Cholinergic 'blockade' as a model for cholinergic depletion. A comparison of the memory deficits with those of Alzheimer-type dementia and the alcoholic Korsakoff syndrome.

The effect of cholinergic 'blockade' on human memory performance as a model for the effect of cholinergic depletion in clinical disorders was investigated. A wide range of memory functions was assessed in 70 subjects, using tests which were identical or closely similar to those which have previously been employed in clinical studies of Alzheimer and Korsakoff patients. In addition, a physiological measure of the degree of central cholinergic blockade was included, as well as measures of subjective arousal and objective attention. It was found that cholinergic blockade had no significant effect

Different cholinergic pathways are involved in the improvement induced by CCK-8 and by ACTH-(1-24) in massive acute hemorrhage, in rats.

Cholecystokinin octapeptide (CCK-8) (20 micrograms/kg i.v.) and tetracosactide [ACTH-(1-24)] (160 micrograms/kg i.v.) restore blood pressure and allow rats subjected to otherwise invariably fatal acute hemorrhage to survive. Atropine sulphate (2-8 mg/kg i.p.), which crosses the blood-brain barrier, dose-dependently prevents this effect both in the case of ACTH-(1-24) and in that of CCK-8. On the other hand, atropine methyl bromide (2-8 mg/kg i.p.), which does not cross the blood-brain barrier, prevents the effect in the case of CCK-8, but not in that of ACTH-(1-24). These data suggest that a c

Effects of metoclopramide and domperidone on cholinergically mediated contractions of human isolated stomach muscle.

The experiments examine the actions of metoclopramide and domperidone on the responses evoked by electrical field stimulation or by acetylcholine, in longitudinal muscle strips obtained from human stomach. Electrical field stimulation evoked contractions which were predominantly cholinergically mediated; metoclopramide 0.28-28 microM caused a concentration-dependent increase in the height of these contractions. In the presence of atropine and barium chloride, electrical stimulation evoked relaxations of the stomach muscle, probably by stimulating non-adrenergic, non-cholinergic inhibitory nerv

Parasympathetic cardiovascular control in human disease: a critical review of methods and results.

A variety of experimental approaches have been used to alter reflex parasympathetic cardiovascular control in normal human subjects and in patients with cardiovascular diseases. This review critically appraises several of these methods (the Valsalva maneuver, upright tilt, lower body negative pressure, pressor drug infusion, neck suction, and coronary arteriography) and summarizes the normal response patterns of healthy subjects and the abnormal response patterns of some patients with cardiovascular disease. Tests of parasympathetic function tend to be heterogeneous; they are not standardized,

Motor innervation of the toad iris (Bufo marinus).

The sphincter pupillae muscle cells in the iris of Bufo marinus contract autonomously in response to bright light, causing a rapid constriction of the pupil. A strong sympathetic beta-adrenergic inhibition of the sphincter pupillae is apparent in this species. The inhibitory fibers can originate in the second, third, or fourth ventral spinal roots. No strong, consistent excitatory innervation of the toad iris was detected, even by transmural stimulation of the isolated iris. Pupilloconstriction occasionally resulted from stimulation of the 3rd or 5th cranial nerves, but the effect was small (1

Evidence against cholinergic mediation of the effect of angiotensin II on the guinea pig ileum.

The belief that the smooth muscle contracting activity of angiotensin II (angiotensin) in the guinea pig ileum is partly mediated by release of acetylcholine was reexamined, with the following results. 1. Atropine did not reduce the maximum contraction produced by angiotensin, although it caused a shift to the right of the log dose-response curve (dose ratio = 2.2). A similar shift was observed with histamine, bradykinin and BaCl2. 2. A moderate potentiation of angiotensin by eserine was also observed, which was similarly found for the other agonists. 3. A previous report that atropine blocks

Cholinergic mechanisms in central thermoregulation in pigeons.

1. In unanaesthetized pigeons the effect on cloacal temperature was studied of acetylcholine (ACh), carbachol, atropine and (+)-tubocurarine injected into a cannulated lateral cerebral ventricle. The experiments were carried out at an ambient temperature of 19-25 degrees C. 2. ACh or carbachol injected intraventricularly produced hyperthermia, and in larger doses hyperthermia followed by hypothermia. These were central effects because they were not obtained when these drugs were injected in the same doses intravenously. 3. Atropine injected intraventricularly produced hypothermia which was gre

Dysregulation of the autonomic nervous system predicts the development of the metabolic syndrome.

CONTEXT: Stress is suggested to lead to metabolic dysregulations as clustered in the metabolic syndrome. Although dysregulation of the autonomic nervous system is found to associate with the metabolic syndrome and its dysregulations, no longitudinal study has been performed to date to examine the predictive value of this stress system in the development of the metabolic syndrome. OBJECTIVE: We examined whether autonomic nervous system functioning predicts 2-year development of metabolic abnormalities that constitute the metabolic syndrome. DESIGN: Data of the baseline and 2-year follow-up as

Improvements of cardiac function and metabolic parameters by sodium-glucose cotransporter 2 inhibitors with no significant effects on sympathetic or parasympathetic activity in chronic heart failure.

Sodium-glucose cotransporter 2 (SGLT2) inhibitors have demonstrated significant cardiovascular benefits, including reductions in hospitalizations and mortality among patients with heart failure (HF). However, the mechanisms underlying these benefits, particularly their effects on autonomic nervous system activity, remain incompletely understood. This single-center, prospective observational study included 11 patients with chronic HF who were newly initiated on SGLT2 inhibitors. Sympathetic nerve activity was assessed using 123I-metaiodobenzylguanidine (123I-MIBG) scintigraphy, while parasympat

Effects of transcutaneous auricular vagus nerve stimulation on cardiovascular autonomic control in health and disease.

Autonomic nervous system (ANS) dysfunction is a well-known feature of cardiovascular diseases (CVDs). Studies on heart rate variability (HRV), a non-invasive method useful in investigating the status of cardiovascular autonomic control, have shown that a predominance of sympathetic modulation not only contributes to the progression of CVDs but has a pivotal role in their onset. Current therapies focus more on inhibition of sympathetic activity, but the presence of drug-resistant conditions and the invasiveness of some surgical procedures are an obstacle to complete therapeutic success. On the

No modulation of postprandial metabolism by transcutaneous auricular vagus nerve stimulation: a cross-over study in 15 healthy men.

Experimental evidence suggests a crucial role of the autonomic nervous system in whole body metabolism with major regulatory effects of the parasympathetic branch in postprandial adaptation. However, the relative contribution of this mechanism is still not fully clear in humans. We therefore compared the effects of transcutaneous auricular vagus nerve stimulation (taVNS, Cerbomed Nemos) with sham stimulation during an oral glucose tolerance test in a randomized, single-blind, cross-over design in 15 healthy lean men. Stimulation was performed for 150 min, 30 min before and during the entire or

Pathogenesis of Parkinson disease--the gut-brain axis and environmental factors.

Parkinson disease (PD) follows a defined clinical pattern, and a range of nonmotor symptoms precede the motor phase. The predominant early n … e gut and spreads via trans-synaptic cell-to-cell transfer of pathology through the sympathetic and parasympathetic nervous systems to the substantia nigra and the CNS. We also consider how environmental factors might trigger pathogenesis, and the potential for therapeutically targeting the mechanisms of these initial stages.

Neurotrophic factor receptor RET: structure, cell biology, and inherited diseases.

RET (REarranged during Transfection) is a transmembrane receptor tyrosine kinase that is activated by a complex consisting of a soluble glial cell line-derived neurotrophic factor (GDNF) family ligand (GFL) and a glycosyl phosphatidylinositol-anchored co-receptor, GDNF family receptors alpha (GFRalpha). RET signalling is crucial for the development of the enteric nervous system. RET also regulates the development of sympathetic, parasympathetic, motor, and sensory neurons, and is necessary for the postnatal maintenance of dopaminergic neurons. The effect of GFLs on sensory, motor, and dopamine

Vagal tone, development, and Gray's motivational theory: toward an integrated model of autonomic nervous system functioning in psychopathology.

In the last decade, cardiac vagal tone has emerged as a psychophysiological marker of many aspects of behavioral functioning in both children and adults. Research efforts during this time have produced an extensive list of vagal tone correlates that includes temperamental variables as well as both anxious/internalizing and disruptive/externalizing behaviors. This potentially confusing state of affairs is compounded by developmental shifts in vagal tone behavior relations that to date have not been elucidated. In this paper, the vagal tone literature is reviewed, and discrepancies, including th
