Hepatitis B Virus Disrupts the Blood-Testis Barrier via the Induction of mTOR-Dependent Autophagy in Sertoli Cells.

Hepatitis B virus (HBV) is one of the most serious public health threats worldwide. HBV is not only able to pass through the blood-testis barrier (BTB); It can also cause impairment of male fertility. However, the mechanisms involved in this process remain unknown. In this study, we showed that HBV can establish persistent infection in human and mouse testes. Persistent HBV infection triggers inflammatory cell invasion, testes immune homeostasis imbalance, and the disruption of the BTB formed by inter-Sertoli cells. HBV mainly persisted in the Sertoli cells and could induce the autophagy of Se…

ATG5-dependent autophagy in Sertoli cells protects against cadmium-disrupted blood-testis barrier via perturbing CXCL2/CXCR2 axis.

The blood-testis barrier (BTB), a unique structure established through intercellular connections of Sertoli cells, establishes a protective microenvironment for spermatogenesis and male fertility. Cadmium (Cd), known for its toxicity, is ubiquitously present in the environment. Here, our findings revealed that Cd exposure compromises BTB integrity, as demonstrated by decreased expression of BTB-associated proteins and elevated Dsignal/Dradius values. Mechanistically, we demonstrated that activation of the CXCL2/CXCR2 axis contributes to Cd-induced BTB impairment, as evidenced by experiments us…

Manganese Deficiency Causes Testicular Developmental Disorders, Blood-Testis Barrier Damage, and Spermatogenesis Disruption via Nrf2-Mediated Oxidative Stress.

Background: Manganese (Mn) is a trace element essential for multiple physiological and biological processes. The testis plays a key role in male reproduction by producing sperm and synthesizing male hormones. This study investigates how Mn deficiency affects testicular development, spermatogenesis, and the blood-testis barrier (BTB), and evaluates associated variations in oxidative stress to explore potential mechanisms. Methods: A Mn-deficient diet was used to induce Mn deficiency in mice, with MnCl2 administered via intraperitoneal injection. Mn levels in testicular tissue were measured by a…

Chronic dietary exposure to glyphosate-induced connexin 43 autophagic degradation contributes to blood-testis barrier disruption in roosters.

Glyphosate (GLY) is the most universally used herbicide worldwide and its application has caused extensive pollution to the ecological envir … ooster testis by transcriptome analysis. Cx43 generated by Sertoli cells acts as a key component of blood-testis barrier (BTB). To further investigate the cause of GLY-induced downregulation of Cx43 to disrupt BTB, we found that autophagy activation is revealed in GLY-exposed rooster testis and primary avian Sertoli cells. Moreover, GLY-induced Cx43 downregulation was …

Regulation of complement by Sertoli cells may contribute to the immune protective environment within the blood-testis barrier.

Sertoli cells are a crucial component of the blood-testis barrier (BTB), which isolates the adluminal compartment of the seminiferous tubules from the rest of the testis thus forming an environment to immunely protect the developing germ cells. The mechanisms of regulating immune responses within this environment are currently under investigation. Here, we focused on Sertoli cell regulation of the complement system.

WIN18,446 enhances spermatogonial stem cell homing and fertility after germ cell transplantation by increasing blood-testis barrier permeability.

Spermatogonial stem cells (SSCs) possess a unique ability to recolonize the seminiferous tubules. Upon microinjection into the adluminal compartment of the seminiferous tubules, SSCs transmigrate through the blood-testis barrier (BTB) to the basal compartment of the tubule and reinitiate spermatogenesis. It was recently discovered that inhibiting retinoic acid signaling with WIN18,446 enhances SSC colonization by transiently suppressing spermatogonia differentiation, thereby promoting fertility restoration. In this study, we report that WIN18,446 increases SSC colonization by disrupting the BT…

The cell adhesion molecule BT-IgSF is essential for a functional blood-testis barrier and male fertility in mice.

The Ig-like cell adhesion molecule (IgCAM) BT-IgSF (brain- and testis-specific Ig superfamily protein) plays a major role in male fertility in mice. However, the molecular mechanism by which BT-IgSF supports fertility is unclear. Here, we found that it is localized in Sertoli cells at the blood-testis barrier (BTB) and at the apical ectoplasmic specialization. The absence of BT-IgSF in Sertoli cells in both global and conditional mouse mutants (i.e. AMHCre and Rosa26CreERT2 lines) resulted in male infertility, atrophic testes with vacuolation, azoospermia, and spermatogenesis arrest. Although …

Ablation of the blood-testis barrier in rats and guinea pigs by 48/80, a histamine releaser, and cadmium chloride.

When adult male guinea pigs were injected unilaterally intratesticularly with compound 48/80 (1 mg in 0.05 ml saline), their testicular peritubular capillaries were engorged with blood, intratubular edema was present on the ipsilateral side, and the blood-testicular (BT) barrier (measured by the entrance of acriflavin into the seminiferous tubule) was ablated. The contralateral testis, when injected with saline, showed no pathological changes nor a breakdown of the BT barrier. Subcutaneous injection of guinea pigs with cadmium chloride resulted in a more intense intratubular fluorescence than

Injury and recovery patterns of the blood-testis barrier and spermatogenic function in a surgical unilateral cryptorchidism model in rats.

BACKGROUND: Cryptorchidism is one of the most common congenital disorders of the pediatric urogenital system. In addition to regulating the passage of substances into and out of the seminiferous epithelium, the blood-testis barrier (BTB) plays a critical role in maintaining a stable microenvironment for spermatogenesis. BTB alterations in cryptorchidism remain poorly characterized. This study aimed to establish a surgical unilateral cryptorchidism model in rats (SUCMR) and to investigate the injury and recovery patterns of both the BTB and spermatogenic function within this model. METHODS: Th…

Perfluorohexane Sulfonic Acid Disrupts the Immune Microenvironment for Spermatogenesis by Damaging the Structure of the Blood-Testis Barrier in Mice.

Perfluorohexane sulfonic acid (PFHxS) is extensively used in waterproof coatings and fire-fighting foams, and several studies have found it … ng ≈60-75% of those in the PFHxS-treated group. Additionally, it is found that the structure of the blood-testis barrier (BTB) is damaged after PFHxS treatment, leading to higher expression levels of inflammatory cytokines in the microenvironment for spermatogenesis. Moreover, the expression of proteins associated with mitochondrial biogenesis, including PTEN-induced …

The role of different compounds on the integrity of blood-testis barrier: A concise review based on in vitro and in vivo studies.

Sertoli cells are "nurturing cells'' in the seminiferous tubules of the testis which have essential roles in the development, proliferation and differentiation of germ cells. These cells also divide the seminiferous epithelium into a basal and an adluminal compartment and establish the blood-testis barrier (BTB). BTB shields haploid germ cells from recognition by the innate immune system. Moreover, after translocation of germ cells into the adluminal compartment their nutritional source is separated from the circulatory system being only supplied by the Sertoli cells. The integrity of BTB is i…

Diurnal testosterone oscillations gate drug delivery via phase separation of ZO-1 at the blood-testis barrier in mice.

The blood-testis barrier is essential for sperm development but also blocks the delivery of non-hormonal male contraceptives and other germ-cell-targeting therapies. However, how this barrier opens and closes in a controlled manner remains poorly understood. Here we show that the barrier exhibits time-of-day-dependent changes in permeability, allowing drugs to enter selectively during the light phase while remaining sealed during the dark phase. This gating mechanism is driven by daily fluctuations in testosterone. When testosterone rises at night, it activates calcium channels in supporting c…

Endoplasmic reticulum stress promotes blood-testis barrier impairment in mice with busulfan-induced oligospermia through PERK-eIF2α signaling pathway.

Busulfan, a chemotherapeutic agent for cancer, has detrimental effects on germ cells and fertility, yet the specific mechanisms remain largely uncertain. The blood-testis barrier (BTB) maintains a suitable microenvironment for germ cells self-renewal and spermatogenesis by blocking the interference and damage of deleterious substances. Therefore, we hypothesized that BTB abnormalities might be involved in busulfan-induced oligospermia. To verify the hypothesis, thirty male Balb/c mice were randomly administered with busulfan (at a total dose of 40 mg/kg body weight) by intraperitoneal injectio…

Omega-3 polyunsaturated fatty acids alleviate hydrogen sulfide-induced blood-testis barrier disruption in the testes of adult mice.

Hydrogen sulfide (H2S), a gaseous intracellular signal transducer, participates in multiple physiological and pathological conditions, including reproductive conditions, and disrupts spermatogenesis. The blood-testis barrier (BTB) plays a vital role in spermatogenesis. However, the effect of H2S on the BTB and the underlying mechanism remain unclear. Herein, we examined the effect of H2S and omega-3 polyunsaturated fatty acids (ω-3 PUFAs) on the BTB and testicular functions. ICR male mice were randomly divided into the following groups: control, H2S exposure, and H2S exposure with ω-3 PUFAs in…

ALKBH5 facilitates Srgn stability via m6A demethylation to disrupt blood-testis barrier integrity in the high-fat diet mice.

Obesity has become an important factor affecting male fertility and is often accompanied by abnormal levels of fatty acids, especially saturated fatty acids (SFAs). Palmitic acid (PA; C16:0) is the most frequent SFA in high-fat diet (HFD) and impairs blood-testis barrier (BTB) integrity and decreases sperm quality. RNA N6-methyladenosine (m6A) is involved in lipid metabolism and affects BTB integrity and spermatogenesis. However, it is unclear whether RNA m6A modification plays a role in the BTB damage caused by PA. Herein, we unraveled that PA induced the upregulation of ALKBH5 in TM4 and pri…

Palmitic acid exposure disrupts blood-testis barrier integrity via CHFR-mediated ubiquitination and degradation of SMARCD1 in male mice.

Palmitic acid (PA), a saturated fatty acid abundant in the diet and environment, has been closely associated with male reproductive dysfunction. However, the molecular basis of PA on the blood-testis barrier (BTB) disruption remains unclear. In this study, the chromatin remodeling factor SMARCD1 is identified as a key mediator of PA-induced testicular injury. PA exposure reduced the viability of Sertoli cells and Leydig cells, and markedly downregulated SMARCD1 expression. Although Smarcd1 haploinsufficient (Smarcd1+/-) male mice maintained normal serum testosterone levels, they exhibited comp…
