Metharbital metabolism was impaired in rats after acute hepatic damage induced by carbon tetrachloride. Compared to control rats, hepatic damage prolonged the metharbital sleeping time and reduced the slopes of log metharbital plasma concentration-time curves. Renal contributions to metharbital elimination from plasma were negligible since only about 6% of the metharbital administered was eliminated unchanged in urine. Female Wistar-strain rats were starved for 14-19 days by feeding approximately 1/4 of the amount consumed by ad libitum fed controls. Dimethyl sulphoxide (DMSO), 3 g/kg body weight, administered daily by the intraperitoneal route, potentiated the proteinuria and formation of tubular casts in aminonucleoside of puromycin (PA) induced nephrosis in Sprague-Dawley rats. The effect was evident at 4 as well as 8-9 days following PA administration. In the absence of PA, DMSO did not induce proteinuria or cast formation. The mechanism by which DMSO enhanced proteinuria and cast formation is not known.

Experimental models of meningeal gliomatosis (MG) have been produced by intracisternal inoculation of rat C6 and 9L glioma cells into Wistar and Fischer 344 rats, respectively. Tumor growth was steady and rapid in both MG models when more than 10(6) tumor cells were implanted. The median survival time of the rats inoculated with tumor cells was inversely related to the number of the cells inoculated. The pharmacokinetics of cyanamide after 35 mg kg-1 intravenous and oral administration to fasted and unfasted rats have been investigated. The plasma level are fitted well by a two-compartment open model. The half-life (t 1/2 beta) of cyanamide in the rat was 1 hour and the plasma clearance (C1p) was 0.02 (1 kg-1) min-1. Food does not appear to modify the absolute bioavailability of cyanamide (F = 93.3 per cent fasted, F = 85.5 per cent unfasted), although it does retard drug absorption.

The influence of rifaximin (L/105) on conception and pregnancy in rats up to F1 progeny was reported. Rifaximin showed a good fertility rate and no dose-related differences in the number of viable or dead fetuses per litter (Fo progeny) up to 100 mg/kg. The number of live or dead pups at birth, pup weight, gestation length and survival of offspring to weaning were not influenced up to the highest dose, 100 mg/kg (Fo-F1 progeny).

A standardized stenosis was induced by applying a silver clip around the left renal artery in male rats. Trifluoperazine was administered to pregnant mice and rats by gastric intubation during the period of organogenesis. Dams were treated at doses of 0.5, 5, 50 (mice and rats) and 100 (only rats) mg/kg/day.
