Dysembryoplastic neuroepithelial tumor located in pericallosal and intraventricular area in a child. Case report.

Dysembryoplastic neuroepithelial tumors (DNETs) are frequently seen in children and young adults with intractable epilepsy, and are typically located in the temporal cortex. Extracortical location of DNET is a very rare occurrence. The authors report on a child with a unique extracortical location of DNET with an extensive involvement to the supracallosal cistern and callosum, septum pellucidum, and lateral ventricle ependyma. The authors discuss the radiological and pathological characteristics of the lesion and reiterate the difficulty in differentiating the histological characteristics of c…

Chordoid Glioma of Third Ventricle With an Epidermoid Cyst: Coexistence or Common Histogenesis?

Chordoid glioma (CG) is a World Health Organization classified grade II tumor located exclusively in the region of anterior third ventricle. Association of CG with other lesions is extremely rare. We report a case of CG in a 45-year-old male coexisting with an epidermoid cyst in the third ventricle. Ultrastructural examination of the CG revealed microvilli, junctional complexes, and intermediate filaments within the cytoplasm suggesting origin from specialized ependyma. The association of the 2 lesions appears coincidental as convincing evidence for a common histogenesis was not found.

The sulcus medianus organ in the fourth cerebral ventricle of the rabbit: a previously undescribed circumventricular organ.

The sulcus medianus of the fourth ventricle in the rabbit contains a previously undescribed circumventricular organ. The cells in this region are predominantly nonciliated. The apical surface of each cell has characteristic microvillous projections with pinocytotic vesicles. The cell body is elongated, with the nucleus situated in the body or basal process; the cytoplasm of the cell is pale and contains microtubules and filaments. The general morphology of the cells is similar to tanycytes found in other circumventricular organs. Supraependymal cells were present on the non-ciliated ependyma i…

Intraventricular tumors in tuberous sclerosis.

Eleven of our own cases of tuberous sclerosis with intraventricular tumor and 34 cases from the literature are reviewed. Initial symptoms and identification of tuberous sclerosis and tumor are reviewed. Results of surgical treatment, including direct radical excision and shunting are given. On gross examination, the tumor was most often loosely connected to ependyma near the foramen of Monro, causing hydrocephalus. Giant cell subependymal astrocytoma was frequently found on microscopic examination. The glial origin of the tumor is discussed and direct surgical approach of the tumor is proposed…

Motor protein Kif6 regulates cilia motility and polarity in brain ependymal cells.

Motile cilia on ependymal cells that line brain ventricular walls beat in concert to generate a flow of laminar cerebrospinal fluid (CSF). Dyneins and kinesins are ATPase microtubule motor proteins that promote the rhythmic beating of cilia axonemes. Despite common consensus about the importance of axonemal dynein motor proteins, little is known about how kinesin motors contribute to cilia motility. Here, we show that Kif6 is a slow processive motor (12.2±2.0 nm/s) on microtubules in vitro and localizes to both the apical cytoplasm and the axoneme in ependymal cells, although it does not displ…

Vimentin in ependymal and subventricular proliferative zone cells of rat telencephalon.

Ependymal cells of cerebral ventricles are the most probable candidate progenitor cells. We studied ependymal and subventricular zone cells expressing intermediate filament protein vimentin. The results suggest that the ventricular ependyma represent a homogenous cell population. Some ependymocytes have long processes contacting with blood vessels, which makes them similar to tanycytes of the third cerebral ventricle. New data on the structure of ependymocyte processes attest to their active involvement into creation of a special microenvironment for neural stem cells of the subventricular pro…

Glial choristoma of the tongue: report of a case and review of the literature.

Glial choristoma of the tongue is an extremely rare developmental malformation. The authors report a case of a 5-month-old male baby with a congenital glial choristoma located on the posterior part of midline of the left dorsal tongue. Histological examination of the resected specimen revealed a poorly demarcated submucosal mass containing neuroglial tissue, scattered neuron, choroids plexus and ependyma. In addition to neuroglial tissue, a sheet of leptomeningeal tissue was observed more rarely in the case. The clinical and pathological characteristics of previous cases and their probable emb…

Ventricular-subventricular zone stem cell niche adaptations in a mouse model of post-infectious hydrocephalus.

Congenital post-infectious hydrocephalus (PIH) is a condition characterized by enlargement of the ventricular system, consequently imposing a burden on the associated stem cell niche, the ventricular-subventricular zone (V-SVZ). To investigate how the V-SVZ adapts in PIH, we developed a mouse model of influenza virus-induced PIH based on direct intracerebroventricular injection of mouse-adapted influenza virus at two distinct time points: embryonic day 16 (E16), when stem cells line the ventricle, and postnatal day 4 (P4), when an ependymal monolayer covers the ventricle surface and stem cells…

Differential expression of cell adhesion molecules (CAM), neural CAM and epithelial cadherin in ependymomas and choroid plexus tumors.

A series of frozen specimens of 18 ependymomas and 7 choroid plexus tumors were examined for their expression of cell adhesion molecules, such as neural cell adhesion molecule (NCAM), its polysialylated isoforms (PSA NCAM), and epithelial (E-) cadherin, and of intermediate filament proteins, such as glial fibrillary acidic protein (GFAP) and cytokeratin, using various monoclonal and polyclonal antibodies. Normal choroid plexus and ependyma were taken as controls. Anti-E-cadherin immunoreactivity was observed on the basolateral part of most adult choroid plexus and benign choroid plexus papillo…

Ultrastructure of the wall of the Dandy-Walker cyst. A case report.

The ultrastructure of the wall of the Dandy-Walker cyst has been described rarely. A boy aged 2 years was confirmed clinically, neuroradiologically, and operatively as having a Dandy-Walker cyst in the posterior fossa. The cyst wall obtained during surgery consisted of an outer arachnoid cell layer, intermediate interwoven neurological strands, and an inner layer of cells which lacked the characteristic appearance of ependyma. An unusual finding was a small, buried island of ependymal cells in the intermediate layer of the neuroglial tissue. Ultrastructural study of the cyst wall provides a be…

Ependymal cells: roles in central nervous system infections and therapeutic application.

Ependymal cells are arranged along the inner surfaces of the ventricles and the central canal of the spinal cord, providing anatomical, physiological and immunological barriers that maintain cerebrospinal fluid (CSF) homeostasis. Based on this, studies have found that alterations in gene expression, cell junctions, cytokine secretion and metabolic disturbances can lead to dysfunction of ependymal cells, thereby participating in the onset and progression of central nervous system (CNS) infections. Additionally, ependymal cells can exhibit proliferative and regenerative potential as well as secr…

Ependymal polarity defects coupled with disorganized ciliary beating drive abnormal cerebrospinal fluid flow and spine curvature in zebrafish.

Idiopathic scoliosis (IS) is the most common spinal deformity diagnosed in childhood or early adolescence, while the underlying pathogenesis of this serious condition remains largely unknown. Here, we report zebrafish ccdc57 mutants exhibiting scoliosis during late development, similar to that observed in human adolescent idiopathic scoliosis (AIS). Zebrafish ccdc57 mutants developed hydrocephalus due to cerebrospinal fluid (CSF) flow defects caused by uncoordinated cilia beating in ependymal cells. Mechanistically, Ccdc57 localizes to ciliary basal bodies and controls the planar polarity of e…

Ultrastuctural study on the relations among nerve elements and ependymal cells of the "Bradypus tridactylus".

On hand of the electonmicroscope, the authors study 24 parts of the encephalic ventricles in samples of Bradypus tridactylus, relating the nervous structures to the ependymal lining, chiefly emphasizing the presence of intraventricular unmyelinated fibres. In various of these regions, typical nerve fibres and other intraventricular cell projections, that cannot be taken as nerve structures, because of the continuity they show along with ependymal cells, were observed. Finally they make an analysis of some functions related to arrangements of this nature.

Complement-dependent pathogenicity of brain-specific antibodies in cerebrospinal fluid.

The specificity and potential pathogenicity of autoantibodies vary between neurological diseases. It is often unclear whether their detection in cerebrospinal fluid (CSF) is a consequence or a cause of pathology. The goal was to test whether administration of brain-specific antibodies into CSF would be sufficient for pathology. Purified immunoglobulin G from a neuromyelitis optica patient was injected intrathecally with complement to naïve mice. Histopathological analysis at 7 days revealed damage to the ependyma, disruption of the CSF parenchymal barrier and pathologic lesions, distant from t…

Symptomatic ependymal cysts of the perimesencephalic and cerebello-pontine angle cisterns.

Primary intracranial ependymal cysts are extremely rare. These are congenital, benign ependyma lined, commonly intraparenchymal and uncommonly extraparenchymal cysts in leptomeningeal location of variable size seen in the adult life. The authors report two cases of symptomatic ependymal cysts: one in the perimesencephalic cistern in a 10-month-old infant presenting with delayed mile stones and another in the cerebello-pontine cistern in a morbidly obese 35-year-old woman with known benign intracranial hypertension now presenting with hemifacial spasm. Neurosurgical intervention was curative in…

Naegleria australiensis ssp. italica: experimental study in mice.

A subspecies of Naegleria australiensis, N. australiensis italica, pathogenic for mice, was recently isolated and identified from an Italian thermal spa. We describe the histopathological changes of the central nervous system with experimental infection of albino mice. The histopathological patterns are intermediate to those seen with infection caused by N. fowleri and N. australiensis or Acanthamoeba spp. An acute inflammatory reaction was present within the choroid plexus, ependyma, midbrain, cerebellum, and basal ganglia. Occasional single amebic trophozoites were found within some microabs…

Degeneration of serotonin-specific neurons in the brain in experimental Trypanosoma brucei infection.

ORMEROD and HUSSEIN (1986) have shown that an intracellular stage of Trypanosoma brucei rhodesiense causes the destruction of ependymal cells lining the ventricles of the brain. The ventricular ependymal cells are intimately associated with a plexus of nerves that react specifically with monoclonal antibody raised against serotonin. We have shown that in areas where the ependyma is damaged, the supraependymal plexus also undergoes destruction and that retrograde degeneration of the neuron as far as the dorsal raphe nucleus also occurs. Although the work is as yet unfinished, we suggest that so…
