High-sugar driven microbial dynamics in spontaneous fermentations of Lebanese cooked wines.

Traditional Lebanese cooked wines are produced by boiling grapes to concentrate the must, creating an extreme environment marked by high sugar levels where fermentation occurs spontaneously. This study investigated the microbial dynamics and chemical changes occurring in seven fermentations of cooked must from four wineries. Using high-throughput amplicon sequencing of bacterial 16S and fungal ITS2 regions, combined with culture-dependent for yeast isolation and HPLC analyses, we monitored microbial and chemical changes from fresh juice and along the fermentation. Sugar concentrations increase

Untargeted LC-MS-based metabolic fingerprinting of Escherichia coli-associated urinary tract infections and urosepsis: Insights into the urine, serum, and bacterial interactomes.

Urinary tract infections (UTIs) and urosepsis necessitate a deeper understanding of host-pathogen interactions at the metabolic level. We use LC-MS and GC-MS techniques to characterize metabolic pathway alterations in patients and Escherichia coli isolates during UTI and urosepsis. Our findings reveal substantial metabolic adaptations in the human host, including increased porphyrin metabolism, suggesting oxidative stress response or tissue damage. Activation of the pentose phosphate pathway (PPP) and tricarboxylic acid cycle (TCA) highlights the host's heightened immune and energy demands dur

Integrated metabolomic and gut microbiota analyses reveal TCA cycle dysfunction in broiler skeletal disorders and the protective effect of L‑malic acid.

BACKGROUND: Skeletal disorders pose a significant challenge to intensive broiler production. However, the metabolic mechanisms underlying the development of skeletal abnormalities in broilers remain unclear. This study aims to reveal the underlying mechanisms through multi-omics analyses and evaluate the ameliorative effects of exogenous L-malic acid supplementation. RESULTS: The abnormal group exhibited reduced serum concentrations of calcium and phosphorus, as well as decreased bone calcium and phosphorus content. These changes were accompanied by deterioration in bone geometric properties

Development of new liquid chromatography-tandem mass spectrometry methods for the quantification of seven metabolites in three related breast cancer cell lines.

Metabolites of the tricarboxylic acid (TCA) cycle play crucial roles in cancer biology, and their accurate quantification is essential for understanding energy metabolism, signaling dynamics, and identifying metabolic vulnerabilities in cancer cells. However, traditional liquid chromatograph-tandem mass spectrometry (LC-MS/MS) methods for these polar metabolites often encounter challenges, such as limited retention on reversed-phase columns and ion suppression. This study developed and validated two LC-MS/MS methods for the accurate quantification of seven key TCA cycle metabolites in MDA-MB-2

Sulfur dioxide delays quality deterioration by maintaining energy metabolism and mitochondrial structure in postharvest table grapes.

BACKGROUND: Table grapes are very susceptible to rapid softening, browning, and decay after harvest, which significantly reduces their commercial value. Sulfur dioxide (SO2) treatment is used widely to mitigate these postharvest losses; however, the biological mechanisms underlying its preservative effects remain poorly understood. This study aimed to elucidate how SO2 preserves postharvest fruit quality by modulating energy metabolism in 'Munage' grape berries (Vitis vinifera L. 'Munage'). RESULTS: Sulfur dioxide fumigation (500 μL L-1) effectively maintained the visual quality of grapes dur

Comparative Effects of High-Molecular-Weight Chitosan and Chitosan Oligosaccharide Lactate on Metabolic Adjustment and Salinity Tolerance in Cucumis sativus L.

Salinity is one of the most harmful abiotic factors limiting crop productivity. Chitosan is a natural polysaccharide biostimulant able to alleviate salt stress, but its efficiency depends on its physicochemical properties, which remain insufficiently understood. The present study aimed to determine whether two chemically distinct forms of chitosan, i.e., high-molecular-weight chitosan (ChT) and chitosan oligosaccharide lactate (ChL), differentially mitigate the adverse effects of NaCl-induced salinity in cucumber (Cucumis sativus L.). Plants were grown hydroponically under control conditions o

Cholinergic Ligand-dependent Modulation of Oxidative Phosphorylation Coupling in Digitonin-permeabilized BE(2)-C Neuroblastoma Cells.

The objective of this study was to assess oxidative phosphorylation (OXPHOS) function in cultured cells using defined substrate-inhibitor combinations while retaining cellular structure and cytosolic context lost in isolated mitochondrial preparations. Because intact cells are poorly permeable to several Krebs cycle intermediates, direct assessment of substrate-supported respiration through specific electron transport chain (ETC) entry points is limited. To overcome this, we applied digitonin-mediated selective plasma membrane permeabilization and performed extracellular flux analyzer-based co

Inhibitory effect of Acetylshikonin on drug-resistant Candida albicans.

BACKGROUND: Candida albicans (C. albicans) is among the most common opportunistic fungal pathogens and causes superficial infections and life-threatening invasive candidiasis in immunocompromised individuals. In recent years, C. albicans has become resistant to a wide range of clinical drugs, and the identification of effective antifungal drugs is an extremely urgent medical need. Acetylshikonin (ASK) is a naphthoquinone compound extracted from Lithospermum erythrorhizon that exhibits potent antibacterial activity. However, its efficacy against C. albicans and antimicrobial mechanisms, especia

A comparative analysis of monascin and ankaflavin biosynthesis in Monascus purpureus and its albino variant.

Red mold rice (RMR), a functional food ingredient, is produced via Monascus spp. fermentation, a process that yields a diverse range of bioactive compounds, most notably the Monascus yellow pigments monascin and ankaflavin. This study provides the first report of spontaneous albinism in Monascus species. Although the specific mechanisms and conditions triggering the transition to albino strains remain to be definitively elucidated, our analyses reveal that the albino strain of Monascus purpureus exhibits a faster growth rate and significantly lower yellow pigment production compared to the wil

(p)ppGpp mediates persister formation in Escherichia coli during glucose to fatty acid shift.

Bacterial persistence contributes to antibiotic failure and recurrent infectious disease, yet the metabolic cues that promote persister formation remain poorly understood. Here we investigated Escherichia coli persistence after nutrient downshifts from glucose to various carbons. Compared to shifts to gluconeogenic carbons (pyruvate, malate, succinate, and fumarate), the glucose-to-fatty acid shift induced exceptionally high persister levels, with cells tolerating ampicillin (56%), carbenicillin (22%), and gentamicin (1%) after 24-h treatment. With an RNA-based biosensor and HPLC quantificatio

Inhibition of Cronobacter sakazakii by Apigenin and the Antibacterial Mechanism.

Cronobacter sakazakii is a significant foodborne pathogen commonly associated with dairy products, causing severe infections with high mortality rates, particularly in neonates and immunocompromised adults. In this study, we demonstrated that apigenin exhibits potent antibacterial activity against C. sakazakii, with a minimum inhibitory concentration (MIC) of 160 mg/L. Propidium iodide staining indicated that apigenin at sub-MIC and MIC concentrations had negligible effects on bacterial membrane integrity; only a 4.9% increase in membrane permeability was observed at 2 MIC, suggesting that mem

Impact of leukoreduction on the metabolome of ovine packed red blood cells during refrigerated storage.

BACKGROUND: Blood transfusion is a life-saving intervention for many species of veterinary interest, including sheep. Despite extensive research on the impact of refrigerated storage of packed red blood cells (pRBC) in humans, research on the quality of stored ovine blood is limited and storage guidelines are mostly informed by studies in humans. Human pRBC are currently stored without residual white blood cells, following selective removal of the leukocytes by filtration (leukoreduction). This process delays the onset and mitigates the progression of the storage lesion, a series of molecular

A tumor-derived lactate/ENO1 lactylation feedback loop facilitates osimertinib resistance of lung adenocarcinoma.

Enhanced glycolysis and lactate accumulation are shared features of human cancers. Lactylation is a lactate-derived posttranslational modification. So far, the impact of lactylation on resistance to osimertinib (a third-generation epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI)) in patients with lung adenocarcinoma (LUAD) remains indistinct. Here, we performed 4D label-free proteomics analysis of LUAD tissues from advanced-stage EGFR-mutant patients treated with surgery with or without neoadjuvant osimertinib to reveal a global lactylation profile and explore the role and

Elevation of hepatic de novo lipogenesis in mice with overnutrition is dependent on multiple substrates.

Increased de novo lipogenesis (DNL) contributes to hyperlipidemia, MASLD, and ASCVD in insulin-resistant subjects. However, multiple pathways support lipogenesis and few have sought to quantify the contributions of the discrete metabolic pathways that contribute to lipogenesis. In this study, antisense oligonucleotides (ASOs) targeting glucokinase (Gck), lactate dehydrogenase A (Ldha), and glutamic-pyruvic transaminase 2 (Gpt2) were utilized to restrict substrate flux from lipogenic precursors in C57BL6/J mice, comparing controls (CO) and chronic overnutrition (ON). In CO mice, ASO treatments

Deciphering the impact of NOS-derived NO on nitrogen metabolism and carbon flux in the heterocytous cyanobacterium Aphanizomenon flos-aquae 2012/KM1/D3.

Nitric oxide synthases (NOSs) are heme-based monooxygenases that catalyze the NADPH-dependent oxidation of L-arginine to produce NO and L-citrulline. Over the past five years, the identification and characterization of NOS homologs in cyanobacteria have significantly advanced our understanding of these enzymes. However, the precise mechanisms through which NOS-derived NO influences nitrogen metabolism remain incompletely elucidated. Therefore, the present study aims to investigates the impact of NOS-derived NO on nitrogen metabolism, heterocyte development, and carbon utilization dynamics in A

Variations in Human Milk Metabolites After Gestational Diabetes: Associations with Infant Growth.

BACKGROUND/OBJECTIVES: Gestational diabetes mellitus (GDM) is a condition characterized by hyperglycemia and is associated with increased risk of obesity and diabetes in exposed children. Differences in human milk composition between women with (GDM+) and without GDM (GDM-) suggest that GDM could impact milk production and composition, potentially influencing infant growth. However, this association remains poorly understood. The objective was to study the association between GDM and human milk composition and its influence on infant growth, focusing on metabolites and bioactive molecules invo

Dapagliflozin modulates plasma lipidomic profile and urinary metabolite excretion in type 2 diabetes.

BACKGROUND: Dapagliflozin (DAPA) has shown major nephroprotective effects, improving kidney metabolism and oxigenation. Lipidomics and metabolomics are powerful tools for understanding such effects, providing a comprehensive look at how SGLT2 inhibitors might change the metabolic landscape beyond their primary glucose-lowering action. We investigated changes in plasma metabolomic/lipidomic profile and urinary excretion of metabolites that could occur independent of increased diuresis. METHODS: A two-armed, parallel-design, randomized clinical trial was conducted in subjects with type 2 diabet

Monocarboxylate Transporter 4 Inhibition Reduces Synovial Hyperproliferation and Metabolic Reprogramming Under Hypoxia in Rheumatoid Arthritis.

OBJECTIVES: In hypoxic conditions, the acidic environment characteristic of rheumatoid arthritis (RA) induces metabolic changes in fibroblast-like synoviocytes (FLS), markedly promoting the synovial proliferation. Monocarboxylate transporter 4 (MCT4) plays a crucial role in cellular pH regulation and synovial fibroblast activation by regulating lactate export. This study investigates the impact of MCT4 inhibition on RA and elucidates its underlying mechanisms. METHOD: RA-FLS cells were treated with siMCT4 and VB124 (a selective MCT4 inhibitor), significantly affecting glucose and glutamine up

Fumarate hydratase ameliorates pressure overload induced cardiac remodeling by controlling Elovl7-mediated biosynthesis of unsaturated fatty acids.

Pathological cardiac hypertrophy as a major contributor to heart failure is characterized by complicated mechanisms. Fumarate hydratase (FH) is a crucial enzyme in the tricarboxylic acid cycle. FH mutations and dysfunction have been implicated in various pathological processes including hereditary leiomyomatosis and renal cell cancer, neurodegenerative diseases, metabolic syndrome and cardiovascular diseases. In this study we investigated the role of FH in cardiac hypertrophy. Cardiac hypertrophy was induced in mice by transverse aortic constriction (TAC) surgery as well as in neonatal rat car
