Dynamical modulation of hypersynchronous seizure onset with transcranial magneto-acoustic stimulation in a hippocampal computational model.

Hypersynchronous (HYP) seizure onset is one of the frequently observed seizure-onset patterns in temporal lobe epileptic animals and patients, often accompanied by hippocampal sclerosis. However, the exact mechanisms and ion dynamics of the transition to HYP seizures remain unclear. Transcranial magneto-acoustic stimulation (TMAS) has recently been proposed as a novel non-invasive brain therapy method to modulate neurological disorders. Therefore, we propose a biophysical computational hippocampal network model to explore the evolution of HYP seizure caused by changes in crucial physiological …

Acoustic trauma increases inhibitory effects of amygdala electrical stimulation on thalamic neurons in a rat model.

Acoustic trauma (AT) induced hearing loss elicits plasticity throughout the central auditory pathway, including at the level of the medial geniculate nucleus (MGN). Hearing loss also results in altered neuronal responses in the amygdala, which is involved in sensory gating at the level of the MGN. However, whether these altered responses in the amygdala affect sensory gating at the level of the MGN requires further evaluation. The current study aimed to investigate the effects of AT-induced hearing loss on the functional connectivity between the amygdala and the MGN. Male Sprague-Dawley rats w

Evidence for a bipolar change in distortion product otoacoustic emissions during contralateral acoustic stimulation in humans.

The aim of this study was to investigate the activity of the medial olivocochlear (MOC) efferents during contralateral (CAS) and ipsilateral acoustic stimulation (IAS) by recording distortion product otoacoustic emission (DPOAE) suppression and DPOAE adaptation in humans. The main question was: do large bipolar changes in DPOAE level (transition from enhancement to suppression) also occur in humans when changing the primary tone level within a small range as described by Maison and Liberman for guinea pigs [J. Neurosci. 20, 4701-4707 (2000)]? In the present study, large bipolar changes in DPOA…

The effect of the acoustic nerve chronic electric stimulation upon the guinea pig cochlear nucleus development.

14 newborn guinea pigs have been deafened three days after birth by bilateral destruction of the organ of Corti, and among them 8 have been supplied with a chronic auditory electric stimulation by means of an intracochlear implanted electrode. The histological study and the anatomical reconstructions of these animals' cochlear nuclei demonstrate that this chronic stimulation prevents, at least partially, these auditory pathway medullary formations from atrophy due to the cochlear destruction.

Effect on vestibular function of cochlear implantation by partial deafness treatment-electro acoustic stimulation (PDT-EAS).

PURPOSE: Although the cochlear implantation procedure does not interfere with vestibular structures directly, both the vestibulum and the cochlea share the same inner ear fluid space, and this fluid may be responsible for transferring possibly damaging forces from one to the other. The purpose of the study is to assess postoperative vestibular function after partial deafness treatment-electro-acoustic stimulation (PDT-EAS) cochlear implantation. METHODS: Fifty-five patients were included in the study (30 females, 25 males, age 11-80, mean 41.8 ± 19.35). cVEMP and oVEMP were performed preopera…

Using Neural Response Telemetry to Monitor Physiological Responses to Acoustic Stimulation in Hybrid Cochlear Implant Users.

OBJECTIVE: This report describes the results of a series of experiments where we use the neural response telemetry (NRT) system of the Nucleus cochlear implant (CI) to measure the response of the peripheral auditory system to acoustic stimulation in Nucleus Hybrid CI users. The objectives of this study were to determine whether they could separate responses from hair cells and neurons and to evaluate the stability of these measures over time. DESIGN: Forty-four CI users participated. They all had residual acoustic hearing and used a Nucleus Hybrid S8, S12, or L24 CI or the standard lateral wa…

Effect of contralateral acoustic stimulation on cochlear tuning measured using stimulus frequency and distortion product OAEs.

OBJECTIVE: To study whether a change in cochlear tuning, measured using OAEs, could be detected due to contralateral activation of the efferent system using broadband noise. DESIGN: Cochlear tuning measures based on SFOAE phase gradients and SFOAE-2TS 'Q' were used to test this hypothesis. SFOAE magnitude and phase gradient were measured using a pure-tone sweep from 1248 to 2496 Hz at 50 dB SPL. 2TS curves of SFOAE were recorded with a suppressor frequency swept from 1120 to 2080 Hz at 50 dB SPL. DPOAE f2-sweep phase gradient was also obtained to allow comparisons with the literature. All thr

Effectiveness of intermittent and continuous acoustic stimulation in preventing noise-induced hearing and hair cell loss.

Resistance to noise-induced hearing loss (NIHL) was studied in gerbils exposed either to intermittent or continuous low-level noise prior to an intense noise. Auditory-evoked brainstem response (ABR) thresholds, distortion product otoacoustic emissions (DPOAEs), Q10dB values from compound action potential (CAP) tuning curves, and outer hair cell (OHC) loss were measured for each group. Subjects were exposed to A-weighted noise (octave band noise centered at 2 kHz) on an intermittent (80 dB, 6 h/day) or continuous schedule (74 dB, 24 h/day) for 10 days, allowed to rest in quiet for 2 days, then

Effects of contralateral white noise stimulation on transitory evoked otoacoustic emissions in patients with acoustic neuroma.

Transitory evoked otoacoustic emissions are normal phenomena observed in most persons with hearing levels greater than 35 dB. Further, masking of the contralateral ear produces amplitude reductions in the transitory evoked otoacoustic emissions. We have undertaken a study of transitory evoked otoacoustic emissions in 20 patients with acoustic neuroma. All patients were assessed for transitory evoked otoacoustic emissions bilaterally, with and without contralateral masking with white band noise at 40, 50, and 60 dB. We found that transitory evoked otoacoustic emissions were present in 30% of ea

Selective Auditory Attention Engages Distinct Neural Strategies in Simulated Electric and Acoustic Stimulation Within and Across Ears.

OBJECTIVES: This study aimed to investigate whether electric-acoustic stimulation (EAS) and bimodal stimulation (BS) rely on similar or distinct neural mechanisms to support attention-driven speech comprehension in noisy environments. DESIGN: This cross-sectional study used electroencephalography to measure cortical multidimensional neural responses and compared them between two groups of young adult normal-hearing listeners (15 subjects per group) exposed to simulated EAS or BS listening conditions in a two-talker competing scene. The simulated stimuli were generated using a vocoder to mimic…

Transient acoustic stimulation induces time-dependent synaptic remodeling and enhancement of auditory nerve output after threshold recovery.

BACKGROUND: Acoustic stress can alter cochlear function even in the absence of permanent threshold elevation; however, synaptic consequences of transient acoustic stimulation remain incompletely understood. OBJECTIVE: This study aimed to investigate whether transient acoustic stimulation induces changes in the auditory nerve output and cochlear ribbon synapse morphology following hearing threshold recovery. METHODS: Young adult CBA/CaJ mice were exposed to band-limited acoustic stimulation (45-2,000 Hz, 95 dB SPL, 2 h). Auditory brainstem responses (ABRs), hair cell and spiral ganglion neuro…

Combined Acoustic-Acupuncture Stimulation Demonstrates Efficacy in Primary Tinnitus Rehabilitation: Objective Evidence From EEG Brain Network Analysis.

Tinnitus is a prevalent neurological disorder that significantly impacts patients' quality of life and is difficult to treat. While acupuncture therapy and acoustic stimulation have individually shown therapeutic potential, their combined application remains largely unexplored. The aim of this study was to investigate the efficacy and neural mechanisms of combination therapy based on brain network technology. In this tinnitus intervention study, 240 patients were enrolled, randomly divided into three groups for three treatment schemes: acoustic stimulation therapy, acupuncture stimulation ther…

Revealing Pathological Auditory Central Inhibition in Tinnitus Using Cortical Auditory Evoked Potentials Responses to Contralateral Acoustic Stimulation.

OBJECTIVE: To test the hypothesis that auditory central inhibition is reduced in tinnitus patients and explore whether improving this inhibitory function could alleviate tinnitus severity. METHODS: We recruited 16 chronic tinnitus patients and 14 age-matched healthy controls, all of whom exhibited clinically normal audiometric thresholds. Electroacoustic measures, cortical auditory evoked potentials (CAEP), were used to assess neural activity. Contralateral noise stimulation was employed to evaluate contralateral suppression (CS) of CAEP amplitude. RESULTS: Significant differences in CS of N

Heterogeneity of MYO15A variants significantly determine the feasibility of acoustic stimulation with hearing aid and cochlear implant.

Autosomal recessive nonsyndromic hearing loss 3 (DFNB3) mainly leads to congenital and severe-to-profound hearing impairment, which is caused by variants in MYO15A. However, audiological heterogeneity in patients with DFNB3 hinders precision medicine in hearing rehabilitation. Here, we aimed to elucidate the heterogeneity of the auditory phenotypes of MYO15A variants according to the affected domain and the feasibilities for acoustic stimulation. We conducted whole-exome sequencing for 10 unrelated individuals from seven multiplex families with DFNB3; 11 MYO15A variants, including the novel fr…

Stronger efferent suppression of cochlear neural potentials by contralateral acoustic stimulation in awake than in anesthetized chinchilla.

There are two types of sensory cells in the mammalian cochlea, inner hair cells, which make synaptic contact with auditory-nerve afferent fibers, and outer hair cells that are innervated by crossed and uncrossed medial olivocochlear (MOC) efferent fibers. Contralateral acoustic stimulation activates the uncrossed efferent MOC fibers reducing cochlear neural responses, thus modifying the input to the central auditory system. The chinchilla, among all studied mammals, displays the lowest percentage of uncrossed MOC fibers raising questions about the strength and frequency distribution of the con…

Electrically evoked auditory brainstem response by direct electrical stimulation to the cochlear nerve in acoustic neuroma patients.

The electrically evoked auditory brainstem response (EABR) was recorded by direct electrical stimulation of the cochlear nerve in 8 acoustic neuroma patients. The EABR by electrical stimulation of the cochlear nerve at the fundus of the internal auditory canal was almost similar to those previously reported from cochlear implant patients in waveform morphology and wave latencies. However, the wave corresponding to wave II in human auditory ABR was not recorded in the EABRs by stimulation of the distal part of the intracranial cochlear nerve. These results suggest that the cochlear nucleus does
