Spinocerebellar ataxia type 8 (SCA8) is a slowly progressive ataxia causally associated with untranslated CTG repeat expansion on chromosome 13q21. However, the role of the CTG repeat in SCA8 pathology is not yet well understood. Therefore, we studied the length of the SCA8 CTA/CTG expansions (combined repeats, CRs) in 115 patients with ataxia, 64 unrelated individuals with non-triplet neuromuscular diseases, 70 unrelated patients with schizophrenia, and 125 healthy controls. Only one patient with apparently sporadic ataxia was identified with an expansion of 100 CRs. He had inherited the expa

The blood - brain barrier (BBB) is a dynamic regulator of brain homeostasis, and its dysfunction is a hallmark of many neurological and psychiatric disorders. Yet, in most conditions, the causal relationship between BBB injury and disease progression remains unclear, as shared systemic risk factors, such as inflammation, infection, oxidative stress, and genetic predisposition, produce highly variable patterns of barrier disruption. Epigenetic mechanisms, including DNA and RNA methylation, histone modifications, chromatin remodeling, and noncoding RNAs, have emerged as key regulators of BBB int

Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease that leads to progressive joint destruction, extra-articular manifestations, disability, and increased mortality. Early detection, particularly in seronegative patients, remains challenging because current diagnostic criteria based on joint involvement, serology, and acute-phase reactants may fail to identify disease at its earliest stages. Epigenetic mechanisms, including DNA and RNA methylation, histone modifications, and non-coding RNAs (ncRNAs), provide a dynamic interface between genetic predisposition and environmental tr

The Rhabdoviridae family comprises negative-sense single-stranded RNA viruses that cause severe infections in a wide range of hosts. Increasing evidence indicates that host noncoding RNAs (ncRNAs), including microRNAs (miRNAs), long noncoding RNAs (lncRNAs), and circular RNAs (circRNAs), play central roles in regulating rhabdovirus replication, pathogenesis, and host immune responses. These ncRNAs exert both antiviral and proviral effects through modulation of innate immune signaling pathways, regulation of gene expression, and control of inflammatory responses. Antiviral ncRNAs suppress viral

Synthetic messenger RNA (mRNA) is emerging as next generation vaccine platform. As natural 5'UTRs of particular genes do not work for other than its own mRNA, it is essential to identify an ideal 5'UTR supporting higher translation efficiency for different antigens. Six natural 5'UTRs derived from α, β-globin genes of host animals and other viral genes were cloned upstream to the coding sequences of reporter proteins. The corresponding capped mRNA were produced through in vitro transcription and the translation efficiency was assessed using flow cytometry, bioluminescence, immunofluorescence a

As a major obstacle in cancer treatment, drug resistance is increasingly recognized that non-coding RNA (ncRNA) plays an important role in regulating cell plasticity. This review comprehensively explores how ncRNAs, including microRNAs, long non-coding RNAs, and circular RNAs, drive drug resistance by altering identity plasticity (epithelial-mesenchymal transition and stemness), state plasticity (cell fate selection and metabolic reprogramming), and communication plasticity in the tumor microenvironment. Due to the complexity of the ncRNA network, Chinese medicines (CMs) with multi-target prop

BACKGROUND: Dengue virus (DENV), with its four antigenically distinct serotypes, is the etiological agent of dengue fever, which is endemic in tropical and subtropical regions but has recently spread to previously non-endemic areas such as Europe. Given the growing body of evidence suggesting that sequence diversification in the 3' UTR of DENV contributes to its epidemiological fitness and host adaptation, we conducted phylogenetic and genetic diversity analyses on four DENV 3' UTR datasets (DENV-1, DENV-2, DENV-3 and DENV-4).
METHODS: A maximum likelihood phylogenetic analysis was performed

AIMS: Diabetic cardiomyopathy (DCM) is a major complication of diabetes and a risk factor for cardiovascular disease. Endothelial dysfunction is central to DCM, and endothelial-to-mesenchymal transition (EndMT) is a key form of endothelial dysfunction in diabetes. EndMT in DCM has been well-studied in model systems and has been found to be epigenetically regulated by non-coding RNAs (ncRNAs). However, EndMT in DCM and its associated epigenetic changes need further characterization in human patients. It is also not known if ncRNAs are affected by changes in DNA methylation in DCM. This study ai

BACKGROUND: Schistosomiasis is a prevalent infectious disease caused by the parasitic trematodes of the genus Schistosoma. Praziquantel (PZQ), a safe and affordable drug, is the recommended oral treatment for schistosomiasis. The main pathologic manifestation of schistosomiasis is liver injury. However, the role and interactions of various RNA molecules in the effect of PZQ on the liver after S. japonicum infection have not been elucidated.
RESULTS: In this study, C57BL/6 mice were randomly divided into the control group, infection group, and PZQ treatment group. Total RNA was extracted from

Non-coding RNAs (ncRNAs) are a class of RNA molecules that do not have the potential to encode proteins. Meanwhile, they can occupy a significant portion of the human genome and participate in gene expression regulation through various mechanisms. Gestational diabetes mellitus (GDM) is a pathologic condition of carbohydrate intolerance that begins or is first detected during pregnancy, making it one of the most common pregnancy complications. Although the exact pathogenesis of GDM remains unclear, several recent studies have shown that ncRNAs play a crucial regulatory role in GDM. Herein, we p

Lithium chloride (LiCl) is a widely used and extensively researched drug for the treatment of bipolar disorder (BD). As a result, LiCl has been the subject of research studying its toxicity, mode of action, and downstream cellular responses. LiCl has been shown to influence cell signaling and signaling transduction pathways through protein kinase C and glycogen synthase kinase-3 in mammalian cells. LiCl's significant downstream effects on the translational pathway necessitate further investigation. In yeast, LiCl is found to lower the activity and alter the expression of PGM2, a gene encoding

A great number of studies have confirmed that long noncoding RNA (lncRNA) are involved in the regulation of inflammatory response in acute gouty arthritis (AGA). This paper aimed to survey the regulatory mechanism of H19 on AGA. The expression of serum H19 in all subjects was examined by qRT-PCR. The ROC curve was used to estimate the diagnostic value of H19 for AGA. THP-1 cells were induced by MSU to establish in vitro AGA cell model. The concentrations of cytokines such as IL-1β, IL-8, and TNF-α were tested by ELISA. Luciferase reporter gene analysis was used to verify the interaction betwee

Influenza virus infection through seasonal epidemics and occasional pandemics has been a major public health concern for decades. Incomplete protection from vaccination and increased antiviral resistance due to frequent mutations of influenza viruses have led to a continuous need for new therapeutic options. The functional significance of host protein and influenza virus interactions has been established, but relatively less is known about the interaction of host noncoding RNAs, including microRNAs and long noncoding RNAs, with influenza viruses. In this review, we summarize host noncoding RNA

The heterochromatin environment plays a central role in silencing genes associated with the malaria parasite's development, survival in the host, and transmission to the mosquito vector. However, the underlying mechanism regulating the dynamic chromatin structure is not understood yet. Here, we have uncovered that Plasmodium falciparum Rrp6, an orthologue of eukaryotic RNA exosome-associated RNase, controls the silencing of heterochromatic genes. PfRrp6 knockdown disrupted the singular expression of the GC-rich ncRNA RUF6 family, a known critical regulator of virulence gene expression, through

Steroid-induced necrosis of femoral head (SINFH) is a femoral head necrotic disease caused by prolonged use of hormones. The detailed pathogenesis has not been fully demonstrated. In this study, we employed the bioinformatics approach to probe the roles of SINFH inhibitors. Core dysfunction modules related to SINFH was obtained. Meanwhile, GO and KEGG analysis of genes in dysfunction modules are carried out. Furthermore, the pivot prediction analysis of dysfunction modules related to ncRNA and transcription factor (TF) has been performed. The functions of the enriched modules were focused on m

Head and neck cancer (HNC), which includes epithelial malignancies of the upper aerodigestive tract (oral cavity, oropharynx, pharynx, hypopharynx, larynx, and thyroid), are slowly but consistently increasing, while the overall survival rate remains unsatisfactory. Because of the multifunctional anatomical intricacies of the head and neck, disease progression and therapy-related side effects often severely affect the patient's appearance and self-image, as well as their ability to breathe, speak, and swallow. Patients with HNC require a multidisciplinary approach involving surgery, radiation t

Using a mouse model, our group recently described an association between chronic paternal alcohol use prior to conception and deficits in offspring growth. Here, we sought to determine the impact of alcohol exposure on male reproductive physiology and the association of sperm-inherited noncoding RNAs with the transmission of the observed growth defects. Alcohol exposure did not appreciably alter male reproductive physiology or fertility. However, chronic alcohol use reproducibly induced late-term fetal growth restriction in the offspring, which correlated with a shift in the proportional ratio

For a period in the mid-1990s, soon after the discovery of the involvement of trinucleotide repeat expansions in fragile-X syndrome (both A and E), Huntington's disease, myotonic dystrophy, and a number of hereditary ataxias, there was a clear sense that this new disease mechanism might provide answers for psychiatric disorders. Given the then failures to replicate initial genetic linkage findings for schizophrenia (SCZ) and bipolar disorder (BD), a greater emphasis was placed on the role of complex and non-Mendelian mechanisms, and repeat instability appeared to have the potential to provide

MicroRNA 122 (miR-122) stimulates the replication and translation of hepatitis C virus (HCV) RNA by binding to two adjacent sites, S1 and S2, within the HCV 5'UTR. We demonstrated previously that the miR-122 antagomir miravirsen (SPC3649) suppresses the infection of HCV strain JFH1-based recombinants with HCV genotypes 1-6 5'UTR-NS2 in human hepatoma Huh7.5 cells. However, specific S1 mutations were permitted and conferred virus resistance to miravirsen treatment. Here, using the J6 (genotype 2a) 5'UTR-NS2 JFH1-based recombinant, we performed reverse-genetics analysis of S1 (ACACUCCG, correspo
