Macromolecular crowding is a defining feature of cellular environments and plays an important role in modulating protein aggregation. Here, we examine how mixed macromolecular crowders influence the aggregation of Huntingtin exon-1 protein with a pathogenic polyglutamine length (HD39Q). After disruption of the pancreas with collagenase, islets were isolated by Ficoll density gradient centrifugation, trypsinized to obtain individual cells, and plated in 96-well tissue culture trays. Myeloid and erythroid cultures were prepared from bone marrow aspirates taken from control and T. congolense infected neonatal and six-month-old calves. To study the potential roles of CD40L in immune responses, we generated CD40L-deficient mice by gene targeting.
