Effects of different oxygen concentrations during intermittent hyperoxic training on endurance capacity in well-trained male mice.

While training under intermittent hyperoxia (INT) enhances endurance capacity, the effects of specific oxygen concentrations remain unclear. This study compared 50% O2 (INT50) and 75% O2 (INT75) in well-trained male mice, which had increased maximal work values by 7.7-fold through a 7-week voluntary running program, during a 4-week treadmill program. Results showed that INT75, but not INT50, significantly improved endurance capacity compared to the normoxic-trained (ET) group (Bayes factor, BF ≥3). The enhanced exercise capacity in the INT75 group was linked to higher levels of metabolic enzym

Time-course for onset and decay of physiological adaptations in endurance trained athletes undertaking prolonged heat acclimation training.

Short-term heat acclimation (HA) appears adequate for maximizing sudomotor adaptations and enhancing thermal resilience in trained athletes. However, for enhanced erythropoiesis and transfer effects to exercise capacity in cooler environments, prolonged HA appears necessary. To establish the time-course for physiological adaptations and performance effects, 20 male elite cyclists were divided into an intervention group (HEAT; n = 10) completing 5 weeks of HA (six one-hour HA-training sessions per week) and control (n = 10) tested pre and post in hot (40°C) and cool conditions (20°C). HEAT comp

Effect of aging, endurance training, and denervation on innate immune signaling in skeletal muscle.

Skeletal muscle function relies on mitochondria for energy and for mediating its unique adaptive plasticity. The NLRP3 inflammasome complex is an innate immune mechanism that responds to mitochondrial damage-associated molecular patterns (DAMPs); however, its activity relative to mitochondrial dysfunction in muscle requires exploration. The purpose of this study was to characterize immune signaling and mitochondrial function in muscle during aging, endurance training, and disuse induced by denervation. Denervation led to decreases in muscle mass, mitochondrial content, and impaired respiration…

Stage-Dependent Brain Plasticity Induced by Long-Term Endurance Training: A Longitudinal Neuroimaging Study.

Long-term physical training is known to induce brain plasticity, yet how these neural adaptations evolve across different stages of training remains underexplored. This two-year longitudinal study investigated the stage-dependent effects of endurance running on brain structure and resting-state function in healthy college students. Thirty participants were recruited into three groups based on their endurance training level: high-level runners, moderate-level runners, and sedentary controls. All participants underwent baseline and two-year follow-up MRI scans, including T1-weighted structural i…

Pi-based biochemical mechanism of endurance-training-induced improvement of running performance in humans.

PURPOSE: Endurance training improves running performance in distances where oxidative phosphorylation (OXPHOS) is the main ATP source. Here, a dynamic computer model is used to assess possible biochemical mechanisms underlying this improvement. METHODS: The dynamic computer model is based on the "Pi double-threshold" mechanism of muscle fatigue, according to which the additional ATP usage appears when (1) inorganic phosphate (Pi) exceeds a critical value (Picrit); (2) exercise is terminated because of fatigue, when Pi reaches a peak value (Pipeak); (3) the Pi increase and additional ATP usage…

Resistance and endurance training in intermittent hypoxia reduce body fat mass and blood pressure.

BACKGROUND: Hypoxia is an environmental condition that occurs in sports performed at high altitude. Adaptation to hypoxia is accompanied by changes in body composition and cardiac function that could impair sport performance in altitude. These changes concern mainly to a reduction in muscle mass and an increased heart rate. In this context, a resistance training protocol in a normobaric hypoxia chamber has been implemented. Therefore, the aim of this study was to study the changes in body composition and cardiovascular variables after a training period in intermittent hypoxia. METHODS: A sing

Long-Term Endurance Exercise Training Alters Repolarization in a New Rabbit Athlete's Heart Model.

In the present study, the effect of long-term exercise training was investigated on myocardial morphological and functional remodeling and on proarrhythmic sensitivity in a rabbit athlete's heart model. New-Zealand white rabbits were trained during a 12-week long treadmill running protocol and compared with their sedentary controls. At the end of the training protocol, echocardiography, in vivo and in vitro ECG recordings, proarrhythmic sensitivity with dofetilide (nM) were performed in isolated hearts, and action potential duration (APD) measurements at different potassium concentrations (4.5

Influence of Sodium Glucose Cotransporter 2 Inhibition on Physiological Adaptation to Endurance Exercise Training.

CONTEXT: The combination of two beneficial antidiabetes interventions, regular exercise and pharmaceuticals, is intuitively appealing. However, metformin, the most commonly prescribed diabetes medication, attenuates the favorable physiological adaptations to exercise; in turn, exercise may impede the action of metformin. OBJECTIVE: We sought to determine the influence of an alternative diabetes treatment, sodium glucose cotransporter 2 (SGLT2) inhibition, on the response to endurance exercise training. DESIGN, PARTICIPANTS, AND INTERVENTION: In a randomized, double-blind, repeated measures p

Building strength, endurance, and mobility using an astaxanthin formulation with functional training in elderly.

BACKGROUND: Building both strength and endurance has been a challenge in exercise training in the elderly, but dietary supplements hold promise as agents for improving muscle adaptation. Here, we test a formulation of natural products (AX: astaxanthin, 12 mg and tocotrienol, 10 mg and zinc, 6 mg) with both anti-inflammatory and antioxidant properties in combination with exercise. We conducted a randomized, double-blind, placebo-controlled study of elderly subjects (65-82 years) on a daily oral dose with interval walking exercise on an incline treadmill. METHODS: Forty-two subjects were fed AX

Endurance Times of the Thoracolumbar Musculature: Reference Values for Female Recreational Resistance Training Participants.

The assessment of thoracolumbar muscle endurance (TLME) is common among strength and conditioning professionals and clinicians desiring to quantify baseline muscle performance and determine injury risk. Reference values for such assessments are documented in the literature; however, their utility may be of limited value due to heterogeneous participant selection and limited demographic reporting. Moreover, active cohorts who engage in resistance training (RT) may reach a ceiling effect on existing reference values when testing routinely trained muscles. Thus, the purpose of this study was to e

Endurance training and chronic intermittent hypoxia modulate in vitro salicylate-induced hepatic mitochondrial dysfunction.

Mitochondrial function is modulated by multiple approaches including physical activity, which can afford cross-tolerance against a variety of insults. We therefore aimed to analyze the effects of endurance-training (ET) and chronic-intermittent hypobaric-hypoxia (IHH) on liver mitochondrial bioenergetics and whether these effects translate into benefits against in vitro salicylate mitochondrial toxicity. Twenty-eight young-adult male rats were divided into normoxic-sedentary (NS), normoxic-exercised (NE), hypoxic-sedentary (HS) and hypoxic-exercised (HE). ET consisted of 1h/days of treadmill r

Endurance training and detraining in mitochondrial myopathies due to single large-scale mtDNA deletions.

At present there are limited therapeutic interventions for patients with mitochondrial myopathies. Exercise training has been suggested as an approach to improve physical capacity and quality of life but it is uncertain whether it offers a safe and effective treatment for patients with heteroplasmic mitochondrial DNA (mtDNA) mutations. The objectives of this study were to assess the effects of exercise training and detraining in eight patients with single, large-scale mtDNA deletions to determine: (i) the efficacy and safety of endurance training (14 weeks) in this patient population; (ii) to …

Influence of endurance exercise training on distribution of vascular adaptations in rat skeletal muscle.

We hypothesized that an exercise training program consisting of treadmill running at 32 m/min up a 15% incline, 90 min/day, 5 days/wk for 12-14 wk, would elicit vascular adaptation in skeletal muscle of all fiber types in rats. This hypothesis was based on previous reports that this intensity and duration of training caused increases in oxidative capacity in rat skeletal muscle of all fiber types. Skeletal muscle vascular transport capacity was examined with measurements of total and regional (radiolabeled microspheres) flow capacity, capillary filtration coefficient (CFC), and permeability-su
