Hypothesis Tests of Direct and Indirect Effects Under Various Semicompeting Risks Models.

The semicompeting risks problem involves two time-to-event outcomes where the intermediate outcome may be censored by the primary outcome but not vice versa. It has been shown that semicompeting risks can be formulated as a mediation model, where both the direct effect (DE) and the indirect effect (IE) are studied. This article proposes unified testing procedures based on current causal approaches to evaluate DE and IE under three classic semicompeting risks models: the Clayton copula, gamma frailty, and multistate models. We study the correspondence of the DE and IE with the model parameters

Standardization and Prediction to Control Confounding: Estimating Risk Differences and Ratios for Clinical Interpretations and Decision Making.

What is the added risk for death from smoking? Logistic regression has become the most common statistical method to answer such questions in the biomedical literature. However, the typical analyses estimate odds ratios, a metric too often misunderstood and misinterpreted. Although estimates of risks, and their differences and ratios, offer transparent clinical interpretations, commonly used statistical models have known methodological shortcomings. "Standardization" through modeling, weighting, or matching offers a solution. The goals of this article are to review classical concepts of standar

Survival prediction based on compound covariate under Cox proportional hazard models.

Survival prediction from a large number of covariates is a current focus of statistical and medical research. In this paper, we study a methodology known as the compound covariate prediction performed under univariate Cox proportional hazard models. We demonstrate via simulations and real data analysis that the compound covariate method generally competes well with ridge regression and Lasso methods, both already well-studied methods for predicting survival outcomes with a large number of covariates. Furthermore, we develop a refinement of the compound covariate method by incorporating likelih

Large language models as versatile predictive engines for notifiable infectious diseases.

Accurate forecasting of infectious disease cases and deaths is crucial for public health decision-making. Traditional statistical and machine learning approaches may be challenged by complex temporal patterns and heterogeneous surveillance data. We evaluated large language models (LLMs) for infectious disease forecasting. We collected monthly reported cases and deaths data from China National Notifiable Diseases Surveillance System (NNDSS) from January 2009 to February 2025, and monthly reported cases from the United States NNDSS from 2016 to 2023, covering 79 notifiable infectious diseases an

An evaluation of three statistical estimation methods for assessing health policy effects on prescription drug claims.

BACKGROUND: While the choice of analytical approach affects study results and their interpretation, there is no consensus to guide the choice of statistical approaches to evaluate public health policy change. OBJECTIVES: This study compared and contrasted three statistical estimation procedures in the assessment of a U.S. Food and Drug Administration (FDA) suicidality warning, communicated in January 2008 and implemented in May 2009, on antiepileptic drug (AED) prescription claims. METHODS: Longitudinal designs were utilized to evaluate Oklahoma (U.S. State) Medicaid claim data from January

A sensitivity analysis approach to principal stratification with a continuous longitudinal intermediate outcome: applications to a cohort stepped wedge trial.

Causal inference in the presence of intermediate variables is a challenging problem in many applications. Principal stratification (PS) provides a framework to estimate principal causal effects (PCE) in such settings. However, existing PS methods primarily focus on settings with binary intermediate variables. We propose a novel approach to estimate PCE with continuous intermediate variables in the context of stepped wedge cluster randomized trials (SW-CRTs). Our method leverages the time-varying treatment assignment in SW-CRTs to calibrate sensitivity parameters and identify the PCE under real

Latent Growth Curve Models for Biomarkers of the Stress Response.

Objective: The stress response is a dynamic process that can be characterized by predictable biochemical and psychological changes. Biomarkers of the stress response are typically measured over time and require statistical methods that can model change over time. One flexible method of evaluating change over time is the latent growth curve model (LGCM). However, stress researchers seldom use the LGCM when studying biomarkers, despite their benefits. Stress researchers may be unaware of how these methods can be useful. Therefore, the purpose of this paper is to provide an overview of LGCMs in t

Comparison of AERMOD and CALPUFF models for simulating SO2 concentrations in a gas refinery.

In this study, concentration of SO2 from a gas refinery located in complex terrain was calculated by the steady-state, AERMOD model, and nonsteady-state CALPUFF model. First, in four seasons, SO2 concentrations emitted from 16 refinery stacks, in nine receptors, were obtained by field measurements, and then the performance of both models was evaluated. Then, the simulated results for SO2 ambient concentrations made by each model were compared with the results of the observed concentrations, and model results were compared among themselves. The evaluation of the two models to simulate SO2 conce

Subject-domain approach to the study of air pollution effects on schoolchildren's illness absence.

In this paper, the authors propose a new statistical modeling technique, the subject-domain approach, which is theoretically proven to be equivalent to the time-domain approach in detecting an association between exposure and response with time trends. The authors use an empirical data set from a school absence monitoring study conducted during the 1994-1995 school year in Taiwan to demonstrate this subject-domain approach's application to environmental epidemiologic studies. Because the subject-domain models can control the influential personal confounding factors in the models, they show gre

Divide and recombine approaches for fitting logistic regression to large-scale health surveillance data: application to diabetes risk prediction in BRFSS.

The global rise in diabetes mellitus poses a major public health concern, highlighting the urgent need for effective risk prediction models to support early identification and prevention strategies. Yet, applying conventional statistical techniques to large-scale health surveillance data remains challenging due to memory limitations and high computational demands. One promising alternative, the divide and recombine (D&R) approach partitions the data into smaller subsets, fits logistic models independently within each, and combines the estimates to approximate full-sample maximum likelihood res

An Improved Clinical and Genetics-Based Prediction Model for Diabetic Foot Ulcer Healing.

Objective: The goal of this investigation was to use comprehensive prediction modeling tools and available genetic information to try to improve upon the performance of simple clinical models in predicting whether a diabetic foot ulcer (DFU) will heal. Approach: We utilized a cohort study (n = 206) that included clinical factors, measurements of circulating endothelial precursor cells (CEPCs), and fine sequencing of the NOS1AP gene. We derived and selected relevant predictive features from this patient-level information using statistical and machine learning techniques. We then developed progn

An emission-weighted proximity model for air pollution exposure assessment.

BACKGROUND: Among the most common spatial models for estimating personal exposure are Traditional Proximity Models (TPMs). Though TPMs are straightforward to configure and interpret, they are prone to extensive errors in exposure estimates and do not provide prospective estimates. METHOD: To resolve these inherent problems with TPMs, we introduce here a novel Emission Weighted Proximity Model (EWPM) to improve the TPM, which takes into consideration the emissions from all sources potentially influencing the receptors. EWPM performance was evaluated by comparing the normalized exposure risk va

A Model-Robust G-Computation Method for Analyzing Hybrid Control Studies Without Assuming Exchangeability.

There is growing interest in a hybrid control design for treatment evaluation, where a randomized controlled trial is augmented with external control data from a previous trial or a real-world data source. The hybrid control design has the potential to improve efficiency but also carries the risk of introducing bias. The potential bias in a hybrid control study can be mitigated by adjusting for baseline covariates that are related to the control outcome. Existing methods that serve this purpose commonly assume that the internal and external control outcomes are exchangeable upon conditioning o

Illustrating Implications of Misaligned Causal Questions and Statistics in Settings With Competing Events and Interest in Treatment Mechanisms.

In the presence of competing events, many investigators are interested in a direct treatment effect on the event of interest that does not capture treatment effects on competing events. Classical survival analysis methods that treat competing events like censoring events, at best, target a controlled direct effect: the effect of the treatment under a difficult to imagine and typically clinically irrelevant scenario where competing events are somehow eliminated. A separable direct effect, quantifying the effect of a future modified version of the treatment, is an alternative direct effect notio

Blinded sample size recalculation in randomized controlled trials with analysis of covariance.

In randomized controlled trials, covariate adjustment can improve statistical power and reduce the required sample size compared with unadjusted estimators. Analysis of covariance (ANCOVA) is often used to adjust for baseline covariates when outcomes are continuous. To design a sample size based on ANCOVA, it is necessary to prespecify the association between the outcome and baseline covariates, as well as among the baseline covariates themselves. However, determining these parameters at the design stage is challenging. Although it may be possible to adaptively assess these during the trial an

G-formula with multiple imputation for causal inference with incomplete data.

G-formula is a popular approach for estimating the effects of time-varying treatments or exposures from longitudinal data. G-formula is typically implemented using Monte-Carlo simulation, with non-parametric bootstrapping used for inference. In longitudinal data settings missing data are a common issue, which are often handled using multiple imputation, but it is unclear how G-formula and multiple imputation should be combined. We show how G-formula can be implemented using Bayesian multiple imputation methods for synthetic data, and that by doing so, we can impute missing data and simulate th

Double robust variance estimation with parametric working models.

Doubly robust estimators have gained popularity in the field of causal inference due to their ability to provide consistent point estimates when either an outcome or an exposure model is correctly specified. However, for nonrandomized exposures, the influence function based variance estimator frequently used with doubly robust estimators of the average causal effect is only consistent when both working models (ie, outcome and exposure models) are correctly specified. Here, the empirical sandwich variance estimator and the nonparametric bootstrap are demonstrated to be doubly robust variance es

Multiple Comparisons Procedures for Analyses of Joint Primary Endpoints and Secondary Endpoints.

One of the main challenges in drug development for rare diseases is selecting the appropriate primary endpoints for pivotal studies. Although many endpoints can effectively reflect clinical benefit, their sensitivity often varies, making it difficult to determine the required sample size for study design and to interpret final results, which may be underpowered for some or all endpoints. This complexity is further compounded when there is a desire to support regulatory claims for multiple clinical endpoints and dose regimens due to the issues of multiplicity and sample size constraints. Joint

A mixed-effects Bayesian regression model for multivariate group testing data.

Laboratories use group (pooled) testing with multiplex assays to reduce the time and cost associated with screening large populations for infectious diseases. Multiplex assays test for multiple diseases simultaneously, and combining their use with group testing can lead to highly efficient screening protocols. However, these benefits come at the expense of a more complex data structure which can hinder surveillance efforts. To overcome this challenge, we develop a general Bayesian framework to estimate a mixed multivariate probit model with data arising from any group testing protocol that use
