Association between Staphylococcus aureus nasal carriage and disease phenotype in patients affected by systemic lupus erythematosus.

BACKGROUND: Staphylococcus aureus (SA) is a commensal bacterium representing one of the most important components of the skin microbiome, mostly isolated in the anterior nares. A higher rate of SA nasal colonization in patients affected by Wegener's granulomatosis and rheumatoid arthritis compared with healthy subjects (HS) has been described. No studies focusing on systemic lupus erythematosus (SLE) are available. We aimed at analyzing the prevalence of SA nasal carriers in an SLE cohort and evaluating correlation between nasal colonization and clinical, laboratory and therapeutic features. …

Lactiplantibacillus pentosus JWN01 and Lactiplantibacillus plantarum JWN02 attenuate renal fibrosis and pathological autophagy in hyperuricemic nephropathy via gut-kidney axis.

Hyperuricemic nephropathy (HN) is a worldwide metabolic disorder marked by uric acid (UA) imbalance and renal tubulointerstitial fibrosis, yet therapies that both lower UA and prevent fibrosis remain limited. Targeting the gut-kidney axis with probiotics is a promising strategy, but most candidates are food-derived and not human-adapted. We isolated two Lactiplantibacillus strains, Lactiplantibacillus pentosus JWN01 and Lactiplantibacillus plantarum JWN02, from the healthy newborn skin representing a relatively unperturbed, early-life human microbiome. In vitro, these two human-derived probiot

Postbiotics as Emerging Therapeutics for Allergic Diseases: A Novel Approach Beyond Live Biologics.

Allergic diseases, including asthma, atopic dermatitis, allergic rhinitis, and food allergies, are characterized by immune dysregulation, epithelial barrier dysfunction, and exaggerated type 2 inflammation. Although current therapeutic strategies have improved disease management, many treatments remain symptomatic, costly, and insufficiently effective in a substantial proportion of patients. Increasing recognition of the gut-immune axis has shifted attention toward microbiome-derived therapeutic approaches. However, safety concerns and inconsistent clinical outcomes associated with live probio

The neovaginal microbiome of transgender women post-gender reassignment surgery.

BACKGROUND: Gender reassignment surgery is a procedure some transgender women (TW) undergo for gender-affirming purposes. This often includes the construction of a neovagina using existing penile and scrotal tissue and/or a sigmoid colon graft. There are limited data regarding the composition and function of the neovaginal microbiome representing a major gap in knowledge in neovaginal health. RESULTS: Metaproteomics was performed on secretions collected from the neovaginas (n = 5) and rectums (n = 7) of TW surgically reassigned via penile inversion/scrotal graft with (n = 1) or without (n = 4

The Pathogenesis and Treatment of Hidradenitis Suppurativa.

Hidradenitis suppurativa (HS) is a multifactorial disease involving the skin and subcutaneous tissues characterized by deep-seated, painful nodules and abscesses with draining sinus tracts. It affects mostly younger individuals between the ages of 18 and 34. The discomfort and embarrassment that patients affected by HS experience negatively impact their daily lives. It is associated with decreased quality of life and high rates of comorbid depression and anxiety. The rate of depression in HS was reported to be as high as 26%. Its pathogenesis is multifactorial and as such requires a multimodal

Analysis of the Conjunctival Microbiome in Patients with Atopic Keratoconjunctivitis and Healthy Individuals.

BACKGROUND: Atopic dermatitis (AD) has been clarified that imbalance of bacterial and fungal communities in the skin and gut play key roles in immunologic dysfunction. Atopic keratoconjunctivitis (AKC), one of severe ophthalmic manifestation of AD, could be related with dysbiosis as same as AD. OBJECTIVE: In this case-control study, the roles of conjunctival microbial communities in AKC were evaluated by a comparative analysis with healthy controls (HCs). METHODS: 16S rRNA sequencing was used to construct libraries of compositional information for a total of 30 volunteers including 20 patien

Human Microbiota Network: Unveiling Potential Crosstalk between the Different Microbiota Ecosystems and Their Role in Health and Disease.

The human body is host to a large number of microorganisms which conform the human microbiota, that is known to play an important role in health and disease. Although most of the microorganisms that coexist with us are located in the gut, microbial cells present in other locations (like skin, respiratory tract, genitourinary tract, and the vaginal zone in women) also play a significant role regulating host health. The fact that there are different kinds of microbiota in different body areas does not mean they are independent. It is plausible that connection exist, and different studies have sh

Past, Present and Future Perspectives of Forensic Genetics.

Forensic genetics has experienced remarkable advancements over the past decades, evolving from the analysis of a limited number of DNA segments to comprehensive genome-wide investigations. This progression has significantly improved the ability to establish genetic profiles under diverse conditions and scenarios. Beyond individual identification, forensic genetics now enables the inference of physical traits (e.g., eye, hair, and skin color, as well as body composition), biogeographic ancestry, lifestyle habits such as alcohol and tobacco use, and even the transfer of genital microbiomes post-

Weissella and the two Janus faces of the genus.

The genus Weissella belongs to the lactic acid bacteria group. It occurs naturally in foods and is a component of the human microbiome. A few Weissella species are candidate probiotics due to their potential for survival under the harsh conditions present in the gastrointestinal tract of humans and animals. Various species have also shown potential for treating and preventing periodontal disease, skin pathologies, and atopic dermatitis; some are used as starters for the fermentation of foods due to their production of exopolysaccharides; and others are used as protective cultures due to their

Probiotics and prebiotics in intestinal health and disease: from biology to the clinic.

Probiotics and prebiotics are microbiota-management tools for improving host health. They target gastrointestinal effects via the gut, although direct application to other sites such as the oral cavity, vaginal tract and skin is being explored. Here, we describe gut-derived effects in humans. In the past decade, research on the gut microbiome has rapidly accumulated and has been accompanied by increased interest in probiotics and prebiotics as a means to modulate the gut microbiota. Given the importance of these approaches for public health, it is timely to reiterate factual and supporting inf

An AI-augmented review of childhood atopic dermatitis biomarkers across genetic, immune, microbial, and metabolic domains.

BACKGROUND: Atopic dermatitis (AD) is a prevalent inflammatory skin disease and a major source of disease burden in children. Biomarker studies in childhood AD span genetic, immune, microbial and metabolic domains, but prior reviews have often focused on single molecular layers, specific sample sites or clinical classification. As a result, the field lacks an integrated, systems-level synthesis that compares and contextualizes biomarkers across domains while clearly distinguishing evidence strength. The rapid growth of literature in this field also poses practical challenges for traditional ma

Microbiome-based body site of origin classification of forensically relevant blood traces.

Human blood traces are amongst the most commonly encountered biological stains collected at crime scenes. Identifying the body site of origin of a forensic blood trace can provide crucial information in many cases, such as in sexual and violent assaults. However, means for reliably and accurately identifying from which body site a forensic blood trace originated are missing, but would be highly valuable in crime scene investigations. With this study, we introduce a taxonomy-independent deep neural network approach based on massively parallel microbiome sequencing, which delivers accurate body

Low-dose brain estrogen prevents menopausal syndrome while maintaining the diversity of the gut microbiomes in estrogen-deficient rats.

We evaluated the effects of intracerebroventricular administration (ICV) of brain estrogen and progesterone on menopausal symptoms and their effects on the secretion of follicle-stimulating hormone(FSH) and luteinizing hormone (LH) in estrogen-deficient rats. Three weeks after ovariectomy (OVX) or sham operation, OVX rats were given ICV infusions of either 17β-estradiol (4 μg/day; ICV-E), progesterone(0.8 μg/day; ICV-P), or vehicle (control) for 4 wk. OVX rats in the positive-control group were orally provided 150 μg 17β-estradiol·kg body wt-1·day-1. Sham rats had ICV vehicle infusion (normal-

PPARα deficiency causes skin dysbiosis and triggers innate immunity in keratinocytes.

Peroxisome proliferator-activated receptor-α (PPARα) is a transcription factor expressed in the skin, where its physiological role remains largely unknown. In this work, we found decreased bacterial density and dysbiosis associated with reduced amounts of Staphylococcus in the skin of PPARα-deficient mice compared to littermate controls. The NOD2 pathway was enhanced, as shown by up-regulation of both NOD2 and its downstream targets, including the antimicrobial peptides mBD3 and mBD4. Moreover, the Th17 immune response was enhanced in keratinocytes and epidermal dendritic T cells. Topical trea

MyD88 restricts dysbiosis-mediated inflammation in filaggrin deficient skin.

Atopic dermatitis (AD) is a common inflammatory skin disease associated with epidermal barrier dysfunction, immune dysregulation, and microbial dysbiosis. Loss-of-function mutations in filaggrin, a critical epidermal protein, represent the strongest genetic risk factor for AD and result in compromised skin barrier integrity and altered immune responses. MyD88 is an adaptor protein essential for TLR and IL-1 receptor signaling, with dual roles in promoting inflammation and regulating immune tolerance. However, the function of MyD88 in maintaining skin homeostasis in the context of filaggrin def

Strain-level genomic analysis of Staphylococcus epidermidis across multiple body sites in healthy females.

Staphylococcus epidermidis is a ubiquitous skin commensal that also colonizes the nasal and oral cavities, as well as the gastrointestinal and urinary tracts. Although prior studies have found genetic variation between S. epidermidis from infections and commensal strains, prior studies also have found that individual strains can inhabit multiple sites within an individual. None of these studies, however, have considered urogenital isolates. Here, we collected samples from 76 healthy female participants and performed whole-genome sequencing on 114 S. epidermidis isolates from the skin, nasal ca

Differential skin-bacteriome-mediated defense against chytridiomycosis in two neotropical frog species.

Symbiotic microbial communities have been implicated in host resistance to pathogens, but their effects are rarely demonstrated experimentally in wildlife. This study tested how skin-associated bacterial communities (i.e., bacteriomes) influence infection by the chytrid fungus Batrachochytrium dendrobatidis (Bd) in two tropical frog species, Haddadus binotatus and Ischnocnema henselii, which differ in susceptibility to Bd. Using a 2x2 factorial experimental design, frogs of both species were assigned to treatments crossing Bd exposure and antibiotic-mediated bacteriome suppression. In parallel

Rosacea in Older Adults and Pharmacologic Treatments.

Rosacea is a chronic inflammatory skin condition that is often more severe in older patients. The main clinical features are erythema, telangiectasia, and inflammatory lesions of the face. The pathogenesis of this condition is not fully understood but certainly multifaceted. Immune and inflammatory dysregulation, genetics, neurogenic dysregulation, microbiome dysbiosis, and systemic disease have all been implicated in rosacea pathogenesis. As we better understand the various pathways that lead to rosacea, we acknowledge that the different symptoms may have unique underlying triggers and mechan
