Maternal exposure to di(2-ethylhexyl) phthalate (DEHP) causes multigenerational adverse effects on the uterus of F1 and F2 offspring rats.

Phthalates are one of the ubiquitous chemicals found in day-to-day products like food packaging, children's toys, and other consumer commodities. There is rising concern that repeated exposure to phthalates during pregnancy and lactation could have long-term effects on maternal and fetal health. We hypothesize that exposure to DEHP during the developmental windows might affect the expression of molecules that regulate uterine function and that this effect would be passed on to further generations. Rat dams were treated with olive oil (vehicle) or DEHP (100 mg/kg b.wt./day) orally from gestatio

Risk of adverse maternal outcomes associated with prenatal exposure to moderate-severe depression compared with mild depression: A fellow-up study.

BACKGROUND: Pregnancy is a time of increased vulnerability for the development of anxiety and depression. The purpose of this study was to compare the risk of developing adverse maternal and perinatal outcomes between pregnant women with moderate-severe depression and those who had mild depression. METHODS: Our study was performed in a prospective cohort of 360 depressed pregnant women, recruited and followed up to delivery at the Adventist Hospital in Bekwai Municipality, Ghana. The research began in February 2020 and the follow-up was completed in August 2020. The pregnant women who had dep

In utero exposure to endogenous maternal polyclonal anti-Caspr2 antibody leads to behavioral abnormalities resembling autism spectrum disorder in male mice.

The concept that exposure in utero to maternal anti-brain antibodies contributes to the development of autism spectrum disorders (ASD) has been entertained for over a decade. We determined that antibodies targeting Caspr2 are present at high frequency in mothers with brain-reactive serology and a child with ASD, and further demonstrated that exposure in utero to a monoclonal anti-Caspr2 antibody, derived from a mother of an ASD child, led to an-ASD like phenotype in male offspring. Now we propose a new model to study the effects of in utero exposure to anti-Caspr2 antibody. Dams immunized with

Parental mental health before and during pregnancy and offspring birth outcomes: A 20-year preconception cohort of maternal and paternal exposure.

BACKGROUND: Preterm birth (PTB) and small for gestational age (SGA) are increasingly prevalent, with major consequences for health and development into later life. There is emerging evidence that some risk processes begin before pregnancy. We report on associations between maternal and paternal common mental disorders (CMD) before and during pregnancy and offspring PTB and SGA. METHODS: 398 women with 609 infants and 267 men with 421 infants were assessed repeatedly for CMD symptoms before pregnancy between age 14 and 29 and during pregnancy. Associations between preconception and antenatal C

Prenatal exposure to chlorinated polyfluoroalkyl ether sulfonic acids and perfluoroalkyl acids: Potential role of maternal determinants and associations with birth outcomes.

Transplacental exposure to per/polyfluoroalkyl substances (PFASs) may impact fetal growth, but published evidence are still sparse and not in agreement. Moreover, little is known on the occurrence of emerging chlorinated polyfluorinated ether sulfonates (Cl-PFESAs, 6:2 and 8:2) in maternal-neonatal population. This study investigated eleven PFASs by analyzing 98 cord samples from Hangzhou, China. All target compounds can be transported across placenta, with highest median concentrations of 4.07, 1.05 and 0.731 ng/mL for PFOS, PFOA, and 6:2 Cl-PFESA. Older ages and higher pre-pregnancy BMI were

Prenatal Risk Factors and Perinatal and Postnatal Outcomes Associated With Maternal Opioid Exposure in an Urban, Low-Income, Multiethnic US Population.

IMPORTANCE: The opioid epidemic increasingly affects pregnant women and developing fetuses, resulting in high rates of neonatal abstinence syndrome. However, longitudinal studies that prospectively observe newborns with neonatal abstinence syndrome or with maternal opioid use and examine their long-term physical and neurodevelopmental outcomes are lacking. OBJECTIVE: To examine prenatal risk factors associated with maternal opioid use during pregnancy and the short-term and long-term health consequences on their children. DESIGN, SETTING, AND PARTICIPANTS: This cohort study analyzed data fro

Associations between prenatal traffic-related air pollution exposure and birth weight: Modification by sex and maternal pre-pregnancy body mass index.

BACKGROUND: Prenatal traffic-related air pollution exposure is linked to adverse birth outcomes. However, modifying effects of maternal body mass index (BMI) and infant sex remain virtually unexplored. OBJECTIVES: We examined whether associations between prenatal air pollution and birth weight differed by sex and maternal BMI in 670 urban ethnically mixed mother-child pairs. METHODS: Black carbon (BC) levels were estimated using a validated spatio-temporal land-use regression (LUR) model; fine particulate matter (PM2.5) was estimated using a hybrid LUR model incorporating satellite-derived A

Elevation of 11β-hydroxysteroid dehydrogenase type 2 activity in Holocaust survivor offspring: evidence for an intergenerational effect of maternal trauma exposure.

BACKGROUND: Adult offspring of Holocaust survivors comprise an informative cohort in which to study intergenerational transmission of the effects of trauma exposure. Lower cortisol and enhanced glucocorticoid sensitivity have been previously demonstrated in Holocaust survivors with PTSD, and in offspring of Holocaust survivors in association with maternal PTSD. In other work, reduction in the activity of the enzyme 11β-hydroxysteroid dehydrogenase type 2 (11β-HSD-2), which inactivates cortisol, was identified in Holocaust survivors in comparison to age-matched, unexposed Jewish controls. LPS exposure increases maternal corticosterone levels, causes placental injury and increases IL-1Β levels in adult rat offspring: relevance to autism.

Maternal immune activation can induce neuropsychiatric disorders, such as autism and schizophrenia. Previous investigations by our group have shown that prenatal treatment of rats on gestation day 9.5 with lipopolysaccharide (LPS; 100 μg/kg, intraperitoneally), which mimics infections by gram-negative bacteria, induced autism-like behavior in male rats, including impaired communication and socialization and induced repetitive/restricted behavior. However, the behavior of female rats was unchanged. Little is known about how LPS-induced changes in the pregnant dam subsequently affect the develop

Caffeine-induced fetal rat over-exposure to maternal glucocorticoid and histone methylation of liver IGF-1 might cause skeletal growth retardation.

Several epidemiological investigations, including previous work by our laboratory, indicate that maternal caffeine consumption is associated with intrauterine growth retardation and impaired fetal length growth. Skeletal development is critical for length growth. In the present study, our goals were to determine the effects of prenatal caffeine exposures on fetal skeletal growth and to investigate the mechanisms associated with such effects. Pregnant Wistar rats were injected intragastrically with 120mg/kg of caffeine intragastrically each day from gestational days 11-20. Maternal prenatal caf

Maternal dibutyl benzene-1,2-dicarboxylate exposure accelerates bone marrow mesenchymal stem cells senescence to induce skeletal retardation in male offspring mice.

Dibutyl benzene-1,2-dicarboxylate (DBP), a ubiquitous plasticizer, readily crosses the placenta, posing a risk to male offspring development. Previous studies have found that maternal DBP exposure leads to multiple organ development disorders in male offspring, but the effect on skeletal development in male offspring mice is unclear. In this study, pregnant mice were orally administered corn oil from day 12 of pregnancy or given different doses of DBP by gavage. Our results showed that prenatal DBP exposure induced dose-dependent deterioration in the male offspring's femoral bone microarchitec

Particulate matter exposure induces maternal scalp hair loss after birth in C57/B6 mouse via alteration of inflammatory and apoptotic pathways.

PM 2.5 exposure is associated with a variety of health effects, including effects on the reproductive and skin. However, the relationship between PM2.5 exposure and postpartum hair loss has not been investigated. In this study, we evaluated the effect of PM2.5 exposure on hair loss after birth in mouse model and analyzed possible associated molecular changes. Female mice were exposed to PM2.5 using nasal inhalation method. After 4 weeks, mating tests were conducted and postpartum scalp tissues from PM2.5-exposed mice and those without exposure were harvested and analyzed. Then, human immortali

Maternal high-fat diet exposure is associated with altered hypothalamic microglial development and reduced early postnatal TGFβ1 signaling in male offspring.

BACKGROUND: The hypothalamus plays a central role in maintaining energy homeostasis by integrating peripheral metabolic signals. Maternal high-fat diet (HFD) exposure has been shown to disrupt hypothalamic development and increase offspring susceptibility to metabolic disorders. Microglia, as key regulators of neuroimmune interactions and synaptic remodeling, are increasingly recognized as critical mediators of metabolic programming. During early postnatal development, microglia undergo a transition toward a homeostatic phenotype characterized by markers such as TMEM119, a process that is crit

Maternal liver lipid dysregulation mediates fetal developmental impairments induced by gestational co-exposure to ozone and heat in C57BL/6J Mice.

Global warming has increased the frequency of simultaneous ozone and heat exposure, raising significant public health concerns. This study investigates the impacts of gestational exposure to ozone (1 ppm) and heat (34°C) under real-world conditions on fetal and placental development in C57BL/6 J mice, with a particular focus on the role of maternal hepatic lipid metabolism. Pregnant mice were exposed from gestational day (GD) 0 to GD17. Co-exposure to ozone and heat significantly reduced fetal birth weight and crown-rump length, and exacerbated adverse pregnancy outcomes, while exposure to ozo

Prevention of prolactin reduction: a novel strategy countering multi-generational growth retardation caused by maternal exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin.

Neonatal growth and development are significantly influenced by maternal care and breastfeeding. Our previous research showed that maternal exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in dams reduced prolactin (PRL) levels, nursing behavior, and milk production during lactation. Intracerebroventricular infusion of PRL in TCDD-exposed dams partially reversed these defects in mothers and offspring. However, the mechanism by which maternal TCCD exposure causes reduced PRL levels and multigenerational effects remains unclear. This study aimed to investigate the multigenerational effects…

Maternal dietary deficiencies in folic acid or choline reduce primary neuron viability after exposure to hypoxia through increased levels of apoptosis.

Objective: Ischemic stroke is the leading cause of death and disability globally. By addressing modifiable risk factors, particularly nutrition, the prevalence of stroke and its dire consequences can be mitigated. One-carbon (1C) metabolism is a critical biosynthetic process that is involved in neural tube closure, DNA synthesis, plasticity, and cellular proliferation. Folates and choline are two active components of 1C metabolism. We have previously demonstrated that maternal dietary deficiencies during pregnancy and lactation in folic acid or choline result in worse stroke outcomes in offspr
