Platelet-derived growth factors (PDGFs) and their receptors (PDGFRs) have been linked to vascular smooth muscle cell (SMC) migration and proliferation leading to atherosclerosis, restenosis, and chronic allograft rejection. This study describes the effect of CGP 53716, a specific PDGFR tyrosine kinase inhibitor on SMC proliferation and migration in vitro and in neointimal formation in vivo. CGP 53716 inhibited dose dependently tyrosine phosphorylation of both the known PDGFRs: the PDGFR-alpha and PDGFR-beta. In primary rat SMC cultures, a dose-dependent inhibition of PDGF-AA and PDGF-BB induce

ARPC1B (Actin Related Protein 2/3 Complex Subunit 1B) has been found to be involved in platelet abnormalities of immune-mediated inflammatory disease and eosinophilia. However, its role in prostate cancer (PCa) has not been established. We characterized the role of ARPC1B in PCa invasion and metastasis and investigated its prognosis using in vitro cellular models and PCa clinical data. Higher immunohistochemistry (IHC) expressions of ARPC1B were observed in localized and castrate resistant PCa (CRPC) vs. benign prostate tissue (p < 0.01). Additionally, 47% of patients with grade group 5 (GG) s

Burkitt lymphoma is a highly aggressive non-Hodgkin lymphoma that is characterized by MYC deregulation. Recently, the PI3K pathway has emerged as a cooperative prosurvival mechanism in Burkitt lymphoma. Despite the highly successful results of treatment that use high-dose chemotherapy regimens in pediatric Burkitt lymphoma patients, the survival rate of pediatric patients with progressive or recurrent disease is low. PI3Ks are also known to regulate cell migration, and abnormal cell migration may contribute to cancer progression and dissemination in Burkitt lymphoma. Little is known about Burk

Many studies have demonstrated that berberine inhibited the cell migration and invasion in human cancer cell lines. However, the exact molecular mechanism of berberine inhibiting the cell migration and invasion of human melanoma A375.S2 and A375.S2/PLX (PLX4032 induced resistant A375.S2) skin cancer cells remains unknown. In this study, we investigated the anti-metastasis mechanisms of berberine in human melanoma cancer A375.S2 cells and A375.S2/PLX resistant cells in vitro. Berberine at low concentrations (0, 1, 1.5 and 2 μM) induced cell morphological changes and reduced the viable cell numb

Bladder cancer is one of the most prevalent cancers worldwide. Lapachol is a natural compound with antiproliferative effects, but its activity in bladder cancer cells has not yet been evaluated. Cytotoxic activity was assessed using the MTT assay, while long-term effects were evaluated through the clonogenic assay. Cell migration was assessed by the scratch assay, and cell cycle progression was analyzed via flow cytometry. The expression of the JHDM1D gene and the long non-coding RNAs (lncRNAs) JHDM1D-AS1, SBF2-AS1, and RP11-36E7.4 was investigated. Treatment resulted in a dose- and time-depen

BACKGROUND: Regardless of contemporary improvements in cancer treatment, the results of drug treatment are not always efficacious. Thus, the development of novel approaches that affect cancer cell-specific metabolic pathways is needed. Since much evidence has shown that tumor cell proliferation and motility are stimulated by glutamate via activation of its receptors, use of antagonists to these receptors may be the key to control cancer cell progression. Riluzole noncompetitive metabotropic glutamate receptor 1 (mGluR1) antagonist, commonly used to treat patients with amyotrophic lateral scler

Gastric cancer (GC) is a globally lethal malignancy, with invasion and metastasis driving treatment failure and poor prognosis. MX dynamin like GTPase 1 (MX1) shows tumor-specific functional heterogeneity, while its expression, biological functions and molecular mechanisms in GC remain unclear. Here, we explored MX1's clinical significance and its regulatory mechanism in GC cell migration. We integrated public databases and institutional paired clinical samples for bioinformatics analysis of MX1's correlation with clinical outcomes, and verified its pro-migratory effect via Transwell and wound

Triple-negative breast cancer (TNBC) is an aggressive subtype that is characterized by a high metastatic capacity, limited therapeutic options, and a poor prognosis, largely because of its stemness properties and aberrant signaling. Dynorphin A, an endogenous opioid peptide, has been identified as a potential modulator of cancer progression, although its role in TNBC remains unclear. This study demonstrates that the Dynorphin/κ-opioid receptor (KOR) signaling axis is markedly diminished in TNBC patients and TNBC cell lines, suggesting its tumor-suppressive function. Functional assays revealed

PURPOSE: The expression of CD24 is significantly higher in triple-negative breast cancer (TNBC) compared to estrogen receptor and progesterone receptor-positive (ER+ PR+) breast cancer and normal breast tissue. CD24 overexpression is associated with tumorigenesis, metastasis, and drug resistance in TNBC. Moreover, CD24 functions as a "don't eat me" signal and disrupts macrophages-mediated phagocytosis of cancer cells. We, therefore, aim to investigate the therapeutic potential of downregulating CD24 in TNBC cells.
METHODS: We designed four CD24 siRNAs and evaluated their silencing efficiency

Guanine nucleotide-binding protein subunit gamma-12 (GNG12), a G protein γ-subunit, has been identified as a potential regulator of tumor biology. However, its functional relevance in triple-negative breast cancer (TNBC) remains unclear. In this study, we integrated pan-cancer transcriptomic analyses with experimental validation in TNBC models to investigate the expression patterns, clinicopathological significance, and biological roles of GNG12. GNG12 was found to be broadly downregulated across diverse cancer types, and its reduced expression was significantly associated with unfavorable pat

BACKGROUND: Many different types of human malignancies overexpress heat shock proteins (HSPs), which function as oncogenic regulators and control tumorigenesis. However, their contribution to gastric cancer (GC) remains unclear.
OBJECTIVES: This study aimed to investigate the roles of HSP20 during high-temperature intraperitoneal chemotherapy for GC.
METHOD: Immunohistochemistry and western blotting analysis were used to assess the levels of HSP20. Wild-type human HSP20 was sustainably overexpressed in AGS and HGC-27 cells (HSP20-overexpressing cells). The role of HSP20 in GC cells was furth

BACKGROUND: Lung cancer is the leading cause of cancer-related deaths worldwide, so research and development of potential therapeutic targets is urgent.
METHODS: The target gene, AKR1C2, was screened using the TCGA and FerrDb databases. The expression of AKR1C2 in lung cancer, its correlation with the clinical characteristics of patients, and the biological roles and molecular mechanisms involved were assessed by bioinformatics. The expression levels of AKR1C2 in normal lung epithelial cells and lung cancer cells were compared by qRT-PCR and Western blot. The effects of AKR1C2 knockdown on th

Cellular senescence leads to stable cell cycle arrest and an inflammatory senescence-associated secretory phenotype that varies with stressor and cell type. To mitigate these effects and improve health, senotherapeutics (e.g., senolytics and senomorphics) have been developed. Senescent-like endometrial stromal cells (eSCs) lining the uterus of patients with endometriosis and infertility are proposed to impair decidualization, a differentiation process required for uterine receptivity in humans. Quercetin, a natural flavonoid senolytic, dramatically improves decidualization and reduces endometr

Only 20% of patients with muscle-invasive bladder carcinoma respond to cisplatin-based chemotherapy. Since the natural phytochemical sulforaphane (SFN) exhibits antitumor properties, its influence on the adhesive and migratory properties of cisplatin- and gemcitabine-sensitive and cisplatin- and gemcitabine-resistant RT4, RT112, T24, and TCCSUP bladder cancer cells was evaluated. Mechanisms behind the SFN influence were explored by assessing levels of the integrin adhesion receptors β1 (total and activated) and β4 and their functional relevance. To evaluate cell differentiation processes, E- a

Regenerative cell therapy to replenish the missing neurons and glia in the aganglionic segment of Hirschsprung disease represents a promising treatment option. However, the success of cell therapies for this condition are hindered by poor migration of the transplanted cells. This limitation is in part due to a markedly less permissive extracellular environment in the postnatal gut than that of the embryo. Coordinated interactions between enteric neural crest-derived cells (ENCDCs) and their local environment drive migration along the embryonic gut during development of the enteric nervous syst

Background and objectivesESC or Ehrlich solid carcinoma is a type of tumor originating from a spontaneous mammary adenocarcinoma in mice. It is a highly aggressive and fast-growing carcinoma that can create a solid mass when inserted under the skin. Its solid, undifferentiated form makes it an ideal model for researching cancer biology, tumor immunology, and testing various anti-cancer treatments. Additionally, arctiin has multiple beneficial properties, such as anti-proliferative, anti-oxidative, anti-adipogenic, and anti-bacterial. This study aimed to explore the potential anti-cancer benefi

Complement factor H (CFH), a major soluble inhibitor of complement, is a plasma protein that directly interacts with the endothelium of blood vessels. Mutations in the CFH gene lead to diseases associated with excessive angiogenesis; however, the underlying mechanisms are unknown. The present study aimed to determine the effects of CFH on endothelial cells and to explore the underlying mechanisms. The adenoviral plasmid expressing CFH was transduced into HepG2 cells, and the culture medium supernatant was collected and co-cultured with human umbilical vein endothelial cells (HUVECs). Cell prol
