Disruption of urinary odor preference and lordosis behavior in female mice given lesions of the medial amygdala.

Previous research showed that axonal inputs to both anterior and posterior subdivisions of the medial amygdala from the main and accessory olfactory bulbs of female mice, respectively, process volatile and non-volatile pheromonal signals from male conspecifics. In the present study we found that bilateral electrolytic lesions that included posterior portions, but not the anterior subdivision alone of the medial amygdala (Me) blocked the preference of estrous female mice to investigate volatile urinary odors from testes-intact vs. castrated males. Similar results were obtained in separate tests

Drosophila olfaction: the end of stereotypy?

Recent work has demonstrated substantial wiring and functional stereotypy in the fly olfactory system. In this issue of Neuron, Murthy et al. demonstrate that in the mushroom body, a site of olfactory associative learning, this initial peripheral stereotypy gives way to functionally nonstereotyped circuits.

Activity-dependent plasticity in an olfactory circuit.

Olfactory sensory neurons (OSNs) form synapses with local interneurons and second-order projection neurons to form stereotyped olfactory glomeruli. This primary olfactory circuit is hard-wired through the action of genetic cues. We asked whether individual glomeruli have the capacity for stimulus-evoked plasticity by focusing on the carbon dioxide (CO2) circuit in Drosophila. Specialized OSNs detect this gas and relay the information to a dedicated circuit in the brain. Prolonged exposure to CO2 induced a reversible volume increase in the CO2-specific glomerulus. OSNs showed neither altered mo

[Neuropathological study on progression of the limbic degeneration in senile dementia of Alzheimer type].

Neuropathological study on the limbic lesion of 33 autopsy cases with senile dementia of Alzheimer type (SDAT) showed as follows. 1) The entorhinal cortex was more atrophied than the hippocampus. 2) Neuronal loss was found in the 2nd and 3rd layers of the entorhinal cortex, irrespective of the different numbers of senile plaques and neurofibrillary tangles (NFTs). 3) Fibrillary gliosis occurred in the stratum lacunosum of the hippocampus, irrespective of the different degrees of neuronal loss in the stratum pyramidale of the hippocampus. 4) The prosubiculum showed gliosis disproportional to ne

Migration of GnRH-immunoreactive neurons from the olfactory placode to the brain: a study using avian embryonic chimeras.

Previous studies suggest that gonadotropin-releasing hormone (GnRH) neurons appear in the olfactory placode and subsequently migrate into the brain during embryonic development. The aim of the present study was to obtain direct evidence for migration of GnRH neurons from the olfactory placode into the brain. Olfactory placodes from quail embryos were transplanted isotopically and isochronically, to replace the unilaterally ablated olfactory placodes of chick embryos. The chimeric embryos were allowed to develop for several days until they reached the embryonic stages when GnRH neurons are seen

Preservation of olfaction in anterior craniofacial approaches.

Through the combined efforts of neurosurgeons, head and neck surgeons, and craniofacial surgeons, the standard transbasal approach to the frontal fossa has been modified to include removal of the orbital roofs, nasion, and ethmoid sinuses. This approach has been combined further with facial disassembly procedures to provide extensive midline exposure to the midface and clival region. Extended frontal approaches, however, necessitate removal of the crista galli and sectioning of the olfactory rootlets with the associated risk of anosmia, cerebrospinal fluid (CSF) leak, and the need for complex

Hypothyroidism disrupts neural development in the olfactory epithelium of adult mice.

Adult mice made hypothyroid with propylthiouracil (PTU) lose their sense of smell. This is prevented by daily administration of thyroxine. As thyroxine is necessary for the correct development of the nervous system it may also be necessary for the genesis of new olfactory receptor neurones, a process that continues into adulthood. Adult mice were treated with PTU, injected with [3H]thymidine after 54 days and killed 5 or 15 days later. Microscopic analysis of the olfactory epithelium after autoradiography revealed similar numbers of labelled nuclei in the basal cell layer of the olfactory epit

Synaptic reorganization in the medial amygdaloid nucleus after lesion of the accessory olfactory bulb of adult rat. II. New synapse formation in the medial amygdaloid nucleus by fibers from the bed nucleus of the stria terminalis.

The rearrangement of terminations from the bed nucleus of stria terminalis (BST) was examined in the medial amygdaloid nucleus (MAN) at 2 months following the lesion of the accessory olfactory bulb (AOB) using an electron microscopy and degeneration study. At 2 days following a BST lesion, the number of degenerating synapses was 0.7 +/- 0.1 (mean +/- S.E.M.) per unit area (2500 microns2) in the molecular layer, and 3.0 +/- 0.3 in the cellular part. At 2 months after an AOB lesion, the degenerating synapses from the AOB had completely disappeared from the MAN. The BST was then lesioned at 2 mon

Indocyanine Green as a Marker for Nose-to-Brain Delivery Pathways, Brain Distribution, and PLGA Nanoparticle Efficiency.

This study aims to assess the rate and duration of rat brain retention after a single intranasal administration of indocyanine green (ICG) as an aqueous solution or encapsulated in poly(D,L-lactide-co-glycolide) (PLGA) nanoparticles. Near-infrared fluorescence emission of ICG from the brain and visceral organs was measured at 1, 4, and 24 h, as well as at 1 and 2 weeks after administration. It was observed that both ICG formulations stained the olfactory bulbs and brainstem, the latter mainly in the basolateral region of the pons. Reduced staining was observed on day 7 after treatment, and the

Recurrent synapses between CO2-sensitive olfactory sensory neurons enable robust CO2 detection in Aedes aegypti mosquitoes.

The mosquito Aedes aegypti's human host-seeking behavior depends on the integration of multiple sensory cues. One of these cues, carbon dioxide (CO2), gates odorant and heat pathways and activates host-seeking behavior. The neuronal circuits underlying processing of CO2 information remain unclear. We used automated serial-section transmission electron microscopy (EM) to image and reconstruct the circuitry of the glomeruli that are innervated by the Ae. aegypti maxillary palp, including the glomerulus that responds to CO2. Specification of GnRH-1 neurons by antagonistic FGF and retinoic acid signaling.

A small population of neuroendocrine cells in the rostral hypothalamus and basal forebrain is the key regulator of vertebrate reproduction. They secrete gonadotropin-releasing hormone (GnRH-1), communicate with many areas of the brain and integrate multiple inputs to control gonad maturation, puberty and sexual behavior. In humans, disruption of the GnRH-1 system leads to hypogonadotropic gonadism and Kallmann syndrome. Unlike other neurons in the central nervous system, GnRH-1 neurons arise in the periphery, however their embryonic origin is controversial, and the molecular mechanisms that co

Gamma and Beta Oscillations Define a Sequence of Neurocognitive Modes Present in Odor Processing.

UNLABELLED: Olfactory system beta (15-35 Hz) and gamma (40-110 Hz) oscillations of the local field potential in mammals have both been linked to odor learning and discrimination. Gamma oscillations represent the activity of a local network within the olfactory bulb, and beta oscillations represent engagement of a systemwide network. Here, we test whether beta and gamma oscillations represent different cognitive modes using the different demands of go/no-go and two-alternative choice tasks that previously were suggested to favor beta or gamma oscillations, respectively. We reconcile previous st

Reproduction phase-related variations in neuropeptide Y immunoreactivity in the olfactory system, forebrain, and pituitary of the female catfish, Clarias batrachus (Linn.).

The aim of this study was to determine whether neuropeptide Y (NPY) immunoreactivity in the cells and fibers in the forebrain and pituitary of Clarias batrachus is linked to the annual reproductive cycle. A steady rise in luteinizing hormone (LH) immunoreactivity was seen in the pituitary through preparatory (February-April) and prespawning (May-June) phases; it was greatly reduced during spawning (July-August; P < 0.001) and partially replenished during postspawning (September-November; P < 0.01) through resting (December-January) phases. Although NPY immunoreactivity in olfactory receptor ne

Olfactory identification in combat-related posttraumatic stress disorder.

Recent neuropsychological conceptualizations of posttraumatic stress disorder (PTSD) implicate dysfunction of the fronto-limbic system, a brain system thought to be involved in the mediation of emotion. However, few studies have examined fronto-limbic subregions, such as the orbitofrontal cortex, in PTSD. As a measure of orbitofrontal integrity, olfactory identification was assessed in 26 Vietnam War veterans with PTSD, 25 Vietnam War veterans without mental disorders, and 17 Vietnam-era, non-war-zone veterans without mental disorders. Relative to veterans without PTSD, those diagnosed with PT

Metabolic mapping of functional activity in the olfactory system of normal and hypogonadal (hpg) mice.

The hypogonadal mouse, which lacks gonadotropin-releasing hormone, has been suggested as an animal model of Kallmann's syndrome, one symptom of which is hyposmia. We have determined the metabolic activity of the olfactory system, in normal and hypogonadal mice, using [14C]-2-deoxyglucose quantitative autoradiography. In the olfactory lobes, deoxyglucose uptake was greatest in the glomerular and granule cell layers and low in the olfactory nerve layer and bulb core. The pattern of uptake was similar in both hypogonadal and normal mice breathing filtered air. Exposure of normal mice to ethyl ace

Purification and characterization of an antigen that is spatially segregated in the primary olfactory projection.

The monoclonal antibody RB-8 heavily labels axons from the ventrolateral olfactory epithelium and their terminals in the glomeruli of the ventrolateral olfactory bulb, but leaves the axons from the dorsomedial epithelium unstained or lightly stained. RB-8 reacts with a 125 kDa membrane protein in both olfactory nerve and other parts of the CNS (Schwob and Gottlieb, 1986). Here we report further characterization of the molecular nature and cellular localization of the RB-8 antigen. The RB-8 antigen is exposed on the surface of olfactory axons. Individual axons and axon bundles stain when explan

Convulsants antagonise inhibition in the olfactory cortex slice.

The effects of some gamma-aminobutyric acid (GABA) antagonists and other "convulsants" have been tested on neurones of the isolated olfactory cortex slice preparation of the guinea-pig using single cell intracellular and gross extracellular recording techniques. Bicuculline, picrotoxin, strychnine, leptazol, bemegride, and also theophylline and d-tubocurarine all increased the duration and amplitude of the excitatory postsynaptic potential, thereby producing a seizure-like discharge. These drugs reduced and shortened the peak conductance increase during the inhibitory postsynaptic potential (i

Contribution of astrocytes to familial risk and clinical manifestation of schizophrenia.

Previous studies have implicated several brain cell types in schizophrenia (SCZ), but the genetic impact of astrocytes is unknown. Considering their high complexity in humans, astrocytes are likely key determinants of neurodevelopmental diseases, such as SCZ. Human induced pluripotent stem cell (hiPSC)-derived astrocytes differentiated from five monozygotic twin pairs discordant for SCZ and five healthy subjects were studied for alterations related to high genetic risk and clinical manifestation of SCZ in astrocyte transcriptomics, neuron-astrocyte co-cultures, and in humanized mice.
