Bile is the major route of cholesterol excretion from the body. It is concentrated in the gallbladder, and often results in supersaturation of cholesterol. The high levels of cholesterol in gallbladder bile has clinical implications with respect to cholesterol gallstone formation and cholesterolosis of the gallbladder wall. Epstein-Barr virus (EBV) persistently infects B-cells in humans and can be shed into the saliva. The cellular source of infectious virus is uncertain. Hairy leukoplakia, an AIDS-associated lesion of the tongue, supports EBV replication in epithelial cells. However, the general significance of this observation has remained doubtful. The authors describe a technique with which one can perform electron microscopy on conjunctival epithelial cells acquired by impression cytology. With adjacent excisional and impression biopsies of conjunctival epithelium in patients having mucopolysaccharidoses (MPS), typical fibrillo-granular inclusions are shown. This noninvasive procedure has potential for the study of subcellular, cellular, and intercellular morphology in diseases of the ocular surface.

Biopsies from the oral mucosa of three patients (two with pemphigus vulgaris and one with pemphigus erythematosus) were taken for both freeze-fracturing and thin-sectioning studies. Cells obtained by tryptic digestion of the surface of intact adult and fetal Fischer 344 rat lungs were plated on glass fragments. Epithelial cell lines were readily established by selecting fragments with 2 to 10 cells 2 days after plating and growing them in F12 K media containing 10% fetal bovine serum (FBS). These cell lines and new lines that can be easily obtained a provide a reliable source of diploid, density-inhibited epithelial cells. These cells of mesothelial origin may serve as models for the study of mesothelial cells in situ.
