Application of quantitative ethanol detector (QED) test kit to measure ethanol concentration in blood samples.

In this paper, the applicability of the quantitative ethanol detector (QED) test kit for screening of ethanol concentrations in blood samples was investigated. The pretreatment of blood using the sulfosalicylic acid solution and the three-way stopcock followed by membrane filtration gave satisfactory results. The ethanol concentrations in whole blood samples (n=61) determined by QED correlated well with those determined by gas chromatography; the correlation coefficient indicated 0.990. Because a high correlation coefficient (0.928) was also confirmed in trial by investigators, QED test should…

Effects of ageing and intermittent ethanol exposure on rat locus coeruleus and ethanol-withdrawal symptoms.

In this study, the effects of ethanol and age on the morphology of the locus coeruleus (LC) and on the severity of ethanol-withdrawal symptoms were studied during a 5-week intermittent ethanol exposure. Young (3-4 months) and old (29-30 months) male Wistar rats were given highly intoxicating doses of ethanol by intragastric intubations for 4 days, followed by a 3-day ethanol-withdrawal period. This 7-day cycle of ethanol exposure and withdrawal was repeated five times. A non-treated group and a sucrose-fed group of both ages were used as control groups. The severity of ethanol-withdrawal sympt…

MK-801 administration during ethanol withdrawal in neonatal rat pups attenuates ethanol-induced behavioral deficits.

Alcohol exposure during development can produce central nervous system dysfunction, resulting in a wide range of behavioral alterations. The various mechanisms by which alcohol causes these behavioral changes, however, remain unknown. One mechanism that has been suggested is NMDA receptor-mediated excitotoxic cell death produced by ethanol withdrawal. The present study examined whether MK-801, an antagonist of the NMDA receptor that has been shown to protect against NMDA receptor-mediated excitotoxicity, could block alcohol's adverse effects on behavior. Sprague-Dawley rat pups were exposed to…

Comparison of ethanol metabolism in isolated periportal or perivenous hepatocytes: effects of chronic ethanol treatment.

Ethanol metabolism in rat hepatocytes isolated either from the periportal (pp) or the perivenous (pv) area by collagenase gradient perfusion was compared to reveal metabolic factors that could be associated with the development of perivenous alcoholic liver damage. Cells were also isolated from rats given ethanol (E) chronically by addition to the drinking fluid. One group (EM) received in addition the alcohol dehydrogenase inhibitor 4-methylpyrazole, which potentiated the ethanol treatment by causing sustained elevated diurnal blood ethanol levels. Fatty degeneration ensued in only one-third …

S-Allyl-L-cysteine sulfoxide, a garlic odor precursor, suppresses elevation in blood ethanol concentration by accelerating ethanol metabolism and preventing ethanol absorption from gut.

Alcoholic beverages are enjoyed together with meals worldwide, but their excessive intake is associated with an increased risk of various diseases. We investigated whether S-allyl-L-cysteine sulfoxide (ACSO), a sulfuric odor precursor of garlic, suppresses elevation in plasma ethanol concentration by accelerating ethanol metabolism and preventing ethanol absorption from the gut in rats. ACSO and garlic extract with a high ACSO content (Garlic-H) suppressed elevation in concentrations of ethanol and acetaldehyde in plasma and promoted the activities of alcohol dehydrogenase and aldehyde dehydro…

Ethanol intake in the juvenile, adolescent, and adult rat: effects of age and prior exposure to ethanol.

BACKGROUND: Initial ingestion of ethanol by naïve rats has seemed to decrease dramatically with age. During the preweanling period, infant rats consume large quantities of high concentrations of ethanol without initiating procedures, in some instances exceeding doses required for severe motor incoordination. During adulthood, however, initial ingestion of ethanol without initiation procedures is low and infrequent. In the present study, the ontogeny of ethanol intake was measured in juvenile, adolescent and adult rats using a technique [consume off the floor (COF)] similar to that used to stud…

Fetal or infantile exposure to ethanol promotes ethanol ingestion in adolescence and adulthood: a theoretical review.

BACKGROUND: Despite good evidence that ethanol abuse in adulthood is more likely the earlier human adolescents begin drinking, it is unclear why the early onset of drinking occurs in the first place. A review of experimental studies with animals complemented by clinical, epidemiologic and experimental studies with humans supports the idea that precipitating conditions for ethanol abuse occur well before adolescence, in terms of very early exposure to ethanol as a fetus or infant. Experimental studies with animals indicate, accordingly, that ethanol intake during adolescence or adulthood is pot…

Increased specificity of ethanol's discriminative stimulus effects in an ethanol-pentobarbital-water discrimination in rats.

Ethanol's modulation of a number of receptor systems results in a heterogeneous discriminative stimulus complex. A previous study found that these heterogeneous discriminative stimulus effects were seemingly diminished when rats were trained to discriminate ethanol (2.0 g/kg) from pentobarbital (10.0 mg/kg). The present experiment was designed to extend these findings by using a lower training dose of ethanol (1.0 g/kg). Adult male Long-Evans rats (n = 7) discriminated pentobarbital (10.0 mg/kg; intragastric (i.g.)) from ethanol (1.0 g/kg; i.g.) from water (2.3 ml; i.g.) in a 3 lever, food-rei…

Prenatal ethanol exposure: susceptibility to convulsions and ethanol's anticonvulsant effect in amygdala-kindled rats.

The present experiments assessed the effects of prenatal ethanol exposure on the susceptibility to convulsions and on the anticonvulsant effect of ethanol using the electrical kindling model of epilepsy in rats. Adult male Sprague-Dawley rats from prenatal ethanol (E), pair-fed (PF), and ad libitum-fed control (C) treatment groups were tested following the implantation of a stimulation electrode in the left amygdala complex. The same rats were tested in four consecutive experiments. Both E and PF rats showed a slightly slower rate of kindling than C rats, as measured by convulsion class but no…

Effects of pregnanolone and dehydroepiandrosterone on ethanol intake in rats administered ethanol or saline during adolescence.

BACKGROUND: Adolescent alcohol use may contribute to long-term changes in the receptors and neuroactive steroids that may mediate its effects and to subsequent alcohol abuse and dependence as an adult. Therefore, in this study, ethanol preference and intake as an adult were examined after adolescent ethanol or saline administration. In addition, ethanol intake in the same groups was examined after administration of 2 neuroactive steroids with modulatory effects at GABA(A) receptors. METHODS: Two groups of male Long-Evans rats were administered 15 intraperitoneal (i.p.) injections of either et…

Long lasting effects of rearing by an ethanol-consuming dam on voluntary ethanol consumption by rats.

For exposure to alcohol early in life to potentiate alcohol abuse in adolescence or adulthood, consequences of early exposure to alcohol must be of considerable duration. In two experiments using Norway rats as subjects, we examined effects of exposure during weaning to a dam consuming ethanol on adolescents' later affinity for ethanol. In a preliminary experiment, we offered rat pups a choice between 8% ethanol and water for 7 days immediately after they were weaned at 26 days of age. Pups whose dam had ingested 8% ethanol for 6 days either immediately or 1 week before we weaned them drank mo…

Extended blood ethyl glucuronide persistence after controlled ethanol administration: Implications for ethanol-negative delayed DUI cases.

Delayed blood sampling complicates forensic interpretation in driving-under-the-influence investigations when ethanol concentrations have declined below routine reporting thresholds. Blood ethyl glucuronide (EtG) may provide evidence of recent alcohol exposure, but its late-phase persistence under controlled conditions remains incompletely characterized. This secondary analysis evaluated blood EtG persistence following controlled administration of 1.0 g/kg ethanol in 34 participants using a predefined operational threshold of 0.09 mg/L and sampling through 15 h after completion of drinking. Th…

Dpp6 Homozygous Knockout Mice Exhibit Increased Ethanol Conditioned Place Preference and Acute Ethanol-Induced Anxiolytic Behavior.

The gene DPP6 has been associated with behavioral phenotypes of alcohol use disorder (AUD) in recent human genome wide association studies. To further assess the role of this gene in ethanol-related traits, we tested Dpp6 knockout (KO) mice for ethanol conditioned place preference (CPP), locomotor activity, and ethanol-induced anxiolysis. Male homozygous KO mice (HOM) showed greater preference for the ethanol-paired context compared to wild type littermates (WT) and heterozygous KO mice (HET), while female mice showed no genotypic difference. HOM of both sexes exhibited greater novelty-induced…

Effect of 5-HT 1b/1d agonist on ethanol withdrawal syndrome and ethanol withdrawal induced anxiety.

INTRODUCTION: Ethanol dependence and abuse is an important problem of public health worldwide and its withdrawal shows some severe behavioural complication. Management of ethanol withdrawal syndrome (EWS) is still a challenge, thus the presented report postulates the possible mechanism involved in the development of EWS. MATERIAL AND METHODS: EWS was induced by administration of ethanol for 21 days and 5-hydroxytryptamine (5-HT) 1b/1d agonist treated group receives Zolmitriptan (ZMT) at 30 mg/kg i.p. 30 min prior to ethanol withdrawal. The effect of 5-HT 1b/1d receptor agonist on EWS was dete…

Adolescent but not adult ethanol binge drinking modulates ethanol behavioral effects in mice later in life.

BACKGROUND: Alcohol use disorder is a serious illness marked by uncontrollable drinking and a negative withdrawal state when not using. Alcohol is one of the most commonly used drugs among adolescent populations. Given that adolescence is a unique developmental stage during which alcohol has long-term effects on future drug-taking behavior; it is essential to understand how early exposure to ethanol during adolescence may affect the abuse liability of the drug later in life. Our studies focused on characterizing how exposure to alcohol in adolescence alters later adult alcohol dependence behav…

Deciphering the relationship between vulnerability to ethanol-induced behavioral sensitization and ethanol consumption in outbred mice.

Ethanol (EtOH)-induced behavioral sensitization (EIBS) is proposed to play a role in early and recurring steps of alcohol dependence, but its impact on alcohol abuse is not clear. EIBS development is dependent upon animal species, strain and also individual factors. We proposed here to decipher the co-expression of EIBS and EtOH intake in individual animals among outbred Swiss mice, which exhibit heterogeneity that parallels what may occur in humans. To do so, mice were exposed to a two-bottle choice with free access to water or 10% EtOH for 6 days just before and immediately after chronic int…

Silver sub-nanoclusters electrocatalyze ethanol oxidation and provide protection against ethanol toxicity in cultured mammalian cells.

Silver atomic quantum clusters (AgAQCs), with two or three silver atoms, show electrocatalytic activities that are not found in nanoparticles or in bulk silver. AgAQCs supported on glassy carbon electrodes oxidize ethanol and other alcohols in macroscopic electrochemical cells in acidic and basic media. This electrocatalysis occurs at very low potentials (from approximately +200 mV vs RHE), at physiological pH, and at ethanol concentrations that are found in alcoholic patients. When mammalian cells are co-exposed to ethanol and AgAQCs, alcohol-induced alterations such as rounded cell morpholog…

Bolivia's Path to Lower Emissions: Sugar Cane Ethanol and GREET Model Insights into Ethanol-Gasoline Blends.

Bolivia's transition toward sustainable energy is advancing through the integration of sugar cane ethanol as a viable alternative to fossil fuels. This study presents the first gate-to-gate (GtG) life cycle assessment (LCA) of Bolivian ethanol production, developed using Aspen Plus v14 with a base unit of 1 MJ of energy. The assessment considers a sugar cane milling capacity of 23,000 tons per day and an annual production potential of 108 million liters of anhydrous ethanol. Distillation emerged as the most significant contributor to the GtG global warming potential (GWP) at 31.0 gCO2eq/L; how…

Ethanol inhibits dopamine uptake via organic cation transporter 3: Implications for ethanol and cocaine co-abuse.

Concurrent cocaine and alcohol use is among the most frequent drug combination, and among the most dangerous in terms of deleterious outcomes. Cocaine increases extracellular monoamines by blocking dopamine (DA), norepinephrine (NE) and serotonin (5-HT) transporters (DAT, NET and SERT, respectively). Likewise, ethanol also increases extracellular monoamines, however evidence suggests that ethanol does so independently of DAT, NET and SERT. Organic cation transporter 3 (OCT3) is an emergent key player in the regulation of monoamine signaling. Using a battery of in vitro, in vivo electrochemical…
