Wastewater treatment plants have been highlighted numerous times as hotspots for anthropogenic environmental pollution with antibiotic resistance genes and resistant bacteria. However, demonstrating horizontal gene transfer events in these environments has been challenging. The role of SOS response, that has been shown to promote horizontal gene transfer through other mechanisms such as transduction, is often overlooked. In the present study, we evaluated whether SOS response could be induced in influent and effluent samples from wastewater treatment plants and compared them to reference point

Bacteria have evolved a wide array of immune systems to detect and defend against external threats including mobile genetic elements (MGEs) such as bacteriophages, plasmids, and transposons. MGEs are often selfish, exploiting their bacterial hosts to propagate, however they can also provide adaptive advantages through horizontal gene transfer. Gene transfer agents (GTAs), which are non-infectious domesticated prophages, represent a unique class of beneficial MGEs that facilitate bacterial gene transfer. Despite their domestication, GTAs retain phage-like features, including the requirement for

The Clp protease regulates antibiotic resistance, biofilm formation, stress response, and virulence in microorganisms through a post-transcriptional mechanism. Substrate recognition by the ClpS subunit is essential for the target protein degradation. However, the function of ClpS in Riemerella anatipestifer (R. anatipestifer) remains unclear. In this study, a clpS gene (B739_1609) in-frame deletion strain of R. anatipestifer was constructed using homologous recombination. The ΔclpS gene mutant exhibited no influence on the growth rate and shape of R. anatipestifer. However, the survival rate w

Gene expression heterogeneity has an impact on human bacterial pathogens responses, including antimicrobial resistance genes expression. Promoter region variability in resistance genes may include different regulatory boxes involved in metabolic processes essential for bacterial survival. In this study, a molecular tool was developed to characterize promoter region variability and resistance gene expression. For these, 46 bacterial clinical isolates were used. Sequence analysis of resistance genes promoter region showed the existence of different variants for each promoter and identified gene-

Co-localization of spatial transcriptome information of host and pathogen can revolutionize our understanding of microbial pathogenesis. Here, we aimed to demonstrate that customized bacterial probes can be successfully used to identify host-pathogen interactions in formalin-fixed-paraffin-embedded (FFPE) tissues by probe-based spatial transcriptomics technology. We analyzed the spatial gene expression of bacterial transcripts with the host transcriptomic profile in murine lung tissue chronically infected with Mycobacterium abscessus embedded in agar beads. Customized mycobacterial probes were

Gene regulatory networks allow organisms to generate coordinated responses to environmental challenges. In bacteria, regulatory networks are re-wired and re-purposed during evolution, though the relationship between selection pressures and evolutionary change is poorly understood. In this study, we discover that the early evolutionary response of Escherichia coli to the antibiotic trimethoprim involves derepression of PhoPQ signaling, an Mg2+-sensitive two-component system, by inactivation of the MgrB feedback-regulatory protein. We report that derepression of PhoPQ confers trimethoprim-tolera

Klebsiella pneumoniae (Kp) is one of the most important nosocomial pathogens worldwide, able to cause multiorgan infections and hospital outbreaks. One of the most widely disseminated lineage of Kp is the clonal group 258 (CG258), which includes the highly resistant "high-risk" sequence types ST258 and ST11. Genomic investigations revealed that very large recombination events have occurred during the emergence of Kp lineages. A striking example is provided by ST258, which has undergone a recombination event that replaced over 1 Mb of the genome with DNA from an unrelated Kp donor. Although sev

Recently, the biased β2-adrenoceptor/β-arrestin pathway was shown to play a pivotal role in crossing of the blood brain barrier by Neisseria meningitidis. We hypothesized that genetic variation in the β2-adrenoceptor gene (ADRB2) may influence susceptibility to bacterial meningitis. In a prospective genetic association study we genotyped 542 patients with CSF culture proven community acquired bacterial meningitis and 376 matched controls for 2 functional single nucleotide polymorphisms in the β2-adrenoceptor gene (ADRB2). Furthermore, we analyzed if the use of non-selective beta-blockers, whic

The acquisition and incorporation of genetic material between nonmating species, or horizontal gene transfer (HGT), has been frequently described for phylogenetically related organisms, but far less evidence exists for HGT between highly divergent organisms. Here we report the identification and characterization of a horizontally transferred fragment of the human long interspersed nuclear element L1 to the genome of the strictly human pathogen Neisseria gonorrhoeae. A 685-bp sequence exhibiting 98 to 100% identity to copies of the human L1 element was identified adjacent to the irg4 gene in so

As diagnostic and surveillance activities are vital to determine measures needed to control antimicrobial resistance (AMR), new and rapid laboratory methods are necessary to facilitate this important effort. DNA microarray technology allows the detection of a large number of genes in a single reaction. This technology is simple, specific and high-throughput. We have developed a bacterial antimicrobial resistance gene DNA microarray that will allow rapid antimicrobial resistance gene screening for all Gram-positive and Gram-negative bacteria. A prototype microarray was designed using a 70-mer b

Chronic non-bacterial osteomyelitis (CNO) is an inflammatory, non-infectious disorder of the skeletal system with unknown etiology. Besides bone-inflammation, patients may present with inflammatory involvement of other tissues. Chronic recurrent multifocal osteomyelitis (CRMO) is the most severe form of CNO. We describe the occurrence of Crohn's disease (CD) in four patients, previously diagnosed with CRMO. Mutations in CARD15, encoding the NOD2 protein, have recently been found in patients with CD. Based on the occurence of CNO and CD in these four and several reported patients, we hypothesiz

Consecutively collected synovial fluids were examined for presence of bacterial DNA (a 700-bp fragment of the bacterial 23S rRNA gene) followed by DNA sequencing of amplicons, and by conventional bacteriological methods. One or more microorganisms were identified in 22 of the 227 synovial fluids (9,7%) originating from 17 patients. Sixteen of the patients had clinical signs of arthritis. For 11 patients molecular and conventional bacterial examinations were in agreement. Staphylococcus aureus, Streptococcus dysgalactiae subspecies equisimilis and Streptococcus pneumoniae, were detected in syno

Extract: The exponential outburst of knowledge in molecular immunology has provided us with an in depth insight into the biological activity of cytokines. These are small, freely diffusible proteins that, together with numerous growth factors and chemokines, act as messengers by which cells of the immune system communicate with each other and with most other tissues in the body. As such, these molecules are able to regulate many aspects of the immune response in which numerous cells and tissues may be involved at any one time. Most often, cytokines are active in extremely low concentrations. I

A novel pathway for degradation of the cyanobacterial heptapeptide hepatotoxin microcystin LR was identified in a newly isolated Sphingomonas sp. (Bourne et al. 1996 Appl. Environ. Microbiol. 62: 4086-4094). We now report the cloning and molecular characterisation of four genes from this Sphingomonas sp. that exist on a 5.8-kb genomic fragment and encode the three hydrolytic enzymes involved in this pathway together with a putative oligopeptide transporter. The heterologously expressed degradation pathway proteins are enzymatically active. Microcystinase (MlrA), the first enzyme in the degrada

Growth of Salmonella typhimurium in a low-oxygen environment induces the ability of these bacteria to enter mammalian cells. We have carried out a search for invasion genes that are expressed under low-oxygen conditions by using Tn5lacZY transcriptional fusions. Several noninvasive oxygen-regulated lacZY insertion strains have been identified. The invasion defect in one of these noninvasive S. typhimurium strains, BJ66, has been complemented by introduction of a cosmid (pBDJ125) from an S. typhimurium SL1344 gene bank. A 1.9-kb EcoRV DNA fragment subcloned from this cosmid, containing a single

Human faecal waste can harbour antibiotic residues, creating reservoirs that sustain and spread antimicrobial resistance (AMR). Black soldier fly larvae (BSFL, Hermetia illucens) bioconversion has emerged as a promising strategy for processing organic waste, particularly in settings with inefficient or absent faecal waste management. However, its effects on AMR in human faecal waste remain poorly characterised. In this study, we investigated ARG dynamics and bacterial community composition during BSFL bioconversion of human faecal waste using real-time qPCR and 16S rRNA gene amplicon sequencin

UNLABELLED: Wastewater treatment plants are considered disseminators of antibiotic resistance bacteria (ARB) and antibiotic resistance genes (ARGs). In the West Bank, Palestine, there are four working wastewater treatment plants (WWTPs) located in Jenin, Tulkarem, Ramallah (Al-Terah), and Al-Bierh. Most of which have secondary treatment facilities that depend on the activated sludge process. In this study, Illumina high-throughput sequence analysis was used to determine the profile of ARB and ARGs in Al-Bierh and Al-Terah WWTPs. Raw wastewater sample (influent) and secondary treated water samp

Rapid diagnosis is vital for patients with sepsis. Therefore, this study was conducted to assess the diagnostic performance of rapid Real-Time PCR (rRT-PCR), which detects 32 pathogens and common resistance genes in whole blood within four hours. This study included 160 adult patients with sepsis. Whole-blood samples and BCs were collected from all patients simultaneously, and causative microorganisms were investigated using an rRT-PCR panel and an automated BC system. The panel was also used to detect common resistance genes. The microorganisms most frequently isolated from the BCs were Staph

The release of pathogens and DNA from the cryosphere (glacier, permafrost, and, sea ice) has become a new threat to society and environment. Due to enhanced glacier retreat, the size of glacier forefields has greatly expanded. Herein, we used a combination of metagenomic and metatranscriptomic methods and adopted a sequence-based approach to investigate the distribution and changing patterns of virulence factor genes (VFGs) and antibiotic resistance genes (ARGs) in two glacier forefields. The forefields are separated by approximately 400 km located in the center and north of the Tibetan Platea

The phycosphere is an essential ecological niche for the proliferation of antibiotic resistance genes (ARGs). However, how ARGs' potential hosts change and the driving mechanism of metabolites under antibiotic stress in the phycosphere have seldom been researched. We investigated the response of Chlorella pyrenoidosa and the structure and abundance of free-living (FL) and particle-attached (PA) bacteria, ARGs, and metabolites under sulfadiazine by using real-time quantitative PCR, 16 S rRNA high-throughput. The linkage of key bacterial communities, ARGs, and metabolites through correlations wa
