Mycobacterial DNA-binding protein 1 is critical for BCG survival in stressful environments and simultaneously regulates gene expression.

Survival of the live attenuated Bacillus Calmette-Guérin (BCG) vaccine amidst harsh host environments is key for BCG effectiveness as it allows continuous immune response induction and protection against tuberculosis. Mycobacterial DNA binding protein 1 (MDP1), a nucleoid associated protein, is essential in BCG. However, there is limited knowledge on the extent of MDP1 gene regulation and how this influences BCG survival. Here, we demonstrate that MDP1 conditional knockdown (cKD) BCG grows slower than vector control in vitro, and dies faster upon exposure to antibiotics (bedaquiline) and oxida

Integrative expression profiling and comparative bioinformatics analysis of ALDH7A1 in chickpea (Cicer arietinum L.) under salt and ABA treatments.

BACKGROUND: Soil salinity and abscisic acid (ABA)-related signaling are major constraints affecting chickpea (Cicer arietinum L.) productivity by promoting oxidative stress and aldehyde accumulation. Aldehyde dehydrogenases (ALDHs), particularly members of the ALDH7 family, are involved in aldehyde detoxification and redox homeostasis under adverse conditions. However, the molecular characteristics, expression behavior, and structural features of ALDH7A1 in chickpea under salt and ABA treatments remain insufficiently understood. Therefore, this study aimed to provide an integrative characteriz

Radiation biomarkers in body fluids: translational perspectives for precision radiotherapy-a narrative review.

BACKGROUND AND OBJECTIVE: Radiotherapy and radionuclide therapy are essential components of modern cancer treatment. Despite substantial advances in radiation delivery and treatment planning, considerable interindividual variability remains in therapeutic response and radiation-induced toxicity. Conventional monitoring approaches, including imaging and routine laboratory tests, often provide limited insight into early or systemic biological responses. Body fluid-based biomarkers may provide accessible molecular readouts of radiation-induced damage, host response, and treatment-related toxicity

Spatiotemporal Dissection of the Infection Response to Bacterial Soft Rot in Non-heading Chinese cabbage.

Bacterial soft rot caused by Pectobacterium brasiliense (Pbr) leads to substantial yield losses in cruciferous vegetables, including non-heading Chinese cabbage (pak choi, Brassica rapa subsp. chinensis). This study employed an integrated physiological and multi-tissue transcriptomic approach to decipher the infection response strategies of pak choi against Pbr. Age-dependent resistance was detected, with susceptibility decreasing as plants matured. In susceptible four-week-old plants, Pbr infection induced oxidative stress, characterized by the accumulation of hydrogen peroxide (H₂O₂) and mal

Selective autophagy pathways in ageing and neurodegeneration: Emerging roles for mitophagy and nucleophagy.

Ageing is characterized by the progressive accumulation of molecular and cellular damage, driven by both genetic and nongenetic factors and ultimately leading to functional decline and an increased risk of disease. Convergent evidence indicates that many of the mechanisms influencing ageing are intimately related to the ability of the organism to cope with intrinsic and extrinsic stress. Key determinants of this capacity are the molecular pathways that coordinate stress responses throughout life. Among these, general and cargo-specific autophagy have emerged as integral components of cellular

Insulin resistance: The central node of convergence between unfolded protein response, diabetes, and cancer.

Modern lifestyle patterns, characterized by high-calorie diets and sedentary behaviour, have driven a global surge in obesity, which is a primary driver of insulin resistance (IR) and type 2 diabetes mellitus (T2D). Initially, pancreatic β-cells adapt to IR by increasing proliferation, neogenesis, and insulin secretion to maintain normoglycemia. Also, a prolonged exposure to genetic and environmental stress compromises the β-cell functioning and survival, leading to chronic hyperglycaemia. Beyond its central role in T2D, IR and its metabolic hallmarks, such as hyperinsulinemia, hyperglycaemia,

Crosstalk between endoplasmic reticulum stress and mitochondrial homeostasis: A new perspective on ophthalmic disease treatment.

Endoplasmic reticulum (ER) stress and mitochondrial dysfunction are hallmarks of many ophthalmic diseases; however, they have traditionally been examined as isolated pathological processes. Recent evidence indicates that these organelles are inextricably coupled through mitochondria-endoplasmic reticulum contact sites, also known as mitochondria-associated membranes (MAMs), which coordinate Ca2+ signaling, lipid transfer, mitochondrial dynamics, redox balance, and cell death decisions. Consequently, dysregulated ER-mitochondria communication has emerged as a key vulnerability that links the ce

Incubatory environment of the scalp impacts pre-emergent hair to affect post-emergent hair cuticle integrity.

OBJECTIVES: To determine whether the oxidative stress transmitted to newly grown hair from an unhealthy scalp has physical consequences to the cuticular condition and function. METHODS: A uniquely designed 24-week clinical study included 8 weeks of pretreatment with a cosmetic shampoo and 16 weeks of treatment with either a potentiated zinc pyrithione (ZPT) antidandruff shampoo or a placebo cosmetic shampoo. This clinical design allowed the growth and acquisition of hair samples under conditions of varying but known scalp health as a result of treating a dandruff/seborrheic dermatitis (D/SD)

Intraspecific variation in mycorrhiza-associated traits predicts plant responses to resource limitation.

Variation in arbuscular mycorrhizal fungal (AMF) root colonization and associated root traits among plant species plays a crucial role in determining plant community responses to environmental change. However, species mean trait values often obscure substantial variation among individuals within a species. Because trait-based ecology has focused predominantly on interspecific differences, the intraspecific responses of AMF-associated traits to resource limitations remain poorly understood. To address this gap, we examined intraspecific variation in AMF-associated traits among genetically diver

Identification of the R2R3-MYB gene family in wild jujube (Ziziphus jujuba var. spinosa) and analysis of its expression under drought stress.

BACKGROUND: R2R3-MYB gene family serves as a pivotal regulatory factor in plant growth, development, and responses to environmental stresses. To investigate its function in the drought stress response of wild jujube (Ziziphus jujuba Mill. var. spinosa), a typical eco-economic forest species, this study performed genome-wide identification and relevant analyses of R2R3-MYB genes. RESULTS: A total of 91 R2R3-MYB genes (designated as ZjMYB1 to ZjMYB91) were identified, which were unevenly distributed across 12 chromosomes. These genes mainly encode hydrophilic and unstable proteins, 97.8% of whi

Integrated WGCNA and TO-GCN analysis reveals pathways and genes underlying salt tolerance in Sesbania cannabina.

Salt stress seriously threatens the growth and development of plants. To investigate how Sesbania cannabina responds to salt stress, we constructed a temporal regulatory response network under salt stress by integrating the physiological and transcriptomic data from 'Zhongke Jing No.1' at five time points (0, 6, 12, 24, and 48 h, n = 3) after salt treatment. The results indicated that salt stress increased the relative electrical conductivity (REC) and malondialdehyde (MDA) content, induced the accumulation of osmotic regulators, and enhanced antioxidant enzyme activities. A total of 688,424,7

Mechanisms of Lipid Droplet-Mitochondria Interactions and Their Roles in Diseases.

Lipid droplets (LDs) coordinate cellular lipid utilization, energy homeostasis, redox balance, and autophagy through dynamic inter-organelle communication with mitochondria. This review summarized recent advances in the molecular mechanisms regulating LD-mitochondria interactions and discusses how their dysregulation contributes to disease progression. Accumulating evidence has indicated that the contacts between LD and mitochondria are dynamically regulated through specialized membrane contact sites and protein complexes, such as Perilipin (PLIN) family of proteins, Mitofusin2 (Mfn2), and Rab

Endoplasmic reticulum stress in cardiomyopathies: from the unfolded protein response to therapeutic opportunities.

The endoplasmic reticulum (ER), a central organelle responsible for maintaining protein homeostasis, calcium balance, and lipid metabolism, is essential for cardiovascular integrity. Functional disruption-referred to as endoplasmic reticulum stress (ERS)-has been recognized as a major pathogenic driver across diverse cardiovascular disorders. Under pathological conditions such as hypoxia, nutrient deprivation, or infection, sustained ERS activates the unfolded protein response (UPR). While initially adaptive, prolonged or excessive ERS initiates apoptotic cascades, severely impairing cardiomyo

Proteostasis Disturbances and Inflammation in Neurodegenerative Diseases.

Protein homeostasis (proteostasis) disturbances and inflammation are evident in normal aging and some age-related neurodegenerative diseases. While the proteostasis network maintains the integrity of intracellular and extracellular functional proteins, inflammation is a biological response to harmful stimuli. Cellular stress conditions can cause protein damage, thus exacerbating protein misfolding and leading to an eventual overload of the degradation system. The regulation of proteostasis network is particularly important in postmitotic neurons due to their limited regenerative capacity. Ther

Spatial and Single-Nucleus Transcriptomic Profiling Reveal Polarity and Intracellular Responses to Mitochondrial Dysfunction.

Organelle retrograde signaling studies have exclusively relied on bulk tissues, obscuring cell-type specificity, spatial organization, and heterogeneity within cell types. Here, we use single-nucleus and spatial RNA sequencing to define the cellular and spatial architecture of mitochondrial stress signaling in Arabidopsis thaliana. Inhibition of the mitochondrial electron transport chain by antimycin A and myxothiazol rapidly redistributed nuclei among transcriptional states within epidermal, leaf pavement, and mesophyll lineages, revealing that mitochondrial dysfunction reshapes cell identity

A mechanistic modeling framework to interpret ACTH stimulation tests across HPA axis adaptation states and glucocorticoid feedback dynamics.

The hypothalamic pituitary adrenal (HPA) axis is a key regulatory system coordinating endocrine responses to physiological and psychological stress. While the ACTH stimulation test remains a cornerstone of adrenal function assessment, its interpretation is complicated by the dynamic and adaptive nature of the HPA axis under chronic stress exposure. In particular, prolonged stress induces glandular remodeling, glucocorticoid receptor (GR) resistance and delayed feedback recovery, all of which may alter test outcomes without indicating primary adrenal failure. In this study, we present a mechani

Melatonin protects the mouse testis against heat-induced damage.

Spermatogenesis, an intricate process occurring in the testis, is responsible for ongoing production of spermatozoa and thus the cornerstone of lifelong male fertility. In the testis, spermatogenesis occurs optimally at a temperature 2-4°C lower than that of the core body. Increased scrotal temperature generates testicular heat stress and later causes testicular atrophy and spermatogenic arrest, resulting in a lower sperm yield and therefore impaired male fertility. Melatonin (N-acetyl-5-methoxytryptamine), a small neuro-hormone synthesized and secreted by the pineal gland and the testis, is w

Astrocyte Bioenergetic Remodeling as a Central Trait of Disrupted Glucocorticoid Signaling: Mechanisms and Implications for Stress Vulnerability.

Glucocorticoids (GCs) are central to the organism's adaptation to stress, coordinating systemic energy distribution and neuroendocrine signaling. While acute effects of GCs are adaptive, chronic GC exposure is increasingly recognized as an important factor contributing to the pathophysiology of neuropsychiatric disorders, such as post-traumatic stress disorder (PTSD) or major depressive disorder (MDD). A piling evidence points to astrocytes as a central integrator of brain response to stress hormones, including GCs. In this review, we discuss a biphasic regulation of astrocyte metabolism by GC

Mechanisms for enhancing ethanol tolerance of Saccharomyces cerevisiae through nano-selenium supplementation during Chinese rice wine brewing.

BACKGROUND: The toxic environment created by elevated ethanol levels presents significant challenges to Saccharomyces cerevisiae, leading to incomplete fermentation of the raw materials, poor flavor of the product, and even increased difficulty in post-processing of the product. Therefore, enhancing the ethanol tolerance and metabolic capacity of strains is critical for the brewing of Chinese rice wine. RESULTS: Results revealed that 1-3 mg L-1 nano-selenium significantly increased viable bacteria counts, reproduction rates and antioxidant enzyme activities, while reducing malondialdehyde lev

Nitric Oxide Dynamics in High-Altitude Medicinal Plants: Role in Stress Adaptation, Signaling, and Phytohormonal Interactions.

Nitric oxide (NO) is a pivotal signaling molecule that plays a crucial role in the high-altitude adaptation and stress tolerance of medicinal plants. Under hypoxic conditions prevalent in high-altitude environments, NO facilitates oxygen sensing, regulates mitochondrial function, and influences ethylene biosynthesis, thereby enhancing plant resilience. This review elucidates NO's modulation of oxidative stress responses through interactions with reactive oxygen species (ROS) to maintain redox homeostasis. Additionally, NO's interaction with key phytohormones, including abscisic acid, ethylene,
