Fumonisin contamination of food: progress in development of biomarkers to better assess human health risks.

Fumonisins, fungal toxins produced by Fusarium moniliforme, contaminate maize based foods and feeds throughout the world. They cause liver and kidney toxicity in animals in addition to leukoencephalomalacia in horses and pulmonary edema in pigs. Fumonisin B(1) is carcinogenic in rats and mice. Ecological studies have linked consumption of fumonisin contaminated maize with oesophageal cancer in human populations in South Africa and China. This review discusses the potential health risks for people exposed to the fumonisins, and describes how mechanistic studies of toxicity in animal models have

Designer Enzymes: Food-System-Oriented Biocatalysts for Mitigating Mycotoxin Contamination and Reducing Postharvest Grain Loss.

Mycotoxin contamination remains a major challenge to food and feed safety, agricultural productivity, and postharvest resource utilization. Although numerous mycotoxin-transforming enzymes have been discovered and increasingly improved through protein engineering, only a limited number have progressed from laboratory characterization to practical food or feed applications. This translational gap arises because catalytic efficiency alone does not address matrix interference, processing stress, scalable production, formulation, degradation-product safety, and application-specific regulatory requ

Advancement in Technologies and Practices to Control Listeria monocytogenes Contamination within the Food Processing Environment.

In the expanding food processing environment, humans still face risks from certain harmful organisms that cause food contamination. One such ubiquitous organism is Listeria monocytogenes (L. monocytogenes), a facultative anaerobe, one of the infectious bacteria that can cause contamination in food processing settings. Food processing environments have inherent challenges in implementing preventive measures, including sanitation protocols, hygiene practices, advanced detection technologies and regulatory compliance. Advanced technologies and best practices are applied to control and prevent L. …

Highly Sensitive Detection of Food Contaminants Based on an Efficient Quadrupedal DNA Walker.

This research details the development of a highly efficient quadrupedal DNA walker designed for the detection of food contaminants. A cruciform DNA probe was synthesized through a straightforward methodology. Upon specific recognition between target analytes and aptamer sequences, the quadrupedal DNA walker is activated. This activation obviates the need for complex assembly procedures, thereby improving both operational simplicity and efficiency. Under equivalent conditions, the quadrupedal walker demonstrates a reduced balancing time and enhanced signal amplification efficiency. The proposed

Applications of NIR Spectroscopy and Chemometrics for Food Authentication and Safety: Detection of Adulterants and Contaminants.

Adulteration and contamination represent an ever-growing global problem, due to the emergence of increasingly sophisticated illicit commercial practices involving high-value and widely consumed products. Traditional chromatographic methods, such as liquid chromatography coupled with mass spectrometry, offer high sensitivity but their application is often limited by long analysis time, complex sample preparation, and the use of non-eco-friendly organic solvents. In contrast, enzymatic and immunoassay-based methods are rapid and require minimal sample preparation, but their applicability may be

Unravelling the Mechanistic Approach of Aflatoxin Contaminated Food on Neurodegenerative Diseases-A Novel Approach.

Aflatoxins (AFs) are a group of toxic secondary metabolites and a dietary toxin produced predominantly by Aspergillus species such as Aspergillus flavus and Aspergillus parasiticus. The four most common and harmful forms of AFs include Aflatoxin B1 (AFB1), Aflatoxin B2 (AFB2), Aflatoxin G1 (AFG1), and Aflatoxin G2 (AFG2), which pose a significant health threat due to their widespread contamination of food and feed products. Particularly, AFB1 has raised a major global health concern. Noxious neurological outcomes have been associated with chronic exposure to AF-contaminated food, contributing

Endonuclease V activated Pyrococcus furiosus Argonaute for the detection of food contaminated bacteria.

Pyrococcus furiosus Argonaute (PfAgo) is a novel programmable nuclease that has been used in nucleic acid detection due to its excellent performance. Traditional PfAgo based detection methods relies on the input of exogenous guide DNA (gDNA), which restricted its flexibility and universality in application. Here, we designed primers with deoxy inosine base which can be recognized and cleaved by endonuclease V, following turn into gDNA to activate PfAgo for target gene detection. Therefore, an Endo V activated PfAgo based nucleic acid detection (VPN) method was developed. The detection limit of

AMPK and metabolic disorders: The opposite roles of dietary bioactive components and food contaminants.

AMPK is a key player in a variety of metabolic and physiological processes, which might be considered one of the most promising targets for both prevention and treatment of metabolic syndrome and its associated diseases. Many dietary components and contaminants have been recently demonstrated to prevent or promote the development these diseases via AMPK-mediated pathways. AMPK can be activated by diverse phytochemical substances such as EGCG, chicoric acid, tomatidine, and others, all of which have been found to contribute to preventing or ameliorating chronic disorders. On the other hand, rec

Constitutively active microglial populations limit anorexia induced by the food contaminant deoxynivalenol.

Microglia are involved in neuroinflammatory processes during diverse pathophysiological conditions. To date, the possible contribution of these cells to deoxynivalenol (DON)-induced brain inflammation and anorexia has not yet been evaluated. DON, one of the most abundant trichothecenes found in cereals, has been implicated in mycotoxicosis in both humans and farm animals. DON-induced toxicity is characterized by reduced food intake, weight gain, and immunological effects. We previously showed that exposure to DON induces an inflammatory response within the hypothalamus and dorsal vagal complex

Interference on Iodine Uptake and Human Thyroid Function by Perchlorate-Contaminated Water and Food.

BACKGROUND: Perchlorate-induced natrium-iodide symporter (NIS) interference is a well-recognized thyroid disrupting mechanism. It is unclear, however, whether a chronic low-dose exposure to perchlorate delivered by food and drinks may cause thyroid dysfunction in the long term. Thus, the aim of this review was to overview and summarize literature results in order to clarify this issue. METHODS: Authors searched PubMed/MEDLINE, Scopus, Web of Science, institutional websites and Google until April 2020 for relevant information about the fundamental mechanism of the thyroid NIS interference indu

Listeria monocytogenes contamination of ready-to-eat foods and the risk for human health in the EU.

Food safety criteria for Listeria monocytogenes in ready-to-eat (RTE) foods have been applied from 2006 onwards (Commission Regulation (EC) 2073/2005). Still, human invasive listeriosis was reported to increase over the period 2009-2013 in the European Union and European Economic Area (EU/EEA). Time series analysis for the 2008-2015 period in the EU/EEA indicated an increasing trend of the monthly notified incidence rate of confirmed human invasive listeriosis of the over 75 age groups and female age group between 25 and 44 years old (probably related to pregnancies). A conceptual model was us

Single versus combined exposure of Hyalella azteca to zinc contaminated sediment and food.

The amphipod Hyalella azteca was exposed for 28 d to different combinations of Zn contaminated sediment and food. Sediment exposure (+clean food) resulted in increased Zn body burdens, increased mortality and decreased body mass when the molar concentrations of simultaneously extracted Zn were greater than the molar concentration of Acid Volatile Sulfide (SEM(Zn)-AVS>0), suggesting that dissolved Zn was a dominant route of exposure. No adverse effect was noted in the food exposure (+clean sediment), suggesting selective feeding or regulation. Combined exposure (sediment+food) significantly inc
