Randomized controlled trial of neurofeedback on chemotherapy-induced peripheral neuropathy: A pilot study.

BACKGROUND: Chemotherapy-induced peripheral neuropathy (CIPN) is a significant problem for cancer patients, and there are limited treatment options for this often debilitating condition. Neuromodulatory interventions could be a novel modality for patients trying to manage CIPN symptoms; however, they are not yet the standard of care. This study examined whether electroencephalogram (EEG) neurofeedback (NFB) could alleviate CIPN symptoms in survivors. METHODS: This was a randomized controlled trial with survivors assigned to an NFB group or a wait-list control (WLC) group. The NFB group underw

Effect of Tailored Dose-Dense Chemotherapy vs Standard 3-Weekly Adjuvant Chemotherapy on Recurrence-Free Survival Among Women With High-Risk Early Breast Cancer: A Randomized Clinical Trial.

IMPORTANCE: Standard dosing of chemotherapy based on body surface area results in marked interpatient variation in pharmacokinetics, toxic effects, and efficacy. Whether tailored dosing can improve outcomes is unknown, as is the role of dose-dense adjuvant chemotherapy. OBJECTIVE: To determine whether tailored dose-dense adjuvant chemotherapy improves the outcomes of early breast cancer compared with a standard 3-weekly chemotherapy schedule. DESIGN, SETTING, AND PARTICIPANTS: A randomized, open-label, phase 3 trial of women aged 65 years and younger who had surgery for nonmetastatic node-po

Taxane-Induced Peripheral Neuropathy-Driven Treatment Modification and Pathologic Complete Response After Neoadjuvant TCHP in HER2-Positive Breast Cancer: A Retrospective Cohort Study.

BACKGROUND: Neoadjuvant docetaxel/carboplatin/trastuzumab/pertuzumab (TCHP) is a widely used regimen for human epidermal growth factor receptor 2 (HER2)-positive breast cancer, achieving high rates of pathologic complete response (pCR). Taxane-induced peripheral neuropathy (CIPN) represents a clinically relevant dose-limiting toxicity that may lead to premature taxane discontinuation. Evidence guiding the optimal neoadjuvant strategy after CIPN onset remains limited in real-world, toxicity-driven treatment modification scenarios. PATIENTS AND METHODS: We conducted a retrospective cohort study

Disulfide-Crosslinked mPEG-PLA-(LA)4 Nanomicelles for Taxane Delivery in Breast Cancer Therapy.

Taxanes represent a crucial class of chemotherapeutic agents clinically employed for the treatment of various malignancies, including breast cancer. Three commonly utilized taxanes-paclitaxel (Taxol), docetaxel (Taxotere), and cabazitaxel-are substantially limited by inherent drawbacks such as poor aqueous solubility, rapid clearance, and non-specific distribution. To enhance their anti-tumor efficacy against breast cancer, an mPEG-PLA-(LA)4 nanomicelle carrier was designed and synthesized in this study for the loading of these three taxanes. These nanomicelles were designed to improve the aqu

Predictive factors for failure of neoadjuvant docetaxel, cisplatin, and 5-fluorouracil therapy in esophageal squamous cell carcinoma.

Recently, neoadjuvant chemotherapy comprising docetaxel, cisplatin, and 5-fluorouracil showed promising efficacy for esophageal squamous cell carcinoma. However, some patients do not achieve curative resection despite neoadjuvant chemotherapy using these drugs. This study aimed to clarify the pretherapeutic characteristics of these patients. We included 113 patients who underwent neoadjuvant chemotherapy with docetaxel, cisplatin, and 5-fluorouracil for potentially resectable esophageal squamous cell carcinoma and compared the clinical characteristics between patients who achieved curative res

Amphiphilic albumin-based nanoparticles designed for the efficient delivery of taxanes.

In cancer chemotherapy, taxanes such as Paclitaxel (PTX), Docetaxel (DTX), and Cabazitaxel (CTX) play a pivotal role but often require surfactants and solvents for solubilization, which cause adverse effects. Plain human serum albumin (HSA) nanoparticles, such as Abraxane, utilize nab technology, which is not universally applicable to all taxanes, limiting its applicability. To address this limitation, this study introduces a novel HSA-PLA nanoparticle platform, designed to universally enhance the delivery of taxanes. This nanoplatform utilizes a thiol-maleimide reaction to form core-shell str

Docetaxel, cyclophosphamide, and epirubicin: application of PBPK modeling to gain new insights for drug-drug interactions.

The new adjuvant chemotherapy of docetaxel, epirubicin, and cyclophosphamide has been recommended for treating breast cancer. It is necessary to investigate the potential drug-drug Interactions (DDIs) since they have a narrow therapeutic window in which slight differences in exposure might result in significant differences in treatment efficacy and tolerability. To guide clinical rational drug use, this study aimed to evaluate the DDI potentials of docetaxel, cyclophosphamide, and epirubicin in cancer patients using physiologically based pharmacokinetic (PBPK) models. The GastroPlus™ was used

Synthesis, in vitro characterization, and anti-tumor effects of novel polystyrene-poly(amide-ether-ester-imide) co-polymeric micelles for delivery of docetaxel in breast cancer in Balb/C mice.

OBJECTIVE: The goal of the present work was to make novel co-polymeric micellar carriers for the delivery of docetaxel (DTX). SIGNIFICANCE: Co-polymeric micelles can not only solubilize DTX and eliminate the need for toxic surfactants to dissolve it, but also cause passive targeting of the drug to the tumor and reduce its toxic side effects. METHODS: Poly(styrene-maleic acid) (SMA) was conjugated to poly (amide-ether-ester-imide)-poly ethylene glycol (PAEEI-PEG). Copolymer synthesis was proven by Fourier transform infrared (FTIR) and 1H-nuclear magnetic resonance (1H-NMR). The SMA-PAEEI-PEG

Uterine leiomyosarcoma: Epidemiology, contemporary treatment strategies and the impact of uterine morcellation.

Leiomyosarcoma, a rare tumor subtype, accounts for 1% of all uterine malignancies, but contributes to a significant proportion of uterine cancer deaths. Surgery is considered the mainstay of treatment for all soft tissue sarcomas, including uterine variants. However, uterine leiomyosarcoma is challenging to diagnose preoperatively and can mimic the appearance of benign uterine leiomyomas. Recently, concerns have grown in this regard, as surgeons have utilized uterine morcellation and myomectomy procedures unknowingly in the setting of occult uterine sarcoma. Because of aggressive tumor biology
