Cytoskeleton and tissue origin in the anterior cynomolgus monkey eye.

We studied cytoskeletal proteins and other markers for embryologic origin in the outflow pathways of the aqueous humor, cornea, sclera, and ciliary muscle of the cynomolgus monkey. The corneal endothelium and trabecular cells stained with markers for vimentin, smooth muscle cell alpha-actin, F-actin, spectrin, vinculin, and talin. The endothelium of Schlemm's canal stained with markers for vimentin, spectrin, and F-actin. These results suggest that trabecular cells are a kind of myofibroblast and support the belief that the endothelial cells of Schlemm's canal are vascular in origin. Fibrillar

Impairment of the myocardial ultrastructure and changes of the cytoskeleton in dilated cardiomyopathy.

This study was designed to determine the morphological correlate of chronic heart failure. Myocardial tissue from eight patients undergoing transplantation surgery because of end-stage dilated cardiomyopathy was investigated by electron microscopy and immunocytochemistry using monoclonal antibodies against elements of the cytoskeleton: desmin, tubulin, vinculin, and vimentin. The tissue showed hypertrophy, atrophy of myocytes, and an increased amount of fibrosis. Ultrastructural changes consisted of enlargement and varying shape of nuclei, numerous very small mitochondria, proliferation of T t

Effects of electric fields on cytoskeleton of corneal stromal fibroblasts.

Low-level, steady electric fields (6-10 volts/cm) stimulated cultured corneal stromal fibroblasts to undergo directional orientation and translocation. The orientative movements (galvanotropism) consisted of somatic elongation of the cells into spindle shapes along an imaginary axis perpendicular to the field; the cathodal edge of the cell underwent retraction, while the anodal edge and the longitudinal ends developed ruffled membranes and lamellipodia. The translocational movements (galvanotaxis) consisted of directed migration of the cells towards the anode. While most actin-containing stres

Rac1 inhibition prevents axonal cytoskeleton dysfunction in transthyretin amyloid polyneuropathy.

Hereditary transthyretin amyloidosis with polyneuropathy (ATTRv-PN) is characterized by the deposition of amyloidogenic transthyretin, particularly in dorsal root ganglia (DRG) and peripheral nerve axons, resulting in sensorimotor axonopathy. Here, we investigated cytoskeleton alterations in peripheral axons from an ATTRv-PN mouse model, the hTTRA97S knockin mice. Proteomics of hTTRA97S sural nerves revealed dysregulation of actin-related proteins. hTTRA97S DRG neurons presented a defective actin distribution in growth cones along with a reduction in axonal actin trails, impacting synaptic ves

Nicotinic acid modulates intracellular calcium concentration and disassembles the cytoskeleton.

Nicotinic acid (NA), a member of the vitamin B family, is well known for its functions in the treatment and prevention of atherosclerosis due to decreasing plasma levels of low-density lipoprotein cholesterol. In recent years, the major side effect of NA, cutaneous flushing, has also attracted extensive attention. However, the effects of NA in other aspects of physiology or cell biology have remained elusive. The present study provided evidence that high concentrations of NA were able to first reduce and later elevate intracellular [Ca2+] in the NIH3T3 cell line. The reduction of the intracell

Calponin 3 regulates actin cytoskeleton rearrangement in trophoblastic cell fusion.

Cell-cell fusion is an intriguing differentiation process, essential for placental development and maturation. A proteomic approach identified a cytoplasmic protein, calponin 3 (CNN3), related to the fusion of BeWo choriocarcinoma cells. CNN3 was expressed in cytotrophoblasts in human placenta. CNN3 gene knockdown promoted actin cytoskeletal rearrangement and syncytium formation in BeWo cells, suggesting CNN3 to be a negative regulator of trophoblast fusion. Indeed, CNN3 depletion promoted BeWo cell fusion. CNN3 at the cytoplasmic face of cytoskeleton was dislocated from F-actin with forskolin

Cleavage of tensin during cytoskeleton disruption in YTX-induced apoptosis.

Yessotoxin (YTX) is a marine algal toxin previously shown to induce apoptosis in L6 and BC3H1 myoblast cell lines. Disassembly of the F-actin cytoskeleton and cleavage of tensin, a cytoskeletal protein localised at the focal adhesion contacts, appear during this apoptotic process. Tensin binds to actin filaments at the focal adhesion contacts and it links the actin cytoskeleton to the extracellular matrix (ECM). This binding occurs via integrin receptors and it makes tensin a potential link between the actin cytoskeleton and signal transduction. This study evaluates disruption in the F-actin c

WASP family proteins act between cytoskeleton and cellular signaling pathways.

This review considers the proteins of the WASP (Wiskott-Aldrich syndrome protein) family and their role in the regulation of actin-based motility. It contains detailed classification of the WASP family proteins and data on their subcellular localization. Impairments of expression of the WASP family proteins cause certain cell pathologies. The review also deals with domain organization of these proteins and proteins interacting with various domains of the WASP proteins. Special attention is given to analysis of the role of the WASP family proteins in initiating directed actin assembly in the le

Labeling Strategies for Monitoring Cytoskeleton Dynamics in Cultured Epithelial Cells.

Collective cell migration relies on coordinated cytoskeletal remodeling, yet the impact of live-cell actin filament probes on these dynamics remains poorly characterized. Here, we systematically compared the performance and cellular effects of fluorogenic jasplakinolide-based probes, SiR-actin and SiR-XActin, with the genetically encoded Lifeact reporter in epithelial monolayers. Using an injury-free wound healing assay combined with widefield images, particle image velocimetry, kymographs, and FUCCI-based cell cycle tracking system, we assessed how probe choice, efflux inhibition, and genetic

TNXB Regulates Spermatogenesis by Maintaining Cytoskeleton and Cell Junction Stability.

Tenascin-X (TNXB) is an extracellular matrix glycoprotein known for its structural and regulatory roles in connective tissues, yet its pathophysiological role in testicular development and spermatogenesis remains uncharacterized. Here, we identified TNXB as clinically implicated in human spermatogenic disorder, with a significant downregulation in the testes of patients with non-obstructive azoospermia (NOA) revealed by isobaric tags for relative and absolute quantification (iTRAQ)-based proteomics. Developmental profiling in mice revealed a progressive increase in TNXB expression with testicu

Mitofusin 2 regulates neutrophil adhesive migration and the actin cytoskeleton.

Neutrophils rely on glycolysis for energy production. How mitochondria regulate neutrophil function is not fully understood. Here, we report that mitochondrial outer membrane protein Mitofusin 2 (MFN2) regulates neutrophil homeostasis and chemotaxis in vivoMfn2-deficient neutrophils are released from the hematopoietic tissue, trapped in the vasculature in zebrafish embryos, and not capable of chemotaxis. Consistent with this, human neutrophil-like cells that are deficient for MFN2 fail to arrest on activated endothelium under sheer stress or perform chemotaxis on 2D surfaces. Deletion of MFN2

Proteinuric Kidney Diseases: A Podocyte's Slit Diaphragm and Cytoskeleton Approach.

Proteinuric kidney diseases are a group of disorders with diverse pathological mechanisms associated with significant losses of protein in the urine. The glomerular filtration barrier (GFB), comprised of the three important layers, the fenestrated glomerular endothelium, the glomerular basement membrane (GBM), and the podocyte, dictates that disruption of any one of these structures should lead to proteinuric disease. Podocytes, in particular, have long been considered as the final gatekeeper of the GFB. This specialized visceral epithelial cell contains a complex framework of cytoskeletons fo

KBP-cytoskeleton interactions underlie developmental anomalies in Goldberg-Shprintzen syndrome.

Goldberg-Shprintzen syndrome (GOSHS, MIM #609460) is an autosomal recessive disorder of intellectual disability, specific facial gestalt and Hirschsprung's disease (HSCR). In 2005, homozygosity mapping in a large consanguineous family identified KIAA1279 as the disease-causing gene. KIAA1279 encodes KIF-binding protein (KBP), whose function is incompletely understood. Studies have identified either the mitochondria or the cytoskeleton as the site of KBP localization and interactions. To better delineate the KIAA1279-related clinical spectrum and the molecular mechanisms involved in GOSHS, we s

[Listeria-induced host cellular actin cytoskeleton rearrangement and phospholipase D].

Either phagocytosis of macrophage to pathogen or pathogen-induced invasion into non-professional phagocytes, such as epithelial cells, require actin cytoskeletal rearrangements and remodeling of the plasma membrane, which are regulated precisely by monogeric GTPase and the correlated proteins. As a key signaling molecule in the cell, phosphotidicphospholipase D (PLD) regulates or interacts directly with cellular actin cytoskeleton rearrangement. Phospholipase D plays an important role in FcgammaRI and C-reactive protein-mediated phagocytosis and phosphorylated cofilin, a ADF (actin depolymeriz

Vascular smooth muscle actin cytoskeleton in cerebral artery forced dilatation.

BACKGROUND AND PURPOSE: We investigated the role of actin polymerization in regulating arterial diameter in response to increasing pressure and modulating forced dilatation of cerebral arteries at pressures above the upper limit of autoregulation. METHODS: Posterior cerebral arteries (n = 12) were isolated and pressurized in a special arteriograph that allowed control of intravascular pressure and measurement of lumen diameter. Intact arteries in the absence (control) or presence of 3.0 mumol/L cytochalasin B (CB), an inhibitor of actin polymerization, were subjected to stepwise increases in

Cytoskeleton in normal and reactive human retinal pigment epithelial cells.

The cytoskeleton of normal and reactive retinal pigment epithelium (RPE) was analyzed immunohistochemically in five light microscopically normal formalin-fixed, paraffin-embedded human eyes enucleated because of orbital tumor and in 44 eyes with a uveal melanoma. In 26 eyes, the RPE overlying the tumor was morphologically normal or atrophic; in 18 eyes, hyperplastic changes were present. Normal RPE cells lacked vimentin, but it was present in 35 of 44 eyes with uveal melanoma. Antibodies that recognize cytokeratins CK8 and CK18 labeled normal and reactive RPE cells in all specimens. Although C

Understanding the impact of cytoskeleton disruption on synaptic plasticity in Alzheimer's disease.

Tau acetylation plays a critical role in the pathogenesis of Alzheimer's disease (AD) by disrupting cytoskeletal dynamics and synaptic plasticity. Acetylated tau aggregates contribute to the formation of neurofibrillary tangles (NFTs), leading to microtubule destabilization, impaired intracellular transport, and dysregulation of synaptic plasticity mechanisms. The cytoskeletal proteins, comprising microfilaments, microtubules, and intermediate filaments, are essential for maintaining neuronal structure, polarity, and signaling. The disruption of cytoskeletal integrity impairs dendritic spine f

Roles of the tubulin-based cytoskeleton in the Toxoplasma gondii apical complex.

Microtubules (MTs) play a vital role as key components of the eukaryotic cytoskeleton. The phylum Apicomplexa comprises eukaryotic unicellular parasitic organisms defined by the presence of an apical complex which consists of specialized secretory organelles and tubulin-based cytoskeletal elements. One apicomplexan parasite, Toxoplasma gondii, is an omnipresent opportunistic pathogen with significant medical and veterinary implications. To ensure successful infection and widespread dissemination, T. gondii heavily relies on the tubulin structures present in the apical complex. Recent advances
