Molecular dosimetry of urinary aflatoxin-N7-guanine and serum aflatoxin-albumin adducts predicts chemoprotection by 1,2-dithiole-3-thione in rats.

Hepatocellular carcinoma has one of the poorest 5 year survival rates of any human cancer. Preventive measures offer the best possibility of ameliorating this disease and chemoprotective agents are being developed for this purpose. The dithiolethiones, including oltipraz and the unsubstituted molecule 1,2-dithiole-3-thione, have been shown to be potent inhibitors of aflatoxin-induced hepatic tumorigenesis in rats. However, subsequent evaluation of dithiolethiones or other chemoprotective agents in human clinical trials will require the development of intermediate, non-invasive biomarkers to ev

Cobalt-Doped CeO2 Nanozyme with Peroxidase-Like Activity for Broad-Spectrum and Recyclable Degradation of Aflatoxins.

Aflatoxins (AFs) contamination in the food chain poses a serious health threat, and current degradation strategies are limited by insufficient efficiency and stability. We developed a series of cobalt-doped CeO2 (Co@CeO2) nanozymes via a one-step hydrothermal method. The optimized nanozyme (Co-to-CeO2 mass ratio of 2:1) exhibited remarkable peroxidase-like activity, enabling efficient broad-spectrum degradation of aflatoxin B1, G1, and M1 under mild conditions, with removal efficiencies exceeding 90% in aqueous solution and approximately 80% in spiked edible oil samples. Density functional the…

Imidazolium polymerized ionic liquid-based membranes for the early warning detection of aflatoxins in fermented foods.

This study uses porous ionic liquid gel membranes (PILs) for early-warning detection of aflatoxins (AFTs) in fermented foods. We prepared a stable, cross-linked membrane by embedding imidazolium-based ILs into hydroxyethyl cellulose‑sodium polyacrylate. Its mild, tunable adsorption captures unstable AFT precursors and enables accurate, nondestructive quantification in complex food matrices. [AVIM]Br membranes selectively bind coumarin-type AFTs via amine groups (91.7-99.3% recovery); [BVIM]Br membranes bind anthraquinone-type precursors via phenylboronic acid (73.1-99.4% recovery). Coupled wit…

Detection of Total Aflatoxins in Herbal Medicines Based on Lateral Flow Assay with Contamination Ratio Model.

In this study, we developed a colloidal gold immunochromatographic strip (CGIS) method that used the matrix-matched calibration curves of contamination ratio models to quantitatively determine the total aflatoxin in five herbal medicines. This approach addresses issues related to false results and poor accuracy associated with conventional methods. The CGIS was analyzed using a Vertu touch reader, and the matrix-matched calibration was established based on the absorbance ratios of the T and C lines, as well as the logarithmic values of the total aflatoxin concentrations. The total aflatoxins c…

Presence of unreported carcinogens, Aflatoxins and their hydroxylated metabolites, in industrialized Oaxaca cheese from Mexico City.

Aflatoxins (AFs) are toxic secondary metabolites of the fungi Aspergillus flavus, A. parasiticus and A. nomius. The fungi produce these AFs in cereals, oilseeds and spices. AFs have damaging effects on all organisms, including humans, and their symptoms can be classified as acute (vomiting, hemorrhage and death) or chronic (immunodepression, Reye syndrome, Kwashiorkor, teratogenesis, hepatitis, cirrhosis, and various cancers). Basic AFs (AFB1, AFB2, AFG1, and AFG2) are metabolized in the liver or by microbes that produce hydroxylated metabolites (AFM1, AFM2, and AFP1) and aflatoxicol (AFL), so…

Identification and quantification of aflatoxins and aflatoxicol from poultry feed and their recovery in poultry litter.

Aflatoxins (AF) are toxic fungal secondary metabolites and are known mycotoxins pathological to animals and humans. Poultry litter is frequently used as a food supplement for ruminants, and when poultry feed contains AF, the litter becomes contaminated as well, thus having an effect on livestock health. This study identified and quantified AF (AFB(1), AFB(2), AFG(1), and AFG(2)) from poultry feed and their recovery, together with their metabolites (AFM(1), AFM(2), AFP(1), and aflatoxicol) in litter. An experiment with 25 Hy-Line W-36 hens, in their second production stage, 121 wk old, was carr…

Microalgae: A promising strategy for aflatoxin control in poultry feeds.

Aflatoxins are toxic compounds produced by certain molds, primarily Aspergillus species, which can contaminate crops such as grains and nuts. These toxins pose a significant health risk to animals and humans. Aflatoxin B1 (AFB1) is the most potent of these compounds and has been well-characterized to lead to diminished growth and feed efficiency by disrupting nutrient absorption and metabolism in poultry. AFB1 can trigger apoptosis and inflammation, leading to a decline in immune function and changes in blood biochemistry in poultry. Recently, there has been growing interest in using microalga…

Pathogen producing aflatoxin in contaminated sandwich: Identification and preservation.

The toxicity of the contaminated powder contributed to toxic aflatoxins has been well-known in the literature. However, before this study, the specific fungal strain behind aflatoxin production remained unidentified. Our research aimed to isolate and identify fungi from the tainted sandwiches while also assessing the preservation of sandwiches in ambient conditions. The study pinpointed Aspergillus flavus as the fungus responsible for aflatoxin production. Analysis revealed that the sandwich samples contaminated with pure A. flavus exhibited a significant Aflatoxin B1 (AFB1) concentration of 5…

65 Years on-Aflatoxin Biomarkers Blossoming: Whither Next?

Aflatoxins are mycotoxins produced by Aspergillus flavus and several other related organisms and are common contaminants of numerous grains and nuts, especially maize (corn) and peanuts. Although, undoubtedly, aflatoxins have been present in the food of humans for millennia, their toxic effects were not discovered until 1960, first becoming evident as a non-infectious outbreak of poisoning of turkeys (Turkey X disease) arising from contaminated groundnut meal. The elucidation of specific chemical structures in 1963 led to the rapid characterization of aflatoxins as among the most potent chemic…

Impedimetric nanoimmunosensor platform for aflatoxin B1 detection in peanuts.

This article developed a novel electrochemical immunosensor for the specific detection of aflatoxin B1 (AFB1). Amino-functionalized iron oxide nanoparticles (Fe3 O4 -NH2 ) were synthesized. Fe3 O4 -NH2 were chemically bound on self-assembly monolayers (SAMs) of mercaptobenzoic acid (MBA). Finally, polyclonal antibodies (pAb) were immobilized on Fe3 O4 -NH2 -MBA. The sensor system was evaluated through atomic force microscopy (AFM), cyclic voltammetry (CV), and electrochemical impedance spectroscopy (EIS). A reduction in the anodic and cathodic peak currents was observed after the assembly of t

Aflatoxin B1 impairs sperm quality and fertilization competence.

Aflatoxins are poisonous byproducts of the soilborne fungus Aspergillus, involved in the decomposition of plant materials. Aflatoxins can be found in various food products, such as maize, sorghum, millet, rice and wheat. AFB1 is the most toxic of these, classified as a carcinogen and mutagen for both humans and animals. AFB1 has been detected in human cord blood and placenta; however, its toxic effect on sperm is less known. The current study examines sperm responses associated with AFB1 exposure. These included acrosome integrity and function, mitochondrial polarity, DNA fragmentation, fertil…

Aflatoxin exposure in Nigerian children with severe acute malnutrition.

Aflatoxin exposure is an important public health concern in sub-Saharan Africa as well as parts of Latin America and Asia. In addition to hepatocellular carcinoma, chronic aflatoxin exposure is believed to play a role in childhood growth impairment. The most reliable biomarker of chronic aflatoxin exposure is the aflatoxin-albumin adduct, as measured by ELISA or isotope dilution mass spectrometry (IDMS). In this report, we have used high resolution LC-MS/MS with IDMS to quantitate AFB1-lysine in an extremely vulnerable population of Nigerian children suffering from severe acute malnutrition. T

Effects of Zinc Chelators on Aflatoxin Production in Aspergillus parasiticus.

Zinc concentrations strongly influence aflatoxin accumulation in laboratory media and in food and feed crops. The presence of zinc stimulates aflatoxin production, and the absence of zinc impedes toxin production. Initial studies that suggested a link between zinc and aflatoxin biosynthesis were presented in the 1970s. In the present study, we utilized two zinc chelators, N,N,N',N'-tetrakis (2-pyridylmethyl) ethane-1,2-diamine (TPEN) and 2,3-dimercapto-1-propanesulfonic acid (DMPS) to explore the effect of zinc limitation on aflatoxin synthesis in Aspergillus parasiticus. TPEN but not DMPS dec

Negligible effects of tryptophan on the aflatoxin adsorption of sodium bentonite.

The main objective of this study was to determine if the competitive adsorption of tryptophan (Trp) and aflatoxin B₁ (AFB₁) could potentially affect the ability of a sodium bentonite (NaB) to prevent aflatoxicosis in monogastric animals. The adsorption of Trp and AFB₁ on this adsorbent is fast and could be operating on the same time-scale making competition feasible. In vitro competitive adsorption experiments under simulated gastrointestinal conditions were performed. A high affinity of the clay for Trp and NaB was observed. The effect of an excess of KCl to mimic the ionic strength of the ph

Aflatoxin exposure and its relationship to kwashiorkor in African children.

The urinary excretion of aflatoxins in normal children from a periurban area, as well as those hospitalized with kwashiorkor and marasmus was assessed. In no case was aflatoxin isolated from the urine. It is concluded that aflatoxin exposure is unusual in this population and that aflatoxins do not play a primary role in the pathogenesis of kwashiorkor.
