Use of patients' sera for immunoperoxidase demonstration of infectious agents in paraffin sections.

Using patients' sera diluted from 1:10 to 1:1,000 as the primary antibodies in indirect immunoperoxidase staining, the authors visualized a variety of infectious agents in formalin-fixed, paraffin-embedded tissue sections. The target lesions included 1) pyoderma caused by Staphylococcus aureus, 2) cryptococcal infection, 3) dermal sporotrichosis, 4) colon ulcer caused by amebic dysentery, 5) cutaneous leishmaniasis, and 6) chronic liver abscess containing ova of Ascaris lumbricoides. The infectious agents were clearly identified in the respective lesions. Paraffin sections of other kinds of in

Enhanced 5-aminolevulinic acid photodynamic therapy with potassium iodide for sporotrichosis: in vitro optimization and a proof-of-concept clinical case.

BACKGROUND: Sporotrichosis, a chronic fungal infection caused by pathogenic Sporothrix species, involves therapeutic challenges due to the toxicity of conventional antifungals. This study investigated the enhancement of 5-aminolevulinic acid-based photodynamic therapy (ALA-PDT) with potassium iodide (KI) through in vitro and a proof-of-concept clinical case. METHODS: In vitro testing on S. globosa conidia involved preincubation with 100 mM KI followed by ALA-PDT using 0.6 M ALA and 108 J/cm2 red light irradiation. The efficacy of this combined approach was systematically compared with that of…

Deep fungal co-infection with Trichosporon asahii and Candida parapsilosis in an immunocompetent individual: a case report and review of literature.

Deep fungal infections are endemic to tropical and subtropical regions and can occur in individuals who are both immunocompetent and immunocompromised. Clinically, these infections may present as verrucous plaques, subcutaneous nodules, abscesses or ulcerations. Common causative organisms in immunocompetent hosts include Sporothrix schenckii, Madurella mycetomatis and Fonsecaea pedrosoi. However, emerging evidence suggests that Trichosporon asahii and Candida parapsilosis are increasingly implicated in these infections. A 64-year-old Cambodian man with a history of recently treated pulmonary t…

Clinical Epidemiology of Sporotrichosis in Jilin Province, China (1990-2019): A Series of 4969 Cases.

INTRODUCTION: This study was aimed to examine the clinical and epidemiological characteristics of sporotrichosis in China and specifically J … otal of 241 strains were identified by molecular biotechnology; among these, 217 were identified as Sporothrix globosa and 24 were identified as S. schenckii sensu stricto. DISCUSSION: The results add clarity to the clinical epidemiology of sporotrichosis in China and specifically Jilin Province. We believe these data will help improve the epidemiology know …

Bone sporotrichosis: 41 cases from a reference hospital in Rio de Janeiro, Brazil.

BACKGROUND: Bone sporotrichosis is rare. The metropolitan region of Rio de Janeiro is hyperendemic for zoonotic sporotrichosis and the bone … the patients-amputations (7.3%) and ankylosis (4.9%), while 22% died in the course of the disease. Sporothrix brasiliensis was the causative agent in all the 9 (22%) performed cases. CONCLUSIONS: Bone sporotrichosis is a chronic, challenging condition with prolonged treatment, often with poor results and sequelae.

Imbalance of Th1, Th17, and Treg cells in peripheral blood of patients with lymphocutaneous sporotrichosis: a comparative study.

BACKGROUND: Sporotrichosis is an infection caused by the microscopic fungus, Sporothrix schenckii. The disease follows the traumatic inoculation of fungus through injuries involving soil, inhalation of conidia, or zoonotic transmission especially from cat scratches. OBJECTIVES: The objective of the retrospective cohort study was to investigate Th1, Th17, and Treg cell counts and host immunity in patients with lymphocutaneous sporotrichosis. MATERIALS AND METHODS: From January 2017 to December 2018, 88 patients, diagnosed with sporotrichosis, were retrospectively reviewed. The patients were d…

Sporotrichosis in sub-himalayan India.

Sporotrichosis is endemic in the Sub-Himalayan belt, which ranges from the northern to the north-eastern Indian subcontinent. Similar to many parts of the developing world, sporotrichosis is commonly recognized clinically in this region however consolidated epidemiological data is lacking. We report epidemiological, clinical and microbiological data from a hundred culture positive cases of sporotrichosis. Out of 305 clinically suspicious cases of sporotrichosis, a total of 100 isolates were identified as Sporothrix schenckii species complex (S. schenckii) on culture. Out of the culture proven …

Effect of anti-glycosphingolipid monoclonal antibodies in pathogenic fungal growth and differentiation. Characterization of monoclonal antibody MEST-3 directed to Manpalpha1-->3Manpalpha1-->2IPC.

Similar inhibitory results were observed when these fungi where incubated with a different mAb, which recognizes GIPCs bearing terminal residues of beta-D-galactofuranose linked to mannose (mAb MEST-1). Oral potassium iodide for the treatment of sporotrichosis.

BACKGROUND: Sporotrichosis is a subacute or chronic disease, usually affecting the skin caused by a dimorphic (existing in two forms), aerobic (oxygen requiring) fungus called Sporothrix schenckii. Oral potassium iodide is widely used for cutaneous sporotrichosis in clinical medicine with more and more reports published. However, the benefits and adverse reactions of these treatments have not yet been systematically reviewed. OBJECTIVES: To assess the effects of oral potassium iodide for the treatment of sporotrichosis. SEARCH STRATEGY: In July 2009 we searched the Cochrane Skin Group Specia…

Mode of action of itraconazole: morphological aspects.

The broad spectrum of antifungal activity of itraconazole is verified by morphologic criteria at the light and electron microscopical level. Yeast and fungal species examined are Candida albicans, Cryptococcus neoformans, Paracoccidioides brasiliensis, Sporothrix schenckii, Pityrosporum ovale, Trichophyton rubrum and Aspergillus fumigatus. Exposures of cultures of these yeasts and fungi to itraconazole results in dose- and time-dependent alterations which vary in nature and intensity from one species to another. The most striking gross morphological change is seen in the biphasic species and c…

An epidemic of sporotrichosis in Rio de Janeiro, Brazil: epidemiological aspects of a series of cases.

The first epidemic of sporotrichosis in humans as a result of zoonotic transmission was identified in Rio de Janeiro, Brazil, in 1998. A cross-sectional study was conducted applying questionnaires to patients seen in 2002 at Evandro Chagas Clinical Research Institute, Fiocruz, with a confirmed diagnosis of sporotrichosis. A total of 73 dwellings were studied, where 255 individuals, including 94 patients and 161 healthy household contacts, lived with 133 cats with sporotrichosis. Most dwellings were houses with 83% having complete basic sanitation. Among patients, there was a predominance of wo

A comparison of mycelial filtrate - and yeast lysate - paracoccidioidin in patients with paracoccidioidomycosis.

A cytoplasmic antigen was obtained from P. brasiliensis yeast cells by water lysis and sonication, with the aim of detecting delayed hypersensitivity to the fungus. This antigen was studied in patients with paracoccidioidomycosis (active and inactive), tuberculosis and histoplasmosis as well as in normal individuals, and it was compared with a mycelial filtrate antigen employed in the past for the same purpose. The yeast paracoccidioidin proved superior to the mycelial preparation (62.0 vs 24.1%) in patients with active paracoccidioidomycosis; however, in inactive cases, both preparations gave
