2 antibodies after the second vaccine dose decline in the following months; the administration of an additional vaccine dose (booster) is able to restore the immune system in the short period significantly reducing the risk of a severe disease. In the winter of 2021, a new particularly infectious variant caused the urgent need to increase the coverage of the booster dose. OBJECTIVES: to present, using real data, an evaluation of the efficacy of the booster dose in reducing the severe disease of SARS-CoV-2 infection in terms of hospital admissions, inten
