Mechanisms of disease and immunity in cholera: a review.

The adenyl cyclase-activating enterotoxin of Vibrio cholerae was shown to contain two types of subunit: six smaller units (L) that are responsible for the binding to cell membrane receptors and a larger unit (H) that mediates the toxic action. The receptor was identified as the ganglioside GM1 (galactosyl-N-acetylgalactosaminyl [sialosyl] lactosyl ceramide), and the results suggested that penetration of the toxin molecule into the membrane follows the rapid binding to GM1. The relationship of these findings to the mechanism of protective immunity, which is mediated by antibodies to the enterot

Acupuncture for Poststroke Insomnia Regulating the Gut-Brain Axis: The Evidence Mapping of Clinical Studies.

The intestinal flora is an important physiological component of the gastrointestinal system and one of the sources of neurotransmitters. About 90% of neurotransmitters in the brain are produced in the human intestine. Poststroke insomnia is caused by damage to the internal and external structures of the brain, characterized by difficulties in falling asleep, shallow sleep, easy awakenings, and challenges in returning to sleep, all impacting patients' quality of life. Hypnotics have many adverse reactions in the treatment of insomnia, which can lead to drug resistance and relapse. The systemati

Paraneoplastic Cushing Syndrome in Gastrointestinal Neuroendocrine Tumour.

Ectopic production of adrenocorticotropic hormone (ACTH) by gastrointestinal neuroendocrine tumours (NETs) is relatively uncommon. We report a rare case of a liver metastatic G1 low-grade NET of the intestine that induced hypercortisolism after surgical resection. A 50-year-old man was admitted for an intestinal obstruction caused by a tumour of the intestine. Paraneoplastic Cushing syndrome was diagnosed more than a year later following the appearance of cushingoid symptoms, despite stable disease according to RECIST criteria but chromogranin A increase. The gut mucosal immune response to Toxoplasma gondii.

As an orally acquired pathogen, the immune response to Toxoplasma gondii unfolds in the small intestinal mucosa. There, an array of regulatory and effector immune cells are elicited to combat the parasite through secretion of inflammatory mediators, normally resulting in host protection and pathogen control. Recent studies largely in mice have found that a productive immune response requires the combined recognition of parasite- and commensal-derived antigens by mucosal leucocytes. However, despite the fine-tuned regulatory mechanisms in place to prevent immunopathology, dysregulated responses

Substance P stimulates human airway submucosal gland secretion mainly via a CFTR-dependent process.

Chronic bacterial airway infections are the major cause of mortality in cystic fibrosis (CF). Normal airway defenses include reflex stimulation of submucosal gland mucus secretion by sensory neurons that release substance P (SubP). CFTR is an anion channel involved in fluid secretion and mutated in CF; the role of CFTR in secretions stimulated by SubP is unknown. We used optical methods to measure SubP-mediated secretion from human submucosal glands in lung transplant tissue. Glands from control but not CF subjects responded to mucosal chili oil. Similarly, serosal SubP stimulated secretion in

Cholinergic stimulation of neuropeptide Y secretion from the isolated perfused rat stomach.

Neuropeptide Y (NPY) is present in both extrinsic sympathetic adrenergic nerve terminals and intrinsic nerves of the gastrointestinal (GI) tract. Based on this localization a number of functions have been attributed to GI NPY including regulation of blood flow, intestinal fluid and electrolyte transport, and motility. There is nothing currently known, however, about the regulation of its secretion from GI nerves. The effect of cholinergic agonists and antagonists on secretion of NPY immunoreactivity (NPY-IR) from the isolated perfused rat stomach was investigated in the present study. Perfusat

Profile of neurohumoral agents on mesenteric and intestinal blood flow in health and disease.

The mesenteric and intestinal blood flow is organized and regulated to support normal intestinal function, and the regulation of blood flow is, in part, determined by intestinal function itself. In the process of the development and adaptation of the intestinal mucosa for the support of the digestive processes and host defense mechanisms, and the muscle layers for propulsion of foodstuffs, a specialized microvascular architecture has evolved in each tissue layer. Compromised mesenteric and intestinal blood flow, which can be common in the elderly, may lead to devastating clinical consequences.

Effect of Lansoprazole on the Control of the Intragastric pH in a Patient with Short Bowel Syndrome.

A 40-year-old man suffered from short bowel syndrome. Since a large amount of intestinal output and watery diarrhea hampered his quality of life, we tried to control the intestinal output by reducing the secretion of gastric acid with lansoprazole. Because the small intestine was only 10 cm in length and effective absorption of oral lansoprazole was doubtful, we monitored his intragastric pH for 24 hours and confirmed that the holding time above pH 3.0 was 14.5 hours (60.4%). He spent his home life eating porridge during the day, and receiving total parenteral nutrition of 1,100 mL/day at nigh

In the absence of nutrients, pancreatic-biliary secretions in the jejunum do not exert feedback control of human pancreatic or gastric function.

Feedback inhibition of basal pancreatic enzyme secretion by luminal pancreatic enzymes appears to be an important regulator of pancreatic secretion in some laboratory animals. To determine whether pancreatic enzymes in the jejunum influence pancreatic or gastric functions in healthy man, we intubated six subjects with a gastric sump tube and a four-lumen duodenal tube which provided (1) a duodenal perfusion site, (2) a duodenal aspiration site, (3) an inflatable balloon immediately distal to the aspiration site, and (4) a jejunal perfusion site immediately beyond the balloon. In this way, the

2-Arachidonoyl glycerol potently induces cholecystokinin secretion in murine enteroendocrine STC-1 cells via cannabinoid receptor CB1.

Cholecystokinin (CCK) is a peptide hormone secreted from enteroendocrine cells and regulates the exocrine pancreas, gastric motility, and appetite. Dietary triacylglycerols are hydrolyzed to fatty acids (FA) and 2-monoacylglycerols (2-MAG) in the small intestine. Although it is well known that FA stimulate CCK secretion, whether 2-MAG have the CCK-releasing activity remains unclear. We examined the CCK-releasing activity of four commercially available 2-MAG in a murine CCK-producing cell line, STC-1, and the molecular mechanism underlying 2-MAG-induced CCK secretion. CCK released from the cell

Effects of Caulis Spatholobi Polysaccharide on Immunity, Intestinal Mucosal Barrier Function, and Intestinal Microbiota in Cyclophosphamide-Induced Immunosuppressive Chickens.

The protective effects of Caulis Spatholobi polysaccharide (CSP) on immune function, intestinal mucosal barrier, and intestinal microflora in cyclophosphamide (CY)-induced immunosuppressed chickens have been rarely reported. This study was designed to investigate the cecal microbiota in chickens and to elucidate the immune mechanism involved in the CSP effect on CY induced-immunosuppressed chickens. A total of 288 cocks were equally divided into six groups and used to evaluate the immune effect of CSP. Results showed that the CSP increased the body weight and immune organ index of immunosuppre

Effects of a probiotic on the morphology and mucin composition of pig intestine.

Although the use of probiotics in human and animal medicine is growing, their mode of action remains poorly understood. This study examined the effects of a multi-strain probiotic (SLAB51™) on the morphology and carbohydrate composition of mucins secreted by goblet cells of intestinal crypts in growing-finishing pigs. Sections of duodenum, caecum and colon from pigs fed for 12 weeks with an orally administered control basal diet (No-Pro) or one with a probiotic blend (Pro) were processed for microscopic analysis and stained with (1) haematoxylin-eosin for structural and morphometrical investig

Reduction of bile salt secretion in children with coeliac disease in activity and in remission.

To assess the role of intestinal absorption in bile secretion, the effects of milk perfusion into the duodenum on bile salt secretion were studied in children with active coeliac disease. Bile secretion was evaluated by measuring the bile salt concentration and output in the duodenum in response to a perfusion with a milk containing a casein hydrolysate as the protein source (4 ml/min/m2) followed by i.v. injection of 2 U/kg cholecystokinine (CCK). Bile salt output was estimated taking into account the dilution of the BSP perfused into the first part of the duodenum and collected 15 cm further

Epicatechin alleviates ulcerative colitis by restoring intestinal mucosa via regulating the NCR+ILC3 cell counts.

BACKGROUND: Promoting damaged intestinal mucosal repair and restoring epithelial barrier function are critical strategies for treating ulcerative colitis (UC). Interleukin-22 (IL-22), a key cytokine for intestinal epithelial homeostasis, is mainly secreted by group 3 innate lymphoid cells (ILC3s). ILC3s exhibit substantial plasticity: NCR⁻ILC3 preferentially secrete IL-17A, whereas NCR⁺ILC3 primarily produce IL-22; under inflammatory conditions, NCR⁺ILC3 and ILC1 can undergo reciprocal conversion. Hypoxia-inducible factor-1α (HIF-1α) signaling is involved in regulating ILC3 subset balance and

Vasoactive intestinal peptide interacts with alpha-adrenergic-, cholinergic-, and substance-P-mediated responses in rat parotid and submandibular glands.

Secretory dose-response curves were obtained with both acetylcholine and phenylephrine treatment in rat parotid and submandibular glands. Vasoactive intestinal peptide (VIP), which produced relatively low volumes of protein-rich saliva in rat salivary glands, also enhanced acetylcholine-, phenylephrine-, and substance-P-mediated fluid and protein secretion when administered in combination with these agents. The specific mechanisms involved in the synergistic actions of VIP with substances such as acetylcholine, phenylephrine, and substance P, which are primarily linked to the production of flu

Autonomic nervous control of venous pressure and secretion in submandibular gland of anesthetized dogs.

In dogs anesthetized with pentobarbital sodium, hilar venous pressure (Phv) and secretion were measured from the submandibular gland receiving spontaneous blood flow or vascular perfusion at the normal resting flow rate. Parasympathetic nerve stimulation and ACh-induced secretion increased Phv and its pulse pressure; Phv also showed an obvious arterial (or perfusion pressure)-like waveform. Vasoactive intestinal polypeptide (VIP) exerted similar effects on Phv but produced negligible secretion. Sympathetic nerve stimulation, phenylephrine, and clonidine did not induce secretion and had no sign

Effects of vasoactive intestinal polypeptide on acetylcholine stimulation of rat submandibular gland.

Studies were carried out on the role of vasoactive intestinal polypeptide (VIP) in the regulation of secretion and blood flow in the rat salivary gland. The first experiments to investigate the spontaneous secretory pattern revealed a clear diurnal fluctuation with a significant increase at night, so that the subsequent experiments were performed during the daytime where the secretion was consistently low. Intravenous administration of VIP at a dose smaller than 40 pmole caused a dose-dependent vasodilatory response, but at a high dose such a local effect was hampered by a decrease in systemic

An integrated transcriptomics and proteomics analysis of the secretome of the helminth pathogen Fasciola hepatica: proteins associated with invasion and infection of the mammalian host.

To infect their mammalian hosts, Fasciola hepatica larvae must penetrate and traverse the intestinal wall of the duodenum, move through the peritoneum, and penetrate the liver. After migrating through and feeding on the liver, causing extensive tissue damage, the parasites move to their final niche in the bile ducts where they mature and produce eggs. Here we integrated a transcriptomics and proteomics approach to profile Fasciola secretory proteins that are involved in host-pathogen interactions and to correlate changes in their expression with the migration of the parasite. Prediction of F.

The feasibility of a crossover, randomized controlled trial design to assess the effect of probiotics and prebiotics on health of elite Swiss para-athletes: a study protocol.

BACKGROUND: Spinal cord injury (SCI) may cause an autonomic imbalance in the gastrointestinal tract, leading to deficits in colonic motility, mucosal secretions, vascular tone, and an increase of intestinal barrier permeability. Autonomic denervation and factors such as age, physical activity, antibiotic use and stress may cause intestinal bacterial translocation, decreased microbiota diversity, known as gut dysbiosis and thus increase susceptibility to experiencing gastrointestinal discomfort. Probiotic treatment in individuals with SCI may normalize the gut microbiota and improve overall hea
