Extravascular albumin concentration of the uvea.

The hypothesis that uveal vessels absorb fluid was tested by measuring the albumin in extravascular uveal tissues and in plasma. From these results the effective albumin concentration was calculated in both rabbits and monkeys. Three separate methods were used to measure uveal albumin, and the results of these were compared. In method 1, the intravenous fluorescein isothiocyanate (FITC)-albumin concentration found in the uvea 5 min after injection (intravascular tracer) was subtracted from that found 2 hr after injection (intravascular plus extravascular tracer) to determine the extravascular

Interaction of methylmercury compounds with albumin.

The nature of interaction between bovine serum albumin (BSA) and methylmercurial compounds has been investigated by ultrafiltration analysis. Four types of BSA samples, mercaptalbumin, its mixed disulfides with glutathione (GSH) and L-cysteine (CySH), and S-carbamidomethylated derivative, were used for binding assays with methylmercury (MM) chloride (MMC) and three kinds of MM mercaptides of low molecular weight thiols, GSH (GS-MM), CySH (CyS-MM) and cysteinylglycine (CG-MM). Among various ligands tested, MMC showed the highest affinity for all BSA species, and the BSA-bound fraction of the li

Anaphylaxis to human serum albumin.

During her 26th week of pregnancy a 20-year-old woman developed generalized pruritus, urticaria, flushing, tinnitus, and tachycardia during plasmapheresis with 5% human serum albumin (HSA) as adjunctive treatment for anti-Kell isoimmunization. The reaction was controlled with intravenous diphenhydramine. Despite pretreatment with diphenhydramine and betamethasone a subsequent attempt to perform plasmapheresis with infusion of 5% HSA resulted in a more severe reaction which progressed to respiratory distress. Intradermal skin testing with 5% HSA produced a 9 x 11-mm wheal and 17 x 21-mm erythem

Albumin metabolism in health and disease.

Studies performed at the University of Cape Town on the metabolism of albumin have been reviewed. The control of albumin metabolism in protein energy malnutrition, in acute exposure to alcohol and after partial hepatectomy in the rat is discussed.

Albumin and the kidney, a friend or foe.

PURPOSE OF REVIEW: Albumin preparations are the most common colloids used in critically ill patients worldwide. This review is to provide the physiological rationale and evidence for benefits and harms in different clinical settings. RECENT FINDINGS: There is a strong physiological rationale for intravenous albumin administration in critically ill patients. Iso-oncotic and hyperoncotic albumin have both been used for volume resuscitation and to maintain effective plasma volume at an optimal fluid balance. This practice, however, has not proven to be superior to using crystalloids or balanced

Albumin Turnover in Peritoneal and Hemodialysis.

The turnover of albumin is increased in both peritoneal dialysis (PD) and hemodialysis (HD) due to increased external losses, normally leading to compensatory increases in the hepatic albumin synthesis. The normal rate of albumin synthesis is on the order of 12 g/day corresponding to an equally large albumin fractional catabolic rate of ~4% daily. Most albumin catabolism is assumed to occur in the endothelium, but there is also renal and hepatic catabolism and leakage into the gastrointestinal tract. In PD the daily losses are on the order of 5 g/day. There are also external albumin losses in

The role of the podocyte in albumin filtration.

In the past decade, our understanding of the role of podocytes in the function of the glomerular filtration barrier, and of the role of podocyte injury in the pathogenesis of proteinuric kidney disease, has substantially increased. Landmark genetic studies identified mutations in genes expressed by podocytes as a cause of albuminuria and nephrotic syndrome, leading to breakthrough discoveries from many laboratories. These discoveries contributed to a dramatic change in our view of the glomerular filtration barrier of the kidney and of the role of podocyte injury in the development of albuminur

Glycated albumin and diabetes mellitus.

BACKGROUND: Diabetes is a growing worldwide problem that is strongly associated with atherosclerosis. Screening and intervention for diabetes in the earliest stages are advocated for the prevention of diabetic complications and cardiovascular disease. SCOPE OF REVIEW: This review gives a background of and discusses the potential clinical utility of glycated albumin (GA) in diabetes. MAJOR CONCLUSIONS: GA is a ketoamine formed via a non-enzymatic glycation reaction of serum albumin and it reflects mean glycemia over two to three weeks. GA can be used for patients with anemia or hemoglobinopat

Recombinant human serum albumin.

Human serum albumin (HSA) is responsible for 80% of the colloid osmotic pressure of plasma (25-33 mmHg). Its main clinical use is in maintaining colloid oncotic pressure and increasing circulating plasma volume with the typical dosage in excess of 10 g per dose. HSA is isolated by fractionating human plasma, which entails possible contamination by viruses or prions. Recombinant HSA (rHSA) has been successfully produced using a methylotrophic yeast, Pichia pastoris. Due to the fact that the clinical usage of HSA infusion often exceeds 10 g, rHSA preparation requires a high level of purity. rHSA
