A new technique to study nutrient flow in host-parasite systems by carbon stable isotope analysis of amino acids and glucose.

Stable isotope analysis of individual compounds is emerging as a powerful tool to study nutrient origin and conversion in host-parasite systems. We measured the carbon isotope composition of amino acids and glucose in the cestode Schistocephalus solidus and in liver and muscle tissues of its second intermediate host, the three-spined stickleback (Gasterosteus aculeatus), over the course of 90 days in a controlled infection experiment. Similar linear regressions of δ13C values over time and low trophic fractionation of essential amino acids indicate that the parasite assimilates nutrients from

Insights into carbon isotope fractionation on trichloroethene degradation in base activated persulfate process: The role of multiple reactive oxygen species.

Understanding the role of reactive oxygen species (ROS) is essential to elucidate the mechanism of contaminants degradation in in-situ chemical oxidation (ISCO). In this study, compound specific isotope analysis (CSIA) and radicals quenching methods were integrated to investigate the roles of hydroxyl radical (HO), sulfate radical (SO4-), and superoxide radical (O2-) on trichloroethene (TCE) degradation during persulfate (PS) activated with base. The carbon isotope fractionation of TCE was found to be dependent of the base:PS ratios, yielding carbon enrichment factors (ε values) from -9.8 ± 0.

Stable Isotope Dilution Assays for Clinical Analyses of Folates and Other One-Carbon Metabolites: Application to Folate-Deficiency Studies.

Folate deficiency is generally accepted as a potential direct or indirect risk factor for diseases including spina bifida, coronary heart diseases, malfunctions of the central nervous system, and cancer. The direct inclusion of folates in the methylation cycle, including the remethylation of homocysteine and regeneration of S-adenosylmethionine, underlines the importance of these vitamins and other components of one-carbon metabolism. Therefore, the aim of the present study was to develop a multiple stable isotope dilution assay (SIDA) for the respective analytes in plasma and tissue samples t

MTHFR C677T genotype influences the isotopic enrichment of one-carbon metabolites in folate-compromised men consuming d9-choline.

BACKGROUND: Homozygosity for the variant 677T allele in the methylenetetrahydrofolate reductase (MTHFR) gene increases the requirement for folate and may alter the metabolic use of choline. The choline adequate intake is 550 mg/d for men, although the metabolic consequences of consuming extra choline are unclear. OBJECTIVE: Deuterium-labeled choline (d9-choline) as tracer was used to determine the differential effects of the MTHFR C677T genotype and the effect of various choline intakes on the isotopic enrichment of choline derivatives in folate-compromised men. DESIGN: Mexican American men

Variations in expression of carbon isotope fractionation of chlorinated ethenes during biologically enhanced PCE dissolution close to a source zone.

The stable carbon isotope values of tetrachloroethene (PCE) and its degradation products were monitored during studies of biologically enhanced dissolution of PCE dense nonaqueous phase liquid (DNAPL) to determine the effect of PCE dissolution on observed isotope values. The degradation of PCE was monitored in a 2-dimensional model aquifer and in a pilot test cell (PTC) at Dover Air Force Base, both with emplaced PCE DNAPL sources. Within the plume down gradient from the source, the isotopic fractionation of dissolved PCE and its degradation products were consistent with those observed in biod

Carbon and nitrogen isotopic analysis of morphine from opium and heroin samples originating in the four major heroin producing regions.

The forensic analysis of stable isotopes is a valuable tool to geo-source natural or semisynthetic drugs such as cocaine and heroin. The present study describes a novel methodology to isolate morphine from opium for isotopic analysis. Furthermore, this isotopic data from regional sources is corroborated with morphine data obtained from seized heroin (deacetylated to morphine) from the same regions. All five primary alkaloids of opium, namely, morphine, codeine, thebaine, noscapine, and papaverine, were quantified using high performance liquid chromatography with photodiode array (PDA) detector

Carbon isotope ratios of endogenous steroids found in human serum-method development, validation, and reference population-derived thresholds.

In order to detect the misuse of testosterone (T), urinary steroid concentrations and concentration ratios are quantified and monitored in a longitudinal manner to enable the identification of samples exhibiting atypical test results. These suspicious samples are then forwarded to isotope ratio mass spectrometry (IRMS)-based methods for confirmation. Especially concentration ratios like T over epitestosterone (E) or 5α-androstanediol over E proved to be valuable markers. Unfortunately, depending on the UGT2B17 genotype and/or the gender of the athlete, these markers may fail to provide evidenc

The use of alkaline hydrolysis as a novel strategy for chloroform remediation: the feasibility of using construction wastes and evaluation of carbon isotopic fractionation.

Laboratory and field-scale pilot experiments were performed to evaluate the feasibility of chloroform degradation by alkaline hydrolysis and the potential of δ(13)C values to assess this induced reaction process at contaminated sites. In batch experiments, alkaline conditions were induced by adding crushed concrete (pH 12.33 ± 0.07), a filtered concrete solution (pH 12.27 ± 0.04), a filtered cement solution (pH 12.66 ± 0.02) and a pH 12 buffer solution (pH 11.92 ± 0.11). The resulting chloroform degradation after 28 days was 94, 96, 99, and 72%, respectively. The experimental data were describ

Effects of varying growth conditions on stable carbon isotope fractionation of trichloroethene (TCE) by tceA-containing Dehalococcoides mccartyi strains.

To quantify in situ bioremediation using compound specific isotope analysis (CSIA), isotope fractionation data obtained from the field is interpreted according to laboratory-derived enrichment factors. Although previous studies that have quantified dynamic isotopic shifts during the reductive dechlorination of trichloroethene (TCE) indicate that fractionation factors can be highly variable from culture-to-culture and site-to-site, the effects of growth condition on the isotope fractionation during reductive dechlorination have not been previously examined. Here, carbon isotope fractionation by

Flushing time and storage effects on the accuracy and precision of carbon and oxygen isotope ratios of sample using the Gasbench II technique.

Continuous-flow isotope ratio mass spectrometry interfaced with a Gasbench II is used for automated and faster analyses of delta(13)C and delta(18)O in water, carbonate, and air samples that are accurate and highly precise. Prior to online chemistry and measurement using the Gasbench technique, rubber septa-capped glass vials are routinely flushed to remove air. Due to the small amounts of sample gas required for isotope analyses using current techniques, care should be taken to properly flush these vials to avoid contamination of sample gas with air. Our results indicate that isotopic composi

Application of ultrafiltration and stable isotopic amendments to field studies of mercury partitioning to filterable carbon in lake water and overland runoff.

Results from pilot studies on colloidal phase transport of newly deposited mercury in lake water and overland runoff demonstrate that the combination of ultrafiltration, and stable isotope amendment techniques is a viable tool for the study of mercury partitioning to filterable carbon. Ultrafiltration mass balance calculations were generally excellent, averaging 97.3, 96.1 and 99.8% for dissolved organic carbon (DOC), total mercury (Hg(T)), and methylmercury (MeHg), respectively. Sub nanogram per liter quantities of isotope were measurable, and the observed phase distribution from replicate ul

Effects of deep-sea cage culture on water quality and stable carbon and nitrogen isotopes of suspended particulate organic matter in the Yellow Sea Cold Water Mass.

The first submersible cage was launched in the Yellow Sea Cold Water Mass (YSCWM) to culture salmonid. Four cruises were conducted to study the impact of aquaculture activities on the surrounding environment. Results indicated that water quality varied among different sampling times, water layers, and sampling stations. In July and August, low water temperature and high dissolved oxygen concentration were found in the bottom water, which provides a suitable habitat for salmonid culture. Significant correlations were observed between δ13C values in particulate organic matter (POM) and chlorophy

Stable carbon ((12/13)C) and nitrogen ((14/15)N) isotopes as a tool for identifying the sources of cyanide in wastes and contaminated soils--a method development.

The occurrence of iron-cyanide complexes in the environment is of concern, since they are potentially hazardous. In order to determine the source of iron-cyanide complexes in contaminated soils and wastes, we developed a method based on the stable isotope ratios (13)C/(12)C and (15)N/(14)N of the complexed cyanide-ion (CN(-)). The method was tested on three pure chemicals and two industrials wastes: blast-furnace sludge (BFS) and gas-purifier waste (GPW). The iron-cyanide complexes were converted into the solid cupric ferrocyanide, Cu(2)[Fe(CN)(6)].7H(2)O, followed by combustion and determinat

Carbon and hydrogen stable isotope fractionation due to monooxygenation of short-chain alkanes by butane monooxygenase of Thauera butanivorans Bu-B1211.

Multi element compound-specific stable isotope analysis (ME-CSIA) is a tool to assess (bio)chemical reactions of molecules in the environment based on their isotopic fingerprints. To that effect, ME-CSIA concepts are initially developed with laboratory model experiments to determine the isotope fractionation factors specific for distinct (bio)chemical reactions. Here, we determined for the first time the carbon and hydrogen isotope fractionation factors for the monooxygenation of the short-chain alkanes ethane, propane, and butane. As model organism we used Thauera butanivorans strain Bu-B1211

Carbon and hydrogen isotope fractionation of phthalates during photocatalysis reactions in aqueous solution containing Fe(III) complexes or iron minerals.

The hydrogen and carbon isotope fractionation factor (ε2H, ε13C) of dimethyl-, diethyl‑ and dibutyl phthalic acid ester during photosensitized degradation by artificial sunlight with Fe(III) ions and iron minerals (hematite, goethite and magnetite) in aqueous solution were examined by compound-specific isotope analysis (CSIA) in order to analyze the degradation mechanism. Hematite does not catalyze photosensitized degradation of phthalates. The correlation of 2H and 13C isotope fractionation (Λ = Δδ2H/Δδ13C) of phthalates with increasing chain length (dimethyl-; diethyl‑; and dibutyl-) were co

Carbon reservoir perturbations induced by Deccan volcanism: Stable isotope and biomolecular perspectives from shallow marine environment in Eastern India.

The Deccan Traps in Western India is hypothesized to have caused significant fluctuations in climatic condition and organic matter (OM) productivity across the Cretaceous-Paleogene Boundary (K/PgB). The periodic release of large amounts of volatiles into the atmosphere is thought to drive these changes. Yet, direct impact of volcanism on the carbon cycle and ecosystem remains relatively unconstrained. For the first time, we attempt to trace changes in both marine and terrestrial carbon reservoirs from pre- and intervolcanic sedimentary units (infra- and inter-trappeans respectively) from Rajah

Concurrent isotope-assisted metabolic flux analysis and transcriptome profiling reveal responses of poplar cells to altered nitrogen and carbon supply.

Reduced nitrogen is indispensable to plants. However, its limited availability in soil combined with the energetic and environmental impacts of nitrogen fertilizers motivates research into molecular mechanisms toward improving plant nitrogen use efficiency (NUE). We performed a systems-level investigation of this problem by employing multiple 'omics methodologies on cell suspensions of hybrid poplar (Populus tremula × Populus alba). Acclimation and growth of the cell suspensions in four nutrient regimes ranging from abundant to deficient supplies of carbon and nitrogen revealed that cell growt

Anthropogenic impacts on mercury concentrations and nitrogen and carbon isotope ratios in fish muscle tissue of the Truckee River watershed, Nevada, USA.

The lower Truckee River originates at Lake Tahoe, California/Nevada (NV), USA and ends in the terminal water body, Pyramid Lake, NV. The river has minimal anthropogenic inputs of contaminants until it encounters the cities of Reno and Sparks, NV, and receives inflows from Steamboat Creek (SBC). SBC originates at Washoe Lake, NV, where there were approximately six mills that used mercury for gold and silver amalgamation in the late 1800s. Since then, mercury has been distributed down the creek to the Truckee River. In addition, SBC receives agricultural and urban nonpoint source pollution, and

Application of dual carbon-bromine stable isotope analysis to characterize anaerobic micro-degradation mechanisms of PBDEs in wetland bottom-water.

Polybrominated diphenyl ethers (PBDEs), one kind of persistent organic pollutants, were widely detected in coastal wetlands. Microbial reductive debromination is one of the most important attenuation processes for PBDEs in anaerobic environment, whereas the underlying reaction mechanisms remain elusive. Dual-element stable isotope analysis was recently recognized to distinguish different reaction mechanism for degradation of organic pollutants. In this study, the dual carbon-bromine isotope effects associated with the anaerobic microbial degradation were first investigated to characterize the

Simultaneous detection of five one-carbon metabolites in plasma using stable isotope dilution liquid chromatography tandem mass spectrometry.

Disturbance in one-carbon (1-C) cycle occurs due to nutritional deficiencies (vitamin B12/folate) or specific genetic polymorphisms. This leads to altered levels of key 1-C metabolites such as SAM (s-adenosyl methionine), SAH (s-adenosyl homocysteine), methionine, homocysteine and MMA (methyl malonic acid). These 1-C metabolites are determinants of cellular methylation potential and epigenetic modifications of DNA which impairs metabolic pathways in several pathological diseases and developmental programming. Though methods were able to measure these analytes only independently, none of the me
