Effects of cations on ceramide-activated protein phosphatase 2A.

Characterization of ceramide-effector(s), which includes protein phosphatase 2A (PP2A) is an important prelude to understanding the molecular basis of sphingolipid-mediated biological effects such as cell growth, differentiation and apoptosis. Recently, the existence of a metal-dependent form of PP2A has been reported (Cai et al., 1995). In this study, we investigated the effects of metal ions and chelators on ceramide-activated PP2A (CAPP). Our study demonstrates that at 0.5 mM concentration, Mg2+ appears to have no significant effect on either basal or ceramide-stimulated phosphatase activit

Increased Plasma Levels of Select Deoxy-ceramide and Ceramide Species are Associated with Increased Odds of Diabetic Neuropathy in Type 1 Diabetes: A Pilot Study.

Plasma deoxy-sphingoid bases are elevated in type 2 diabetes patients and correlate with the stage of diabetic distal sensorimotor polyneuropathy; however, associations between deoxy-sphingolipids (DSL) and neuropathy in type 1 diabetes have not been examined. The primary aim of this exploratory pilot study was to assess the associations between multiple sphingolipid species including DSL and free amino acids and the presence of symptomatic neuropathy in a DCCT/EDIC type 1 diabetes subcohort. Using mass spectroscopy, plasma levels of DSL and free amino acids in DCCT/EDIC type 1 diabetes partic

Ceramide induces mitochondrial abnormalities in insulin-secreting INS-1 cells: potential mechanisms underlying ceramide-mediated metabolic dysfunction of the beta cell.

C2-ceramide, a cell permeable analogue of ceramide [CER] markedly reduced mitochondrial membrane potential [MMP] in insulin-secreting INS cells, which was followed by a significant accumulation of cytochrome c [Cyt c] into the cytosolic compartment. In a manner akin to CER, exposure of these cells to interleukin-1beta [IL-1beta] also resulted in reduction in MMP and cytosolic accumulation of Cyt c. Further, long-term exposure of these cells to either CER [but not its inactive analogue] or IL-1beta caused a marked reduction in their metabolic viability. However, unlike IL-1beta, which increased

The critical role of ceramide in mental health and psychiatric diseases.

Ceramides (Cer) are lipid signaling molecules regulating cell proliferation, differentiation, senescence and apoptosis, whose metabolic disturbance disrupts organismal homeostasis. Accumulating evidence links ceramide imbalance to psychiatric and neurodegenerative diseases, yet few systematic reviews summarize subtype-specific ceramide functions. This review outlines ceramide anabolic and catabolic pathways, compares functional distinctions among ceramides with distinct acyl chain lengths, describes substrate preferences of ceramide metabolic enzymes, and introduces ceramide detection techniqu…

Solvation and hydration of the ceramide headgroup in a non-polar solution.

The microscopic hydration of the ceramide headgroup has been determined using a combination of experimental-both NMR and neutron diffraction techniques and computational techniques-empirical potential structure refinement (EPSR) and molecular dynamics (MD). The addition of water to ceramide in chloroform solutions disrupts the chloroform solvation of the ceramide headgroup, and the water forms distinct pockets of density. Specifically, water is observed to preferentially hydrate the two hydroxyl groups and the carbonyl oxygen over the amide NH motif. Further assessment of the location and orie

Lung cancer and lung injury: the dual role of ceramide.

Sphingolipids play key roles in cancer, yet our current understanding of sphingolipid function in lung cancer is limited to a few key players. The best characterized of these are sphingosine-1-phosphate and ceramide which are described for their opposing roles in cell fate. However, because sphingolipids as a whole are readily interconverted by a complex enzymatic machinery, no single sphingolipid appears to have exactly one role. Instead, the roles of specific sphingolipids appear to be context specific as demonstrated by findings that ceramide-1-phosphate has both proliferative and apoptotic

Loss of TSC2 confers resistance to ceramide and nutrient deprivation.

Nutrient stress that produces quiescence and catabolism in normal cells is lethal to cancer cells, because oncogenic mutations constitutively drive anabolism. One driver of biosynthesis in cancer cells is the mammalian target of rapamycin complex 1 (mTORC1) signaling complex. Activating mTORC1 by deleting its negative regulator tuberous sclerosis complex 2 (TSC2) leads to hypersensitivity to glucose deprivation. We have previously shown that ceramide kills cells in part by triggering nutrient transporter loss and restricting access to extracellular amino acids and glucose, suggesting that TSC2

Anticancer activity of a ceramide analog containing a disulfide linkage.

The effects of exogenous short-chain ceramide (1) on the arrest of growth of cancer cells in vitro and induction of apoptosis have been well documented. In the present study, an analog of 1 with a disulfide linkage, N-(4',5'-dithiaheptanoyl)-D-erythro-ceramide (2), was synthesized and found to be significantly more antiproliferative and cytotoxic than 1 in BT549, A549, and DU145 cancer cells. The activity was correlated with a reduction in cellular glutathione (GSH) level. Ceramide analogs with a tri- and tetra-sulfide moiety were also prepared, but they did not deplete cellular GSH levels and

Current status and perspectives in ceramide-targeting molecular medicine.

Ceramide is not only structurally but also functionally a key molecule in diverse kinds of sphingolipids. In the past decade, ceramide has been shown to be of crucial significance in several cell functions including apoptosis, cell growth, senescence, and cell cycle control. Among them, the role of ceramide in apoptosis induction has extensively been studied, and ceramide-targeting molecular medicine for apoptosis-based diseases such as malignant tumors, atherosclerosis and neurodegenerative disorders appears to come out to the clinical field. We here describe the recent advances in research o

Ceramide inhibits the outward potassium current in rat pinealocytes.

In the present study, we investigated the effect of ceramide on the outward K(+) current in rat pinealocytes using whole cell and single channel recordings. Three components of the whole cell outward K(+) current were separated, an iberiotoxin (IBTX)-sensitive K(+) current (I(KCa)), a transient A current (I(A)) and a delayed rectifier current (I(K)). C6-ceramide reduced all three components of the outward K(+) current. C6-ceramide (30 microM) caused a 53% inhibition of I(KCa) [a component that is generated by the IBTX-sensitive K(+) channel (BK channel)], a 27% inhibition of I(A) and a 17% inh

Ceramides and Type 2 Diabetes: A Scoping Review of Mechanisms, Insulin Resistance, and Nutritional Interventions.

This scoping literature review examined the relationship between ceramides and type 2 diabetes mellitus (T2DM), as well as nutrition interventions that are shown to lower circulating ceramides. Ceramides, bioactive sphingolipids implicated in lipid dysregulation, play a critical role in the development of insulin resistance in obesity-related T2DM. Since diet strongly influences ceramide synthesis, targeted nutritional interventions represent a promising strategy to counteract their impact and improve metabolic health. A systematic search following Preferred Reporting Items for Systematic Revi…

Adipose tissue ceramides: emerging lipid components of extracellular vesicles and their influence on metabolic dysregulation.

Adipose tissue dysfunction is at the interphase between obesity and impaired metabolic health. Increased lipid spillover is a key mechanism by which adipose tissue dysfunction contributes to systemic metabolic dysregulation. This, in turn, leads to the accumulation of lipotoxic lipid species, such as ceramides, within metabolically active tissue not suited for lipid storage tissues thereby fueling insulin resistance. However, the buildup of ceramides also occurs in the adipose tissue of individuals with obesity where these sphingolipids may disrupt energy metabolism, insulin sensitivity and af…

Aberrant ORM (yeast)-like protein isoform 3 (ORMDL3) expression dysregulates ceramide homeostasis in cells and ceramide exacerbates allergic asthma in mice.

BACKGROUND: Asthma, a chronic inflammatory condition defined by episodic shortness of breath with expiratory wheezing and cough, is a serious health concern affecting more than 250 million persons. Genome-wide association studies have identified ORM (yeast)-like protein isoform 3 (ORMDL3) as a gene associated with susceptibility to asthma. Although its yeast ortholog is a negative regulator of de novo ceramide biosynthesis, how ORMDL3 contributes to asthma pathogenesis is not known. OBJECTIVES: We sought to decipher the molecular mechanism for the pathologic functions of ORMDL3 in asthma and
