The epidemiology of bovine trypanosomiasis on Galana Ranch, Kenya was studied for one year (July 1980--June 1981), using measurements from an experimental population of 3 types of Boran cattle. The results were applied to the estimated ranch population at risk, and an attempt was made to measure the potential economic loss due to trypanosomiasis. The potential losses in beef production due to trypanosomiasis in the ranch population at risk at Galana were estimated at around K.Shs. 8900/km2, when the stocking rate was 14.2 Tropical Livestock Units per km2. The estimated potential loss in the to…

A search for Glossina fuscipes fuscipes puparia near homesteads in the sleeping sickness focus of Busoga revealed puparia and puparial shells under Coffea canephora (coffee), Musa sp. (banana) and Lantana camara thickets as well as under house verandahs and, once, inside a hut. This is the first description of G.f. fuscipes breeding sites in a peridomestic habitat. The implications of these findings in relation to the transmission of the current epidemic of sleeping sickness in Busoga is discussed.

This paper gives the results of a tsetse fly research program in the preforested area of Ivory Coast. The main investigation tool was the Challier-Laveissière's biconical trap. The ecodistribution, resting and pupal sites of the principal tsetse species, G. palpalis, G. pallicera and G. nigrofusca were described in the T. b. gambiense sleeping sickness focus of Vavoua. Population dynamics, host preferences, activity cycles were compared between tsetse populations in plantations and village surroundings. Man-fly contact was of high intensity where human habitat is scattered in plantations but i

Parasitological examinations of the cerebrospinal fluid of 20 vervet monkeys (Cercopithecus aethiops), that had been infected with Trypanosoma rhodesiense, revealed that the CSF was regularly infested with trypanosomes in the early phase of the disease, at the earliest on the 13th day, in most of the animals in the 3rd or 4th week, after infection. Follow-up examinations of the CSF during the further course of the disease also regularly proved positive for trypanosomes. Histological studies in the animals that died at a mean of 65 days after infection (range 35 to 107 days) revealed encephalit

African trypanosomiasis in humans is caused by trypanosomes, parasitic protozoa that inhabit the blood and tissue spaces. Humans are infected by the bite of the tsetse (Glossina species), which itself acquires the parasites from human or animal hosts. The vector and the disease are confined to sub-saharan Africa where the disease occurs in endemic foci from which epidemics arise. Two forms of African trypanosomiasis in humans are recognized and both are fatal if left untreated. Control strategy is dependent on the capacity of the public health services to diagnose and treat cases, and the main…

Complement-fixing antibody titres decline after successful treatment of trypanosoma infected cattle during a fixed period. For investigating the practicability of the resulting double test procedure in the field, cattle under controlled drug regimes were tested at treatment and again 3 weeks later. Alternatively, the results from groups of treated and untreated animals from the same area were compared. Both serological and parasitological test results indicated an insuffieicnt effectiveness of therapy in the area under investibation.

Field trials in West Africa have shown the value of the estimation of serum immunoglobulin M (IgM) levels as a screening test in endemic trypanosomiasis areas. A further field trial carried out in the Democratic Republic of the Congo is described. The technique used, based on double diffusion on an agar plate, gave consistent results in skilled hands, but the standard required is probably too high for the wide application of the method under normal field conditions. The diameter of the precipitation zones for 200 sera from new patients from a survey population of nearly 10 000 was generally be…

BACKGROUND: Human African trypanosomiasis (HAT) is a neglected tropical disease that usually occurs in rural areas in sub-Saharan Africa. It caused devastating epidemics during the 20th century. Sustained, coordinated efforts by different stakeholders working with national sleeping sickness control programmes (NSSCPs) succeeded in controlling the disease and reducing the number of cases to historically low levels. In 2012, WHO targeted the elimination of the disease as a public health problem by 2020. This goal has been reached and a new ambitious target was stated in the WHO road map for NTDs…

We designed and synthesized a series of symmetric bis-6-amidino-benzothiazole derivatives with aliphatic central units and evaluated their efficacy against bloodstream forms of the African trypanosome Trypanosoma brucei. Of these, a dicationic benzothiazole compound (9a) exhibited sub-nanomolar in vitro potency with remarkable selectivity over mammalian cells (>26,000-fold). Unsubstituted 5-amidine groups and a cyclohexyl spacer were the crucial determinants of trypanocidal activity. In all cases, mice treated with a single dose of 20 mg kg-1 were cured of stage 1 trypanosomiasis. The compound…

BACKGROUND: Human African trypanosomiasis or sleeping sickness, caused by the parasite Trypanosoma brucei, leads to neuroinflammation and characteristic sleep/wake alterations. The relationship between the onset of these alterations and the development of neuroinflammation is of high translational relevance, but remains unclear. This study investigates the expression of interferon (IFN)-γ and IFN-inducible chemokine genes in the brain, and the levels of CXCL10 in the serum and cerebrospinal fluid prior to and during the encephalitic stage of trypanosome infection, and correlates these with sle…

In 2001, the World Health Organization (WHO) established a public-private partnership to fight human African trypanosomiasis (HAT). As a result of this continuous collaboration, and in addition to the coordination with nongovernmental organizations and bilateral cooperation agencies, the number of new cases of HAT annually reported by the WHO has strikingly decreased. In 2012, HAT was included in WHO's roadmap on neglected tropical diseases with a 2020 target date for elimination. Although the prevalence of HAT is decreasing and its elimination is targeted, control approaches must be adapted t…

A series of quinoline-3-carboxylic acid derivatives has been synthesized with easy reaction conditions (in aqueous micellar microreactors and using microwave irradiation) and tested for its biological activity against human African trypanosomiasis (HAT). From this series, several derivatives of quinoline-3-carboxylic acid such as 23 and 24 exhibited potent activity against Trypanosoma brucei in vitro and had low cytotoxicity. The combination of electron withdrawing and donating groups in the structure seems to be the crucial factor to inhibit T. brucei.
