Activation of peripheral muscarinic receptors rescues depressive-like behaviors via gut-brain-axis, involving parasympathetic excitation.

Dysfunction of the autonomic nervous system (ANS) is strongly linked to the pathophysiological mechanisms of depression. Unfortunately, the correlation between depressive-like behaviors and the activation or inhibition of the sympathetic/parasympathetic nervous system has not been systematically explored. This study demonstrated that the pharmacological activation of sympathetic α-/β-adrenergic receptors increased stress susceptibility, whereas their antagonists enhanced stress resistance, thus suggesting a close correlation between sympathetic activity and depressive-like behaviors. Furthermo…

A new class of drugs for heart failure: SGLT2 inhibitors reduce sympathetic overactivity.

Even in the presence of excess glucose, the proximal renal tubules continue to resorb more glucose. Sodium glucose cotransporter 2 (SGLT2) i … ly understood. A common, and perhaps underappreciated, feature of T2DM is chronic activation of the sympathetic nervous system. This elevation of sympathetic activity contributes to the development of hypertension. It is also associated with a poor prognosis and with increased cardiovascular and renal morbidity/mortality independently of the effect on blood pressure. This r …

Involvement of renal sympathetic nerve overactivation in the progression of chronic kidney disease in rats.

Time-dependent changes in the renal sympathetic nerve activity (RSNA) in the progression of chronic kidney disease (CKD) have not been investigated, despite the fact that renal sympathetic nervous system is augmented in the condition of CKD. In the present study, we examined time-dependent changes in RSNA and renal venous norepinephrine concentrations for 12 weeks using 5 of 6 nephrectomized CKD rats. Both RSNA and norepinephrine concentrations were increased during the early phase in the progression of CKD. Urinary protein excretion and systolic blood pressure (SBP) were gradually increased d…

Exaggerated local hand sympathetic but not renin-angiotensin system activation in patients with primary Raynaud's phenomenon.

In Raynaud's disease (RD), an overactivity of sympathetic nervous system (SNS) was hypothesized but only indirect proofs were obtained. Complex interactions between the renin-angiotensin system (RAS) and SNS were reported without clear demonstrations of a RAS involvement. Recently, the use of ACE inhibitors and AT1 receptor antagonists in RD patients showed mixed results. The study of total and regional kinetics of tritiated noradrenaline (NA) and the measurements of local Angiotensin (Ang) I and II arterial-venous gradient were performed in 10 RD patients and 10 controls both in rest conditio…

Psychological factors in cardiac arrhythmias.

Autonomic nervous control of the heart and the blood vessels is essential for optimal physical performance. Afferent nerves conduct impulses to the vasomotor and cardioaccelerator and cardioinhibitor centres resulting in efferent sympathetic and vagus output. Information inputs also from the environment and the person's thought processes are mediated from the cortex by the hypothalamus, the limbic system, and the autonomic nervous system to the heart and blood vessels. Emotional factors like anger, anxiety, and fear increase sympathetic output and influence heart rate. On the other hand, fear

A Dynamic Architecture Linking Autonomic Activity to Emotional Dimensions: Real-Time Estimation From Photoplethysmography.

The present study introduces a novel, real-time emotion (or generalized behavioral) tracking model capable of independently estimating valence, motivation (approach-avoidance), and activation (arousal) through estimations of parasympathetic (PNS), α-adrenergic (α-SNS), and β-adrenergic (β-SNS) sympathetic nervous system activity. These separate autonomic systems correspond to distinct emotional dimensions: valence to the PNS, motivation (approach-avoidance) to the α-SNS, and activation to the β-SNS. This framework enables continuous, real-time, and interpretable estimation of emotion from a si…

Structural and function organization of intrathoracic extracardiac autonomic projections to the porcine heart: Implications for targeted neuromodulation therapy.

BACKGROUND: Mapping the structure/function organization of the cardiac nervous system is foundational for implementation of targeted neuromodulation-based therapeutics for the treatment of cardiac disease. OBJECTIVE: The purpose of this study was to define the spatial organization of intrathoracic parasympathetic and sympathetic efferent projections to the heart. METHODS: Yucatan mini-pigs (N = 11) were anesthetized and the thoracic cavity exposed. Electrical stimulation of the cervical vagi and stellate ganglia was performed individually, and hemodynamic responses were assessed in the intac

The Effect of Surgical Trauma in the Nasal Cavity on the Behavior in the Open Field and the Autonomic Nervous System of Rats.

In this article, the effect of surgical damage to the nasal mucosa in rats under the action of general anesthesia was investigated. The motor activity and heart rate variability (HRV) of rats were studied before and 2 days after the surgery. After surgical exposure, motor activity decreased, the time of fading and grooming increased, VLF increased, and HF decreased. Surgical trauma of the nasal septum in rats at the early postoperative stage promotes a pronounced stress response in the form of a sharp shift in the balance of the autonomic nervous system towards the sympathetic level and causes

Chemoreflexes, sleep apnea, and sympathetic dysregulation.

Obstructive sleep apnea (OSA) and hypertension are closely linked conditions. Disordered breathing events in OSA are characterized by increasing efforts against an occluded airway while asleep, resulting in a marked sympathetic response. This is predominantly due to hypoxemia activating the chemoreflexes, resulting in reflex increases in sympathetic neural outflow. In addition, apnea - and the consequent lack of inhibition of the sympathetic system that occurs with lung inflation during normal breathing - potentiates central sympathetic outflow. Sympathetic activation persists into the daytime

The role and management of sympathetic overactivity in cardiovascular and renal complications of diabetes.

Feedback activation of neurohormonal pathways in the setting of kidney or heart failure contributes to the development and progression of dysfunction in the other. Diabetes and its management independently activate these same pathogenic pathways, feeding into this vicious cycle and contributing to a poor prognosis. One of the most important of these neurohormonal pathways is the sympathetic nervous system (SNS). The activity of the SNS in increased in patients with chronic kidney disease, even in the absence of renal impairment or heart failure. There is a strong relationship between SNS overa…

New aspects in the pathogenesis of abomasal displacement.

Impaired abomasal motility and an increased accumulation of gas are prerequisites for displacement of the abomasum in the cow. Predisposing factors are the breed (e.g. Holstein-Friesian, Simmental-Red-Holstein cross breeds and Guernsey), genetic background, twin pregnancy, first weeks of lactation, metabolic disorders (ketosis, increased lipomobilisation, insulin resistance), high-concentrate and low-fibre diets, as well as other concomitant diseases, such as endometritis, mastitis and claw disorders. There does not appear to be a strong correlation between increased milk yield or endotoxaemia

Interpreting oscillations of muscle sympathetic nerve activity and heart rate variability.

Computer analysis of spontaneous cardiovascular beat-by-beat variability has gained wide credibility as a means of inferring disturbances of autonomic cardiovascular regulation in a variety of cardiovascular conditions, including hypertension, myocardial infarction and heart failure. Recent applications of spectral analysis to muscle sympathetic nerve activity (MSNA) offer a new approach to a better understanding of the relationship between cardiovascular oscillations and autonomic regulation. However, areas of uncertainty and unresolved debates remain, mostly concerning different methodologie

[Role of ciliary muscle in ocular physiology and disease].

The ciliary muscle is the largest intraocular muscle. It is characterized by a complex structure, rich blood supply, and high activity, particularly in the daytime, when visual work is performed, and even during night sleep. The activity of the ciliary muscle involves the adjacent structures. In fact, it continuously massages the anterior and median segments of the eye. Presbiopia leads to a decrease in the activity of the ciliary muscle, deteriorates blood circulation in it, and accelerates age-associated involution processes. Presbiopia is believed to be a risk factor for glaucoma, cataract,

[Sudden loss of consciousness: clinical presentation and pathophysiologic mechanisms].

Most cases of sudden and temporary loss of consciousness [syncope] are caused by hypoperfusion of the formatio reticularis. more rarely by primary neurologic or metabolic disorders. The most common etiology is vasodepressor (vasovagal) syncope, which is caused by peripheral vasodilation due to acute withdrawal of the efferent sympathetic tone, while the parasympathetic outpour is increased at the same time. Although the efferent limb of the baroreflex manifests in a rather uniform way, the afferent parasympathetic limb is very variable, leading to a variety of clinical presentations and trigge

Selective effect of sufentanil on group III (A delta) and group IV (C) somatosympathetic reflexes.

The effect of sufentanil on somatosympathetic reflexes and the subsequent reversal of its effects by naloxone have been observed in dogs anaesthetised with alpha-chloralose, paralysed with suxamethonium and artificially ventilated. When the drug was infused at a rate of 2.5 micrograms kg-1 min-1 the late long latency response evoked by Group IV (C) fibres was totally abolished at a mean dose of 6.40 micrograms kg-1 (+/- 0.99 microgram kg-1) (s.e. mean), while retaining a substantial part of the early short latency response evoked by group III (A delta) fibres; subsequently, during the infusion

But some rare cases of mixed facial unilateral algias associating both mechanisms are possible. Here is studied a series of 12 cases. One may distinguish: 1. one neuralgic vascular type. It starts by lightning and triggered hemifacial pain followed by tenderness and vaso-secretory phenomenous of same topography; 2. the second is of vascular-neuralgic type. It starts like common cluster headache; the lightning pain occuring in the end of paroxysm. Only the first type -- neuralgic vas
