Blood donor exposome and impact of common drugs on red blood cell metabolism.

Computational models based on recent maps of the RBC proteome suggest that mature erythrocytes may harbor targets for common drugs. This prediction is relevant to RBC storage in the blood bank, in which the impact of small molecule drugs or other xenometabolites deriving from dietary, iatrogenic, or environmental exposures ("exposome") may alter erythrocyte energy and redox metabolism and, in so doing, affect red cell storage quality and posttransfusion efficacy. To test this prediction, here we provide a comprehensive characterization of the blood donor exposome, including the detection of co…

The hepato-exposome axis: How endocrine disruptors hijack liver receptors to drive MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is increasing worldwide at a pace that cannot be fully explained by obesity, diet, or genetics alone. Emerging evidence provides mechanistic insight into how defined endocrine-disrupting chemicals (EDCs) rewire hepatic transcriptional and metabolic networks relevant to MASLD. This review focuses on the molecular interfaces through which major EDC families intersect with hepatic metabolic regulation. We propose the "hepato-exposome axis" as a receptor-centric framework that maps specific EDC classes to nuclear receptors (NRs) and …

Exposome-induced dysregulation of glycemic homeostasis: Emerging biomarkers for diabetes risk and progression.

Environmental exposures throughout life profoundly influence the development and progression of diabetes mellitus. The exposome, representing the totality of environmental exposures from conception to adulthood, interacts with genetic and metabolic pathways, leaving measurable signatures termed biomarkers. These biomarkers encompass indicators of exposure, biological effect, and susceptibility, providing mechanistic insights into glycemic regulation and disease progression. This review synthesizes current evidence on exposome-linked biomarkers in both Type 1 and Type 2 diabetes, highlighting c…

The exposome-autoimmunity axis: environmental xenobiotics, gut dysbiosis, and potential pathways to immunosenescence.

The escalating incidence of autoimmune diseases cannot be fully explained by genetics alone. It is increasingly linked to the exposome-the cumulative burden of lifelong environmental exposures. This review examines how pollutants (particulate matter, microplastics, agrochemicals, heavy metals) reshape the gut microbiota-immune axis and disrupt intestinal homeostasis. This triggers profound dysbiosis, characterized by reduced commensal diversity and expanded pathobionts. We highlight three mechanisms driving pollution-induced immune reprogramming: (i) barrier compromise facilitating metabolic e…

Exposome-driven molecular sensitization signatures in pediatric rhinitis and asthma across Spanish regions.

BACKGROUND: Allergic rhinitis and asthma in childhood arise from dynamic interactions between genetic predisposition and the exposome. However, how environmental heterogeneity shapes molecular IgE sensitization profiles during early life remains insufficiently characterized. METHODS: We conducted a prospective, observational, multicentre, cross-sectional study including 354 children (0-15 years) with allergic rhinitis and/or asthma recruited across eight Spanish cities contrasting regional climatic contexts. All patients were skin prick test-positive to at least one aeroallergen and underwent…

Dietary Adjuvanticity in the Modern Plant Exposome: Implications for Immune-Mediated Inflammatory Diseases.

Immune-mediated inflammatory diseases (IMIDs) arise from interactions among genetic susceptibility, epithelial barrier function, microbiota, … heses. Overall, plant-derived exposures are best interpreted as potential modifiers within the IMID exposome, not as primary causes of autoimmunity. Testing this model will require defined exposures, food-matrix and processing studies, biomarkers of barrier and immune activation, patient stratification, and controlled human studies.

Comparing the Effects of Glyphosate and Mycotoxins in the Human Follicular Microenvironment: An Exploratory Exposome Study.

Background: Exposure to anthropogenic and/or natural (e.g., herbicides or mycotoxins) endocrine-disrupting chemicals (EDCs) has been linked to several reproductive disorders. Glyphosate (GLY), a common agricultural agent, is a potential element of the exposome that bioaccumulates and has potential endocrine and oxidative stress-related effects. However, data on its presence in the human ovarian microenvironment remain limited. Our study examined GLY levels in follicular fluid (ff) and serum and their relationships with oxidative stress markers, reproductive hormones, and stress hormones in wom…

Advancements in Assays for Micro- and Nanoplastic Detection: Paving the Way for Biomonitoring and Exposomics Studies.

Although plastic pollution and exposure to plastic-related compounds have received worldwide attention, health risks associated with micro- and nanoplastics (MNPs) are largely unknown. Emerging evidence suggests MNPs are present in human biofluids and tissue, including blood, breast milk, stool, lung tissue, and placenta; however, exposure assessment is limited and the extent of human exposure to MNPs is not well known. While there is a critical need to establish robust and scalable biomonitoring strategies to assess human exposure to MNPs and plastic-related chemicals, over 10,000 chemicals h

Chemical exposomics in biobanked plasma samples and associations with breast cancer risk factors.

BACKGROUND: The chemical exposome includes exposure to numerous environmental and endogenous molecules, many of which have been linked to reproductive outcomes due to their endocrine-disrupting properties. As several breast cancer risk factors, including age and parity, are related to reproduction, it is imperative to investigate the interplay between such factors and the chemical exposome prior to conducting large scale exposome-based breast cancer studies. OBJECTIVE: This pilot study aimed to provide an overview of the chemical exposome in plasma samples from healthy women and identify asso…

External and internal exposome as triggers of biological signalling in systemic sclerosis - A narrative synthesis.

Systemic sclerosis (SSc) is an autoimmune chronic connective tissue disorder with a complex pathogenesis and a strong gene-environment interaction. Despite the low prevalence of SSc, with around 100-250 cases per million, the morbidity and mortality are high and disproportionately affecting women. In this context, we review the influence of the external and internal exposome on the "immunome" in SSc. While several studies have addressed aspects of exposure-induced autoimmunity in general, very few have focused on SSc-specific phenotypes. In epidemiological studies, targeted characterization of…

Converging Neoplasms and the Environmental Exposome: Four Distinct Encounters with Collision Tumors in Dermatology.

Collision tumors - characterized by two or more distinct cell types within a singular lesion - are uncommon yet intriguing dermatological phenomena, presenting diagnostic and therapeutic enigmas. Our case series details four diverse presentations of such tumor intersections in dermatology. Beyond the individual cases, we embark on an exploration into the potential environmental exposome's role in the emergence of these neoplastic overlaps. While the first and fourth cases underscore serendipitous discoveries during an excisional biopsy, the second revolves around diagnostic ambiguity arising f…

Integrating Intersectionality Into the Exposome Paradigm: A Novel Approach to Racial Inequities in Uterine Fibroids.

Intersectionality is a critical theoretical framework that emphasizes the influence of intersecting systems of oppression on the lived experiences of people marginalized by inequity. Although applications of intersectionality are increasing in public health, this framework is absent in environmental health, which has instead focused on the exposome, a paradigm that considers the totality of an individual's environmental exposures across the life course.Despite advancements in the biological complexity of exposome models, they continue to fall short in addressing health inequities. Therefore, w…
