Modulating CD38 enzymatic activity during antibody-based immunotherapy in multiple myeloma: a basic science perspective.

INTRODUCTION: Multiple myeloma (MM) develops within a profoundly immunosuppressive bone marrow (BM) microenvironment. CD38, a multifunctiona … M cells and a representative MM cell line, and (iii) the in vivo dynamics of ADO and its metabolite inosine (INO) in BM and peripheral blood (PB) plasma from MM patients receiving DARA monotherapy. RESULTS: In vitro, both DARA and ISA promoted NAD⁺ degradation with accumulation of ADPR. In vivo, ADO concentrations in BM plasma remained consistently in th …

ADAR regulates APOL1 via A-to-I RNA editing by inhibition of MDA5 activation in a paradoxical biological circuit.

We studied the effects of ADAR expression and A-to-I editing on APOL1 levels in podocytes, human kidney tissue, and a transgenic APOL1 mouse model. In interferon-γ (IFN-γ)-stimulated human pod …

Intracellular calcium signalling and vascular reactivity in Bartter's syndrome.

We investigated patients affected by Bartter's syndrome in the attempt to localize the intracellular defect mediating the reduced intracellular Ca2+ mobilization that may be responsible for the decreased vascular reactivity characteristic of Bartter's syndrome. Using the formylmethionyl-leucyl-phenylalanine (fMLP) receptor system, which causes, intracellular calcium release, we investigated fMLP-stimulated intracellular inositol 1,4,5-trisphosphate (IP3) production as well as the number and affinity of fMLP receptors in neutrophils from Bartter's syndrome patients and healthy controls. Trihexyphenidyl and isoprinosine in the treatment of subacute sclerosing panencephalitis.

Six cases of subacute sclerosing panencephalitis (1 stage I, 3 stage II, 2 stage III) were diagnosed at our institution in the last 10 years. Five patients were treated with isoprinosine and the antiepileptic drug valproic acid. Three patients presented with myoclonic seizures refractory to valproic acid and the usual antiepileptic therapy. They received trihexyphenidyl with good results. We suggest the use of trihexyphenidyl in combination with isoprinosine in patients with subacute sclerosing panencephalitis with myoclonic seizures refractory to valproic acid.

Cellular immunity (T-cell subset using monoclonal antibody) in tuberculosis, melioidosis, pasteurellosis, penicilliosis; and role of levamisole and isoprinosine.

In vitro and in vivo evaluation of cellular immunity (T-cell subset--using monoclonal antibody--immunofluorescent staining technique; E-rosette formation to 2.4-Dinitrochlorobenzene reaction) were made in tuberculosis, melioidosis, pasteurellosis and penicilliosis. The data demonstrate depressed cellular immunity especially a significant decrease in numbers of T-helper cells and T-helper/T-suppressor subset ratio fell to less than 1.0. Of greater importance, in tuberculous patients and patients with melioidosis treated with levamisole, 150 mg/day, twice a week, along with a specific antituberc

Lymphocyte subpopulations in blood and cerebrospinal fluid from patients with subacute sclerosing panencephalitis.

Lymphocyte subpopulations in peripheral blood (PB) and cerebrospinal fluid (CSF) from 26 children affected with subacute sclerosing panencep … bulins in the central nervous system. Longitudinal studies performed in 6 SSPE patients during isoprinosine therapy revealed a time-dependent decrement of the %s of E- and EA-RFC in CSF. It cannot be affirmed whether this is related to the disease or to the therapy.

Rice Bran as a Sustainable Feed Material Substitute to Improve the Meat Quality in Late-Stage Wenchang Chicken.

The growing competition between humans and livestock for corn-soybean based feed calls for sustainable alternatives like rice bran. This study assessed substituting feed with rice bran (0%, 5%, 10%, 15%, 20%) in diets for slow-growing Wenchang chickens, a prized Chinese yellow-feathered breed. Up to 15% substitution maintained final body weight and average daily gain, but 20% substitution reduced body weight. Feed intake rose significantly at 10-20% substitution. Substituting 15-20% rice bran for feed significantly reduced abdominal fat without affecting slaughter performance or serum biochemi

Dietary OPL-rich structured lipid modulates the nutritional characteristics of developing muscle and is associated with lipid and amino acid metabolic changes in juvenile largemouth bass.

1-Oleoyl-2-palmitoyl-3-linoleoylglycerol (OPL) is a representative sn-2 palmitate-containing triacylglycerol (TAG) relevant to human milk fat and early-life lipid nutrition. However, whether dietary OPL-rich structured lipid affects developing muscle nutritional characteristics and associated metabolic features remains unclear. In this study, juvenile largemouth bass were used as a developmental model to compare dietary OPL-rich oil with a mixed oil (MO), which had a similar total fatty acid composition but a lower sn-2 palmitate content. Dietary OPL-rich structured lipid significantly increas

ADAT2-mediated A-to-I tRNA modification promotes oncogenic translation and colorectal cancer progression and chemoresistance.

BACKGROUND: Adenosine-to-Inosine (A-to-I) modification is one of the most common transfer RNA (tRNA) modifications in humans. However, the role of A-to-I tRNA modification in colorectal cancer (CRC) remains poorly understood. METHODS: tRNA modification was profiled by LC-MS in paired CRC and adjacent normal tissues (N = 70). The clinical significance of adenosine deaminase tRNA specific 2 (ADAT2) was evaluated using in-house (N = 157) and TCGA cohorts (N = 283). The function of ADAT2 in CRC was determined in intestine-specific ADAT2 knockout mice. Mechanism of ADAT2 was assessed by integrated …

ADAR-mediated RNA editing in CNS disorders: from pathogenic mechanisms to therapeutic opportunities.

The adenosine deaminases acting on RNA (ADAR) family of enzymes (ADAR1 and ADAR2) catalyze adenosine-to-inosine (A-to-I) RNA editing. This post-transcriptional change is remarkably prevalent in the central nervous system (CNS). ADAR-mediated editing is critical for proper brain development, synaptic plasticity, and immunological homeostasis in the central nervous system (CNS) via recoding neurotransmitter receptors and ion channels. Conversely, a wide range of CNS disorders, such as neurodegenerative diseases (Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis), neurop…

Depletion of nuclear cytoophidia in Alzheimer's disease.

There is considerable evidence for a role for metabolic dysregulation, including disordered purine nucleotide metabolism, in the pathogenesis of Alzheimer's disease (AD). Purine nucleotide synthesis in the brain is regulated with high fidelity to co-ordinate supply with demand. The assembly of some purine biosynthetic enzymes into linear filamentous aggregates called "cytoophidia" (Gk. Cellular "snakes") represents one post-translational mechanism to regulate enzyme activity. Cytoophidia comprised of the nucleotide biosynthetic enzymes inosine monophosphate dehydrogenase (IMPDH) and phosphorib…
