Uptake and biological effects of synthetic glucocorticoids in common carp (Cyprinus carpio).

Uptake and biological effects of synthetic glucocorticoids (GCs) were analyzed using common carp (Cyprinus carpio). Fish were exposed to clobetasol propionate (CP) or clobetasone butyrate (CB) individually or in mixture at 1 μg L(-1) for 21 days. Bioconcentration factor (BCF) of CB was calculated as 100, and BCF of CP was less than 16. No effects were found in fish erythrocyte and leukocyte numbers and serum glucose levels after exposure to the selected GCs. On the other hand, serum concentrations of free amino acids significantly increased in GC-exposed groups. Thus, exposures to synthetic GC

Toxicity of citrate-capped silver nanoparticles in common carp (Cyprinus carpio).

Juvenile common carp (Cyprinus carpio) were used as a model to investigate acute toxicity and oxidative stress caused by silver nanoparticles (Ag-NPs). The fish were exposed to different concentrations of Ag-NPs for 48 h and 96 h. After exposure, antioxidant enzyme levels were measured, including glutathione-S-transferase (GST), superoxidase dismutase, and catalase (CAT). Other biochemical parameters and histological abnormalities in different tissues (i.e., the liver, gills, and brain) were also examined. The results showed that Ag-NPs agglomerated in freshwater used during the exposure exper

Aluminum-induced oxidative stress in lymphocytes of common carp (Cyprinus carpio).

Several studies of fish have shown that aluminum may induce hypoxia, hypercapnia, metabolic acidosis, and respiratory failure. In lymphocytes, morphologic abnormalities and reduced immune activity have been observed. Nevertheless, there is little data on oxidative stress and such data are essential in order to identify its mechanism of action. The common carp Cyprinus carpio, an omnivorous fish commonly used in commercial aquaculture, has been proposed as a test organism in toxicologic assays due to its economic importance and wide geographic distribution. The aim of this work was to evaluate

NMDA modulation of GABA transporter current in carp retinal horizontal cells.

In the present study, the modulatory effect of NMDA on GABA transporter current was investigated on enzymatically isolated horizontal cells of carp retina. After application of NMDA (0.1 mM) for 50 s, the GABA transporter current elicited by GABA (1 mM) was decreased to 78.07+/-3.10% (n=5) of the control level. When the extracellular Ca(2+) was removed from the Ringer's solution, the NMDA inhibitory effect on the GABA transporter current was eliminated. The suppression effect could be attenuated when the Ca(2+) release and Ca(2+) uptake of intracellular Ca(2+) store were blocked after the cell

The biomechanics and evolutionary significance of thermal acclimation in the common carp Cyprinus carpio.

The effects of thermal acclimation were investigated in the common carp Cyprinus carpio L. Acclimation and acute temperature effects were tested during ontogeny from larval [9.5 mm total length (L)] to juvenile (69.0 mm L) stages and between 8 and 21 degrees C. The myosin heavy chain (MHC) composition, myofibrillar Mg(2+)-Ca(2+)-ATPase activity, and muscle strains showed significant thermal acclimation effects. MHCs were only expressed in an acclimation temperature-dependent fashion in fish longer than 37 mm. During fast starts, the temperature had a significant effect on the white muscle stra

Peripheral and central distribution of major branches of the facial taste nerve in the carp.

The major pathways of the peripheral facial taste system in the carp, Cyprinus carpio, are the maxillary (Max), mandibular (Mand), palatine (Pal) and recurrent nerve rami. The peripheral distribution of the sensory fibers of these branches (B) was determined by means of electrophysiological techniques. Max.B., Mand.B. and Pal.B., each of which arises from the gasserian-geniculate ganglionic complexes, were found to innervate respectively, the upper lip and the adjacent skin, the internal and external surface of the lower lip region, and the upper lip and the anterior palate, ipsilaterally. A helix-loop-helix gene orchestrates male fate in common carp.

Fish show exceptional diversity in sex-determination mechanisms, making them a powerful system for studying how these pathways evolve. However, no definitive master sex-determining gene has yet been identified in Cyprinidae, a widely distributed and species-rich fish family. Here, we identified a male-specific gene in common carp encoding Pangu, a helix-loop-helix (HLH) domain peptide localized to Sertoli cells. Loss- and gain-of-function analyses demonstrate that pangu is both necessary and sufficient for male sex determination. Expression and reporter assays indicate that pangu represses cyp

Lactylation of its phosphoprotein interferes with the replication of the spring viremia of carp virus.

The metabolic by-product of glycolysis, lactate, and lactylation-mediated protein modification are involved in various biological processes, including host antiviral responses. The spring viremia of carp virus (SVCV), a rhabdovirus that devastates aquaculture, relies on its multifunctional phosphoprotein (P) for replication and evasion of the host immune system. In this study, we found that SVCV infection increased lactate levels in zebrafish, with lactate modulating SVCV proliferation through lactylation of the viral P protein. ldha-null zebrafish exhibited reduced lactate levels and increase

Mitochondrial responses to anoxia-reoxygenation exposure in crucian carp (Carassius carassius).

The crucian carp (Carassius carassius) is one of the most anoxia-tolerant vertebrates. While physiological underpinnings of its ability to withstand O2 deprivation are well studied, the ability to tolerate the return to normoxia is still enigmatic. Such reoxygenation is associated with detrimental oxidation damage in other organisms, where mitochondria play a central role in the damaging effects. This leads to the question whether mitochondrial adaptations play a central role in the anoxia and reoxygenation tolerance of crucian carp. We here address whether mitochondria from crucian carp circu

A Novel Circular RNA circSPEF2 Regulates Testis Development in Crucian Carp.

Circular RNAs (circRNAs) exhibit significant sex- and development stage-specific expression patterns in the gonads of various fish species, yet their functions and regulatory mechanisms in male reproductive development remain largely unexplored in crucian carp (Carassius auratus). In this study, we characterized the expression features and biological functions of circSPEF2, a circular RNA derived from the reproduction-related gene spef2. Our results showed that circSPEF2 expression was markedly elevated in mature testes and progressively upregulated during gonadal maturation. Functional studie

Genomic localization and immune response of IgT in grass carp (Ctenopharyngodon idellus).

Teleost fish have three types of immunoglobulins (Ig): IgM, IgD, and IgT, of which IgT is unique to teleost fish. Though grass carp (Ctenopharyngodon idella) is one of the most farmed freshwater fish species worldwide, the genomic localization of the heavy chain gene of IgT (IGHT) and the composition and usage of the heavy chain variable region (VH) genes remain unknown. In this study, tissue expression pattern analysis revealed that grass carp IgT is constitutively expressed in various tissues, with higher expression levels in the spleen, kidney, and skin. After immersion immunization with Fl

Establishment of a novel clonal GFP-expressing transgenic ginbuna crucian carp.

The clonal triploid ginbuna crucian carp Carassius auratus langsdorfii, a naturally occurring gynogenetic fish, is suitable for cell transplantation studies to reveal the roles of stem cells and immune cells. To ensure long-term traceability of donor cells within recipient fish, we have established a transgenic ginbuna line that expresses green fluorescent protein (GFP). The Xenopus laevis ef1a promoter was introduced for regulating GFP expression. Tol2 transposon-based transgenesis to ginbuna embryos resulted in producing a putative founder fish (F0) in a mosaic fluorescent fashion; the frequ

Investigating Immunotoxicity in Black Carp (Mylopharyngodon piceus) Fingerlings Exposed to Niclosamide.

Niclosamide (NIC) is a potent salicylanilide molluscicide/helminthicide commonly utilized for parasite and mollusc control in aquatic environments. Due to its persistent presence in water bodies, there is growing concern regarding its impact on aquatic organisms, yet this remains inadequately elucidated. Consequently, this study aims to assess the hepatotoxic effects and detoxification capacity of black carp (Mylopharyngodon piceus) in a semi-static system, employing various parameters for analysis. NIC was applied to juvenile black carp at three different concentrations (0, 10 and 50 μg/L) fo

Characterization and enrichment of spermatogonial stem cells of common carp (Cyprinus carpio).

Spermatogenesis is a systematically organized process that ensures uninterrupted sperm production in which the spermatogonial stem cells (SSCs) play a crucial role. However, the existing absence of teleost-specific molecular markers for SSCs presents a notable challenge. Herein we characterized phenotypically the spermatogonial stem cells using specific molecular markers and transmission electron microscopy. Moreover, we also describe a simple method to suppress common carp spermatogenesis using the combination of Busulfan and thermo-chemical treatment, and finally, we isolate and enrich the u

Identification of microRNAs in Silver Carp (Hypophthalmichthys molitrix) Response to Hypoxia Stress.

Hypoxia is one of the serious stresses in fish culture, which can lead to physical and morphological changes, and cause injury and even death to fish. Silver carp (Hypophthalmichthys molitrix) is an important economic fish and widely distributed in China. MicroRNA is a kind of endogenous non-coding single-stranded small RNA, which is involved in cell development, and immune response and gene expression regulation. In this study, silver carp were kept in the closed containers for hypoxia treatment by spontaneous oxygen consumption. The samples of heart, brain, liver and gill were collected, and

Molecular cloning and characterization of CD63 in common carp infected with koi herpesvirus.

CD63 is a member of the four-transmembrane-domain protein superfamily and is the first characterized tetraspanin protein. In the present study, we cloned the common carp (Cyprinus Carpio) CD63 (ccCD63) sequence and found that the ccCD63 ORF contained 711 bp and encoded a protein of 236 amino acids. Homology analysis revealed that the complete ccCD63 sequence had 84.08% amino acid similarity to CD63 of Sinocyclocheilus anshuiensis. Subcellular localization analysis revealed that ccCD63 was localized in the cytoplasm. Quantitative real-time PCR (qRT-PCR) analysis indicated that ccCD63 was expres
