Cyclization-Modulated Structure and Oxidative Behavior of Methoxy-Substituted Chalcones: Implications for Diesel-Biodiesel Additive Design.

The oxidative instability of diesel-biodiesel blends remains a critical challenge for fuel quality, motivating the search for new antioxidant candidates. In this study, a novel methoxy-substituted cyclic chalcone 1 was synthesized to investigate how intramolecular cyclization influences supramolecular assembly, electronic properties, and oxidative-response behavior in comparison with a structurally related open-chain analog 2. Single-crystal X-ray diffraction revealed that cyclization induces pronounced conformational rigidity and alters the torsion angles governing intermolecular interactions

Identification of a nanomolar, selective MAO-B inhibitor from piperidine-based chalcones with anti-Parkinson potential.

A library of twenty-seven piperidine-based chalcones (PBCs) was designed, synthesised via microwave-assisted methodology, and subsequently evaluated for their inhibitory activity against monoamine oxidases (MAO-A/B), acetylcholinesterase (AChE), and butyrylcholinesterase (BChE). The PBCs exhibited potent and selective inhibition of MAO-B, with IC50 values ranging from 0.014 to >40 μM, while activity towards MAO-A was negligible (>40 μM). Among them, PBC21 was the most active (IC50 = 0.014 ± 0.0026 μM), whereas PBC1, PBC2, and PBC5 showed comparable potencies around 0.5-0.6 μM. Kinetic and reve…

Targeting Blood-Stage Malaria: Design, Synthesis, Characterization, In Vitro, and In Silico Evaluation of Pyrrolidinodiazenyl Chalcones.

Malaria is a pervasive and deadly threat to the global population, and the resources available to treat this disease are limited. There is widespread clinical resistance to the most commonly prescribed antimalarial drugs. To address this issue, we synthesized a range of 4'-pyrrolidinodiazenyl chalcones using a covalent bitherapy approach to study their potential antimalarial properties. We examined the structure-activity relationships of these compounds, which could explain their antimalarial activities. The in vitro blood stage antimalarial activity of the compounds was evaluated against the …

Interruption of Helicobacter pylori-Induced NLRP3 Inflammasome Activation by Chalcone Derivatives.

Helicobacter pylori causes chronic gastritis through cag pathogenicity island (cagPAI), vacuolating cytotoxin A (VacA), lipopolysaccharides … ith non-canonical NLRP3 inflammasome activation. These results show for the first time that certain chalcones could interrupt the activation of the NLRP3 inflammasome in H. pylori-infected THP-1 cells. Therefore, these chalcones may be helpful in alleviating H. pylori-related inflammatory disorders including chronic gastritis.
