Emerging roles of basophils in the resolution of acute respiratory distress syndrome.

BACKGROUND: Acute respiratory distress syndrome (ARDS) is a severe form of lung failure with a high mortality rate and no effective pharmacological therapy. Although the cellular and molecular mechanisms involved in ARDS onset have been extensively studied, those governing its resolution remain unclear. Recent human cohort studies have suggested an association between ARDS severity and low blood basophil count. Therefore, in this study, we investigated the roles of basophils in ARDS pathogenesis and resolution. METHODS: We examined the effects of basophil depletion in lipopolysaccharide-induc…

What happens to basophils and tryptase, LXA4 and CysLTs during aspirin desensitization?

INTRODUCTION: Aspirin desensitization (AD) is an effective treatment in patients with non-steroidal anti-inflammatory drugs (NSAID)-exacerba … mparable with baseline and at the other time points in each group (p > 0.05). CONCLUSION: Although basophils are active during the first 3 months of AD, no releases of CysLT, tryptase or LXA4 exist. Therefore, our results suggest that despite active basophils, inhibition of mediators can at least partly explain underlying the mechanism in the first three m …

Eosinophils, basophils and type 2 immune microenvironments in COPD-affected lung tissue.

Although elevated blood or sputum eosinophils are present in many patients with COPD, uncertainties remain regarding the anatomical distribution pattern of lung-infiltrating eosinophils. Basophils have remained virtually unexplored in COPD. This study mapped tissue-infiltrating eosinophils, basophils and eosinophil-promoting immune mechanisms in COPD-affected lungs.Surgical lung tissue and biopsies from major anatomical compartments were obtained from COPD patients with severity grades Global Initiative for Chronic Obstructive Lung Disease stages I-IV; never-smokers/smokers served as controls.…

Contribution of Basophils to Cutaneous Immune Reactions and Th2-Mediated Allergic Responses.

Basophils are potent effector cells of innate immunity and also play a role in T helper 2 (Th2)-mediated allergic responses. But, although their in vitro functions are well studied, their in vivo functions remain largely unknown. However, several mouse models of basophil depletion have recently been developed and used to investigate basophil functions. For example, in a croton oil-induced model of irritant contact dermatitis in conditionally basophil-depleted transgenic mice, we found that basophils rapidly infiltrate inflamed skin and subsequently induce infiltration of eosinophils. We also s…

Interleukin-17A activation on bronchial epithelium and basophils: a novel inflammatory mechanism.

Basophils are the accessory cell type for T-helper (Th)2 induction and initiators in immunoglobulin E-mediated chronic allergic inflammation. Basophils and Th17 cells accumulate at the inflammatory sites, such as the airways of allergic asthmatic patients. We investigated the activation of interleukin (IL)-17A on the primary human basophils/KU812 basophilic cells and primary human bronchial epithelial cells/BEAS-2B bronchial epithelial cells. Cytokines, chemokines, adhesion molecules and intracellular signalling molecules were assayed by ELISA or flow cytometry. Co-culture of bronchial epithel…

In vitro effects of ultraviolet A on histamine release from human basophils.

BACKGROUND: As long-wave ultraviolet (UV) radiation penetrates the dermis, connective tissue cellular components and circulating blood cells can be possible targets for solar UVA. Basophils, involved in the effector phase of the inflammatory response, play a part in skin diseases such as chronic urticaria, psoriasis, atopic dermatitis, fixed drug eruption, allergic contact dermatitis, urticaria pigmentosa, systemic sclerosis and bullous pemphigoid. OBJECTIVE: The evaluation of the in vitro effect of UVA on histamine release from human basophils. METHODS: Basophils from healthy human voluntee…

[Chronic urticaria. Clinical evolution, autologous serum test, count and activation of basophils].

Recent advances on the pathogenesis of chronic urticaria have defined a group of patients with autoantibodies directed to the IgE or to the … basophil count consistently induced expression of CD63. This effect was abrogated in non-releasing basophils, confirming the presence of antibodies directed to the Fc epsilon RI alpha-IgE. We conclude that functional antibodies are present in only a minority of patients and that their identification does not predict the outcome.

Cell membrane glycoproteins of human mast cells: a biochemical comparison with basophils.

The relationship of mast cells (MCs) to other blood leucocytes, and to basophils in particular, is unclear. The relative distribution and abundance of cell surface glycoproteins of normal and neoplastic blood cells has been established as providing a "fingerprint" allowing assignment to a particular hemopoietic cell lineage. We have examined the major labeled glycoproteins of mast cells, basophils, HL-60 cells (granulocytic), and U937 cells (monocytic), after cell surface tritiation by the periodate-borohydride technique. Mast cells have a characteristic glycoprotein profile, different from th…

H2 antihistamines augment antigen-induced histamine release from human basophils in vitro.

H2 antihistamines, including cimetidine, burimamide, metiamide, and tiotidine, consistently augmented antigen-induced histamine release from human basophils in vitro when control histamine release was less than 20% of total. This effect was specific to the H2-receptor blocking activity of these drugs: equivalent degrees of receptor blockade by four different H2 antihistamines resulted in equipotent enhancement; H1-receptor antagonists did not alter histamine release; and aminoguanidine and amodiaquine, agents that inhibit histamine metabolism but do not block H2 receptors, did not enhance hist…

Ablation of immunity to Amblyomma americanum by anti-basophil serum: cooperation between basophils and eosinophils in expression of immunity to ectoparasites (ticks) in guinea pigs.

Basophils infiltrate the skin and other tissues as part of T lymphocyte and/or antibody-mediated immune responses to certain protein antigens, viruses, tumors, and parasites. Although basophils may comprise a significant fraction of leukocytes in these reactions, their precise role has been poorly understood. Guinea pigs expressing acquired immunity to the tick Amblyomma americanum develop basophil- and eosinophil-rich cutaneous inflammatory responses at tick feeding sites, and tick rejection is associated with extensive local basophil degranulation. We report that a specific anti-basophil ser…

AllergoOncology: Emerging Translational and Clinical Significance of Basophils and Mast Cells in Cancer.

Mast cells and basophils, historically defined by their pathogenic roles in allergic diseases and type I hypersensitivity, are increasingly recognized as influential participants in cancer biology. Emerging research in AllergoOncology highlights their plasticity, diverse functions, and significance beyond classical contributions to allergy. This review summarizes current evidence on their presence, activation states, and roles across multiple cancer types. We examine their interactions with other immune populations, their context-dependent pro- and anti-tumor functions, and their potential uti…

Basophils drive the resolution and promote wound healing in adult and aged mice.

An active resolution is critical to control the duration of inflammation and limit its pathological consequences. Defects in resolution during wound healing allow the emergence of chronic wounds, a common complication in the elderly. Here, we show that basophils accumulate in perivascular areas at the periphery of mouse skin wounds for at least 3 wk, during both the inflammation and resolution phases of wound healing. Depletion of basophils induces an increased secretion of inflammatory molecules, accumulation and activation of pro-inflammatory leukocytes, and delays the wound healing response…

Basophils in Skin-Mediated Sensitization Drive Subsequent Lung Inflammation in Airway-Challenged Mice.

BACKGROUND: Atopic dermatitis (AD) and asthma are observed as epidemiologically linked allergic comorbidities, both characterized by elevated systemic IgE levels and type 2 immunity. Basophils play a pivotal role in these responses by producing interleukin-4 (IL-4), which is essential for IgE synthesis and allergic inflammation. However, their specific impact on the progression from AD to asthma remains unclear. METHODS: We utilized an AD model in basophil-deficient Mcpt8Cre mice and temporarily basophil-depleted mice, where topical application of the vitamin D analog MC903 induced the alarmi…

Expression and regulation of Siglec-6 (CD327) on human mast cells and basophils.

BACKGROUND: Mast cells (MC) and basophils are effector cells of allergic reactions and display a number of activation-linked cell surface antigens. Of these antigens, however, only a few are functionally relevant and specifically expressed in these cells. OBJECTIVE: We sought to identify MC- and basophil-specific surface molecules and to study their cellular distribution and regulation during cytokine-induced and IgE-dependent activation. METHODS: Multicolor flow cytometry was performed to recognize surface antigens and to determine changes in antigen expression upon activation. RESULTS: We…

Increased number and activation of peripheral basophils in adult-onset minimal change disease.

Nowadays, the pathogenesis of minimal change disease (MCD) is still not well-known, and the current understanding on MCD is mainly based on data derived from children, and very few adults. Here, we comprehensively analysed the correlation between the changes of peripheral basophils and the incidence rate and relapse of adult-onset MCD. The results showed that in patients at the onset of MCD, the ratio and activation of basophils were all higher than those of healthy controls (all P < .05). In vitro test results showed that basophils from healthy controls can be activated by the serum taken fro…

Modulation of basophils' degranulation and allergy-related enzymes by monomeric and dimeric naphthoquinones.

Allergic disorders are characterized by an abnormal immune response towards non-infectious substances, being associated with life quality reduction and potential life-threatening reactions. The increasing prevalence of allergic disorders demands for new and effective anti-allergic treatments. Here we test the anti-allergic potential of monomeric (juglone, menadione, naphthazarin, plumbagin) and dimeric (diospyrin and diosquinone) naphthoquinones. Inhibition of RBL-2H3 rat basophils' degranulation by naphthoquinones was assessed using two complementary stimuli: IgE/antigen and calcium ionophore…

Accumulation of eosinophils, mast cells, and basophils in the spleen in anaphylactic deaths.

Death in anaphylactic shock cannot be diagnosed by autopsy alone. Morphological diagnosis of anaphylactic death by counting mast cells in the lung and airways have failed to give consistent results. Previously it has been observed that eosinophils seem to accumulate in the spleen in anaphylaxis. The purpose of this study was to investigate if it is possible to safely diagnose anaphylactic deaths by counting eosinophils, mast cells, and basophils in the spleen. In 43 forensic autopsy cases specific antibodies to mast cells, eosinophil-, and basophil granulocytes were used on sections from lung …
