The clinical characteristics of women with galactorrhea and amenorrhea induced by drugs were studied. In addition, hyperprolactinemia was, experimentally, induced by sulpiride in women, and their endocrinological backgrounds were studied. The findings suggested that the mechanism of amenorrhea due to hyperprolactinemia was primarily the reduction of steroidogenesis in the ovary, and also the disturbance of positive feedback to the hypothalamus. One hundred women who had been employing combination estrogen/progestin oral contraceptives (OCs) for a minimum of six consecutive months were evaluated for the presence of galactorrhea. It was found in 9%. The incidence did not vary significantly with age, gravidity, duration of OC use of estrogen content. Two cases of craniopharyngioma presenting with amenorrhea-galactorrhea syndrome due to hyperprolactinemia are reported. After operation and irradiation, the tumor reduced markedly in size. Coincidental decrease in plasma prolactin level and restoration of menstruation seem to support the view that the hypothalamic prolactin inhibiting factor (PIF) had played an important role in hyperprolactinemia in these two patients.

A 36-year-old man had estrogen-induced galactorrhea and oligospermia. He had been taking an unknown amount of estrogen for a year. While hypogonadism, manifested by decreased libido and potency, was apparent at the time of estrogen administration, galactorrhea appeared two months after estrogen was discontinued.
