Streptococcus pyogenes are Gram-positive opportunistic pathogens residing in the human nasopharynx and skin. Changes in environmental conditions, such as pH, temperature and availability of essential ions, can stimulate the expression of S. pyogenes virulence factors. One such factor could be the availability of an extracellular pool of polyamines. Polyamines are synthesized from amino acids, and are universally present in the environment. Polyamines have been implicated in the ecology of pathogenesis by modulating quorum sensing, host adaptation and virulence. Polyamines mediate pathogenesis

Organohalide respiration is an environmentally important but poorly characterized type of anaerobic respiration. We compared the global proteome of the versatile organohalide-respiring Epsilonproteobacterium Sulfurospirillum multivorans grown with different electron acceptors (fumarate, nitrate, or tetrachloroethene [PCE]). The most significant differences in protein abundance were found for gene products of the organohalide respiration region. This genomic region encodes the corrinoid and FeS cluster containing PCE reductive dehalogenase PceA and other proteins putatively involved in PCE meta

Hematopoietic stem cells (HSCs) are pluripotent cells that give rise to all of the circulating blood cell types. Their unique ability to self-renew while generating differentiated daughter cells permits HSCs to sustain blood cell production throughout life. In mammals, the pool of HSCs shifts from early sites in the aorta-gonad-mesonephros region and the placenta to the fetal liver and ultimately bone marrow. During the past decade, a map of transcriptional activators and repressors that regulate gene expression in HSCs, their precursors and their progeny, at distinct stages of development has

The lambdaZAP II expressed genomic library of B. pseudomallei was screened with pooled melioidosis serum preabsorbed with E. coli host cell. The positive clones were detected by using protein A-CDP-star chemiluminescence. All of 14 positive clones reacted with only the pooled absorbed melioidosis serum and not the pooled absorbed normal serum when tested with the plaque dot blot analysis. The expressed genes were detected by using a combination of immunoscreening, bioinformatics and molecular biology. At least six in vivo expressed genes were identified by this approach. Two were well known vi

The mammalian ovary is unique in that its reproductive life span is limited by oocyte quantity and quality. Oocytes are recruited from a finite pool of primordial follicles that are usually exhausted from the ovary during midadult life. If regulation of this pool is perturbed, the reproductive capacity of the ovary is compromised. TAF4B is a gonad-enriched subunit of the TFIID complex required for female fertility in mice. Previous characterization of TAF4B-deficient ovaries revealed several reproductive deficits that collectively result in infertility. However, the etiology of such fertility

Solanum verrucosum Schlechtendal (2x = 2n = 24) is unique among the clade 4 Solanum Sect Petota species. In addition to being one of the only fully self-compatible diploid potato species, S. verrucosum is the only clade 4 species that lacks prezygotic interspecific reproductive barriers. This allows S. verrucosum to accept pollen from a broad range of Solanum species and thereby serving as a genetic "bridge" between the cultivated or primary potato gene pool and distantly related wild relatives in the tertiary gene pool. The genetic mechanisms underlying self-compatibility in Solanum often und…

Interspecific or introgressive hybridization is one of the driving forces in plant speciation, producing allopolyploids or diploids with rearranged genomes. The process of karyotype reshaping following homoploid interspecific hybridization has not been studied experimentally. Interspecific hybridization is widely used in plant breeding to increase genetic diversity and introgress new traits. Numerous introgression stocks were developed for hexaploid wheat Triticum aestivum L. (2n = 6x = 42, genome AABBDD). Double monosomic lines, containing one alien chromosome from the tertiary gene pool of w…

Lipoblastoma is a benign tumor of adipocytic differentiation originating from embryonal white fat that is commonly described in young children, with rare cases arising in adults. PLAG1 gene (8q11-13) rearrangement is a characteristic feature of lipoblastoma. However, HMGA2 gene rearrangement has also been reported in a few cases of lipoblastoma, with the gene fusion partners reported thus far being EP400, FGD6, and GSN genes. Herein, we report an HMGA2-rearranged lipoblastoma arising in the chin of a 51-year-old female with a novel gene fusion partner, the GRIP1 gene, that has not been previou

Large-scale, population-based genomic studies have provided a context for modern medical genetics. Among such studies, however, African populations have remained relatively underrepresented. The breadth of genetic diversity across the African continent argues for an exploration of local genomic context to facilitate burgeoning disease mapping studies in Africa. We sought to characterize genetic variation and to assess population substructure within a cohort of HIV-positive children from Botswana-a Southern African country that is regionally underrepresented in genomic databases. In brief: In brief The establishment of the primordial follicle pool governs female reproductive lifespan but is poorly understood in ruminants. This study provides the first single-cell atlas of goat ovarian development, revealing key cellular interactions and identifying LUZP2 as a candidate oocyte-associated regulator potentially involved in primordial follicle formation. Abstract: Mammalian primordial follicles, essential for female reproduction, form a finite nonrenewable pool perinatally, with species-specific developmental variations. This study employed single-cell RNA sequencing to ma

The role astrocytes play in brain development and function has garnered greater attention as the diversity of roles they are involved in has become apparent. We have previously shown that ethanol-exposed astrocytes alter neuronal neurite outgrowth in an in vitro co-culture system and that ethanol alters the astrocyte-produced extracellular matrix (ECM) in vitro, with similar alterations in vivo. In this study, we utilized the translating ribosome affinity purification (TRAP) procedure in Aldh1l1-EGFP/Rpl10a transgenic mouse primary cortical astrocyte cultures to transcriptionally and translati

Anthropogenic changes associated with urbanization can bring wildlife species into closer contact with livestock and humans, thereby increasing the risk of pathogen spillover. Recently, urban-adapted (synanthropic) hedgehog species, including Hemiechinus auritus (long-eared) and Paraechinus hypomelas (Brandt's), have been suggested as potential sources of zoonotic pathogens. The increasing number of tick-infested hedgehogs in urban areas and confirmed cases of human rickettsial infections in south-eastern Iran motivated this investigation to identify rickettsial species in ticks collected from

The appearance of human immunodeficiency virus type 1 (HIV-1) plasma viremia is associated with progression to symptomatic disease and CD4+ T cell depletion. To locate the source of systemic viremia, this study employed a novel method to trace HIV-1 infection in vivo. We created JRCSFξnef, a pool of infectious HIV-1 (strain JR-CSF) with highly mutated nef gene regions by random mutagenesis PCR and infected this mutated virus pool into both Jurkat-CCR5 cells and hematopoietic stem cell-transplanted humanized mice. Infection resulted in systemic plasma viremia in humanized mice and viral RNA seq

BACKGROUND: The continuously growing mouse incisor serves as a model for stem cell-driven tooth renewal. Although mice possess maxillary and mandibular incisors, most studies have focused on the mandible. Potential differences in structure and renewal between these serially homologous organs remain insufficiently characterized.
RESULTS: Mandibular incisors are longer and contain greater enamel volume, mineral density, and mineral content than maxillary incisors, whereas maxillary incisors are more curved and display a larger cross-sectional area. Mineral density increases along the apical-to-

ARMC5 is implicated in several pathological conditions, but its function remains unknown. We have previously identified CUL3 and RPB1 (the largest subunit of RNA polymerase II (Pol II) as potential ARMC5-interacting proteins. Here, we show that ARMC5, CUL3 and RBX1 form an active E3 ligase complex specific for RPB1. ARMC5, CUL3, and RBX1 formed an active E3 specific for RPB1. Armc5 deletion caused a significant reduction in RPB1 ubiquitination and an increase in an accumulation of RPB1, and hence an enlarged Pol II pool in normal tissues and organs. The compromised RPB1 degradation did not cau

Janus kinase (JAK) inhibitors are widely used in the treatment of multiple autoimmune and inflammatory diseases. Immunologic and transcriptomic profiling have revealed major alterations on natural killer (NK) cell homeostasis associated with JAK inhibitions, while information on other innate lymphoid cells (ILCs) is still lacking. Herein, we observed that, in mice, the homeostatic pool of liver ILC1 was less affected by JAK inhibitors compared to the pool of NK cells present in the liver, spleen and bone marrow. JAK inhibition had overlapping effects on the transcriptome of both subsets, mainl
