A convenient homogeneous enzyme immunoassay for estradiol is described. Unlike heterogeneous immunoassays, which require time-consuming separation steps or expensive automated systems, homogeneous immunoassays, wherein all reagents are freely suspended in bulk solution, can be simple and fast without costly instrumentation. Degradation of ethinyl estradiol (EE2) by nitrifying activated sludge was studied with micro-organisms grown in a reactor with feedback of sludge fed with only a mineral salts medium containing ammonium as the sole energy source. Ammonium was oxidised by this sludge at a rate of 50 mg NH4+ g(-1) DW h(-1). This activated sludge was also capable of degrading EE2 at a maximum rate of 1 microg g(-1) DW h(-1). The influence of two routes of estradiol administration on pineal melatonin production in postmenopausal women was investigated. Both transdermal and oral estradiol treatment led to an increase as well as decrease of melatonin production in different patients. During a three-year period, 217 ovarian cystic lesions were evaluated by both fine needle aspiration (FNA) cytology and estradiol (E2) assay at Royal Women's Hospital. One hundred three ovarian lesions were subsequently subjected to histologic examination, and 79 cyst fluids were assayed for progesterone. Of the 151 cystic lesions diagnosed by cytology/histology, 50 were follicular cysts, and 90% had E2 content > 20 nmol/L. Administration of exogenous estradiol caused embryos to develop ovaries rather than testes in alligators, softshell turtles, and leopard geckos. Alligators and leopard geckos are known to have environmental sex determination; softshell turtles lack both environmental sex determination and heteromorphic sex chromosomes. Sensitivity to gonadal feminization by estradiol thus appears to be widespread in amniotes.

Ovariectomized female hamsters received unilateral implants of estradiol aimed at the mesencephalic central gray (MCG). Mature male Sprague-Dawley rats were made surgically cryptorchid at 47--50 days of age. Animals were sacrificed as controls and at 7, 14, and 21 days post-surgery. The testes were removed and utilized to determine the effects of cryptorchidism on tissue weight loss, histological changes, and cytoplasmic estradiol binding capacity. A new extended-cycle OC that contains the lowest doses of ethinyl estradiol (EE) and levonorgestrel (LNG) + continuous EE throughout the cycle is now available.
