Identification of pre-diagnostic lipid sets associated with liver cancer risk using untargeted lipidomics and chemical set analysis: A nested case-control study within the ATBC cohort.

In pre-disposed individuals, a reprogramming of the hepatic lipid metabolism may support liver cancer initiation. We conducted a high-resolution mass spectrometry based untargeted lipidomics analysis of pre-diagnostic serum samples from a nested case-control study (219 liver cancer cases and 219 controls) within the Alpha-Tocopherol, Beta-Carotene Cancer Prevention (ATBC) Study. Out of 462 annotated lipids, 158 (34.2%) were associated with liver cancer risk in a conditional logistic regression analysis at a false discovery rate (FDR) <0.05. A chemical set enrichment analysis (ChemRICH) and co-…

Integrative computational analysis of public fecal lipidomics and transcriptomics datasets suggests a candidate association between the COX-2 pathway and CE(20:4) in colorectal adenoma-carcinoma progression.

BACKGROUND: The fecal lipidomic changes underlying the colorectal adenoma-carcinoma sequence remain incompletely characterized, particularly regarding the continuous metabolic trajectory and heterogeneity of precancerous adenomas. We aimed to use publicly available multi-omics data and an integrative computational framework to identify candidate lipidomic features associated with a CRC-like metabolic state in adenomas. METHODS: We developed an extreme-phenotype machine learning strategy using fecal lipidomics from healthy controls and colorectal cancer (CRC) patients (Study ID: ST003798) to b…

Lipidomic and transcriptomic analyses reveal that the gene ZmLACS9 enhances salt stress tolerance by regulating chloroplast lipid metabolism and maintaining photosynthetic performance in maize.

Salt stress is a major environmental factor limiting crop growth and productivity. We found a long-chain acyl-CoA synthetase (LACS) gene ZmLACS9 that was associated with salt tolerance traits in maize (Zea mays). Mutation of ZmLACS9 impaired plant growth, while overexpression of ZmLACS9 improved plant growth vigour under salt stress conditions. Metabolome and transcriptome analyses indicated that ZmLACS9 may regulate the expression of lipid trafficking-related genes, thereby influencing the concentrations of galactolipids and phospholipid in the photosynthetic membrane under salt stress. Genetically proxied circulating PD-1/PD-L1 levels and broadly defined myocarditis: A bidirectional Mendelian randomization study with exploratory lipidomic analyses.

Myocarditis is an inflammatory myocardial disease with potentially severe outcomes. Programmed cell death protein 1 (PD-1) and programmed death-ligand 1 (PD-L1) regulate immune tolerance, but the association between lifelong genetically proxied circulating PD-1/PD-L1 levels and broadly defined myocarditis remains uncertain. We investigated these associations and explored related plasma lipid species. We conducted bidirectional 2-sample Mendelian randomization using proteomic genome-wide association data from the UK Biobank Pharma Proteomics Project and INTERVAL. FinnGen Release 10 was the prim

Integrated lipidomic and transcriptomic analyses reveal stage-specific hepatic lipid remodeling during GCRV-II infection in grass carp (Ctenopharyngodon idella).

Grass carp reovirus genotype II (GCRV-II) is still a major threat to grass carp aquaculture and causes huge economic losses. Viral replication relies on host metabolic resources and metabolic reprogramming is key to virus-host interactions. However, the dynamic hepatic lipid remodeling associated with GCRV-II infection remains poorly understood. Here, the effect of GCRV-II infection on damage condition of liver from grass carp was evaluated and integrated lipidomic and transcriptomic was analyzed. Obviously, GCRV-II infection caused evident hepatic injury, significantly increased viral load an

Lipidome profile of cystic fibrosis-related diabetes, type 1 and type 2 diabetes mellitus: potential links to inflammation and glucose and lipid metabolism.

Cystic fibrosis (CF) is a genetic disease that primarily affects the pancreas and lungs. CF dyslipidemia is characterized by decreased circulating lipids and increased ectopic lipid deposition in liver, pancreas, and lungs. Pancreatic exocrine insufficiency precedes the onset of CF-related diabetes (CFRD). We hypothesized that different mechanisms contribute to CFRD development and progression, including features of type 1 and type 2 diabetes mellitus (T1DM and T2DM). Thus, we compared their plasma inflammatory, metabolic/hormonal, and lipidomic profiles, using Luminex assays and untargeted ma

Benzoyl chloride derivatization improves selectivity and sensitivity of lipidomic quantitation in human serum of pancreatic cancer patients using RP-UHPLC/MS/MS.

Chemical derivatization is a powerful strategy for enhancing the chromatographic behavior and mass spectrometric sensitivity of lipids, which play an essential role in cellular processes and show high potential in cancer biomarker research. In this study, we describe a targeted and validated method that combines benzoyl chloride derivatization with reversed-phase ultrahigh-performance liquid chromatography tandem mass spectrometry (RP-UHPLC/MS/MS) for the quantitative analysis of the human serum lipidome. In total, 450 lipid species from 19 lipid subclasses were identified based on a combinati

Correlation of plasma lipidomic profiles with cardiometabolic disease in transfusion-dependent thalassemia patients with six-month N-acetylcysteine intervention: A prospective cohort study.

OBJECTIVES: Oxidative stress, driven by iron imbalance from recurrent blood transfusions, is a major contributor to cardiometabolic complications in transfusion-dependent thalassemia (TDT). N-acetylcysteine (NAC), a glutathione precursor, is a well-known antioxidant with cardioprotective effects achieved by mitigating the impact of oxidative stress on cell metabolism. Current study aimed to evaluate the effect of a six-month NAC intervention by focusing on previously reported changes in the plasma lipidome in TDT patients. Design & Methods A randomized cohort of 62 Thai TDT patients was divide

Evaluating the Protective Effects of Silymarin Along with a Cholesterol-Supplemented Diet Against Demyelination in an EAE Mouse Model of Multiple Sclerosis: Lipidomic and Inflammatory Aspects.

Demyelination in multiple sclerosis (MS) is associated with chronic inflammation and dysregulation of cholesterol metabolism. In this study, we investigated the protective effects of silymarin, an herbal flavonoid, against inflammation, impairment of brain cholesterol metabolism, and demyelination in the experimental autoimmune encephalomyelitis (EAE) model of MS. Given the well-documented anti-hypercholesterolemic properties of silymarin, we sought to assess its protective effects in combination with a cholesterol-supplemented diet. Additionally, the effects of the cholesterol-supplemented di

Lipid Profiles from Dried Blood Spots Reveal Lipidomic Signatures of Newborns Undergoing Mild Therapeutic Hypothermia after Hypoxic-Ischemic Encephalopathy.

Hypoxic-ischemic encephalopathy (HIE) is associated with perinatal brain injury, which may lead to disability or death. As the brain is a lipid-rich organ, various lipid species can be significantly impacted by HIE and these correlate with specific changes to the lipidomic profile in the circulation. Objective: To investigate the peripheral blood lipidomic signature in dried blood spots (DBS) from newborns with HIE. Using univariate analysis, multivariate analysis and sPLS-DA modelling, we show that newborns with moderate-severe HIE (n = 46) who underwent therapeutic hypothermia (TH) displayed

Application of a Novel Mass Spectral Data Acquisition Approach to Lipidomic Analysis of Liver Extracts from Sitaxentan-Treated Liver-Humanized PXB Mice.

The application of a data-independent acquisition (DIA) method ("SONAR") that employs a rapidly scanning quadrupole is described for the lipidomic analysis of complex biological extracts. Using this approach, the MS acquisition window can be varied between 1 and 25 Da, enabling the isolation of ions prior to their entering the collision cell. By rapidly scanning the resolving quadrupole window over a specified mass range, co-eluting precursor ions are transmitted sequentially into the collision cell, where collision energies are cycled between low and elevated levels to induce fragmentation. Metabolome and Lipidome Profiles of Populus × canescens Twig Tissues During Annual Growth Show Phospholipid-Linked Storage and Mobilization of C, N, and S.

The temperate climax tree species Fagus sylvatica and the floodplain tree species Populus × canescens possess contrasting phosphorus (P) nutrition strategies. While F. sylvatica has been documented to display P storage and mobilization (Netzer et al., 2017), this was not observed for Populus × canescens (Netzer et al., 2018b). Nevertheless, changes in the abundance of organic bound P in gray poplar trees indicated adaptation of the P nutrition to different needs during annual growth. The present study aimed at characterizing seasonal changes in metabolite and lipid abundances in gray poplar an

Mass Spectrometry-Based Metabolomic and Lipidomic Analyses of the Effects of Dietary Platycodon grandiflorum on Liver and Serum of Obese Mice under a High-Fat Diet.

We aimed to identify metabolites involved in the anti-obesity effects of Platycodon grandiflorum (PG) in high-fat diet (HFD)-fed mice using mass spectrometry (MS)-based metabolomic techniques. C57BL/6J mice were divided into four groups: normal diet (ND)-fed mice, HFD-fed mice, HFD with 1% PG extract-fed mice (HPGL), and HFD with 5% PG extract-fed mice (HPGH). After 8 weeks, the HFD group gained more weight than the ND group, while dietary 5% PG extract attenuated this change. The partial least squares discriminant analysis (PLS-DA) score plots showed a clear distinction between experimental g
