Differential accumulation profiles of hordatine glucosides and aglycons during seedling and seed maturation stages in barley.

Hordatines, dehydrodimers of hydroxycinnamoylagmatines, are antifungal compounds that accumulate abundantly in young barley (Hordeum vulgare) seedlings. In addition to the aglycons, several hordatine glucosides have been isolated. However, detailed accumulation profiles of the hordatine glucosides and aglycons remain poorly understood. In this study, we identified the glucosides of hordatines A, B, and C in barley and examined the accumulation profiles of both the glycosides and aglycons during seedling and seed maturation stages. Aglycons accumulated predominantly in shoots, whereas glucoside…

Repairing effect of total glucosides of Paeonia on deoxynivalenol-induced enterohepatic axis damage in weaned piglets.

This study examined the protective effect of total glucosides of Paeonia (TGP) against deoxynivalenol (DON)-induced liver and intestinal injury in weaned piglets. Eighteen 21-day-old piglets (mean body weight ∼7 kg) were randomly assigned to three groups: control (CON, basal diet), DON (DON-contaminated diet), and DON+TGP (DON-contaminated diet supplemented with 200 mg/kg TGP), with six replicates per group and one piglet per replicate. Compared with the CON group, DON exposure significantly decreased serum antioxidant levels. Supplementation with TGP restored serum total antioxidant capacity …

Synthesis of eugenol-derived glucosides and evaluation of their ability in inhibiting the angiotensin converting enzyme.

We report here a series of glucosides which are active as inhibitors of the angiotensin converting enzyme (ACE). They are structurally related to the natural compound eugenol and exhibited significant inhibition values. Their syntheses were expeditious and we could obtain informative docking plots of them complexed to this enzyme. A glucoside derived from eugenol, carrying a carboxylic group in the aglycone, was the most active of them (with an IC50 of 0.4 mM) and showed good binding energies in docking studies with ACE. Moreover, computational prediction of toxicity risks, physicochemical pro…

Identification of rice Os4BGlu13 as a β-glucosidase which hydrolyzes gibberellin A4 1-O-β-d-glucosyl ester, in addition to tuberonic acid glucoside and salicylic acid derivative glucosides.

Gibberellin 1-O-β-d-glucose ester hydrolysis activity has been detected in rice seedling extracts, but no enzyme responsible for this activity has ever been purified and identified. Therefore, gibberellin A4 glucosyl ester (GA4-GE) β-d-glucosidase activity was purified from ten-day rice seedling stems and leaves. The family 1 glycoside hydrolase Os4BGlu13 was identified in the final purification fraction. The Os4BGlu13 cDNA was amplified from rice seedlings and expressed as an N-terminal thioredoxin-tagged fusion protein in Escherichia coli. The purified recombinant Os4BGlu13 protein (rOs4BGlu

Toxicokinetics, Biotransformation, Excretion, and Tissue Residue of Zearalenone-14-Glucoside in Male Sheep.

Zearalenone-14-glucoside (Z14G) is a common modified mycotoxin attracting considerable attention due to its high occurrence in feed and food, and its potential to transform into the parent form zearalenone (ZEN) in vivo. However, the toxicokinetic data in ruminants remain scarce. In this study, male sheep (n = 5) were orally administered a single dose of Z14G at 2 mg·kg-1 BW. Serum, rumen fluid, urine, and fecal samples were collected at designated time points, and tissue samples were obtained at slaughter 168 h post-dosing. Concentrations of Z14G, ZEN, α-zearalenol (α-ZOL), β-zearalenol (β-ZO

Cyanidin-3-glucoside: targeting atherosclerosis through gut microbiota and anti-inflammation.

With the shifting global disease spectrum, atherosclerosis (AS) has emerged as a leading contributor to mortality worldwide, with associated cardiovascular diseases (CVDs) representing the predominant cause of death. AS, a chronic inflammatory pathology, is mechanistically linked to oxidative stress and gut microbiota dysbiosis, which drive excessive reactive oxygen species (ROS) production and elevated levels of pro-inflammatory cytokines. Dietary polyphenols, particularly anthocyanins, are well-characterized for their dual role in modulating gut microbial communities and ameliorating chronic

Diagnostic meaning of urinary ethyl glucoside concentrations in relationship to alcoholic beverage consumption.

Incidents and accidents often involve the drinking of alcoholic beverages. We investigated compounds that indicate the consumption of alcoholic beverages even after ethanol (EtOH) becomes undetectable in blood and urine. Ethyl glucoside (EG) has been isolated as a possible drinking marker, and a GC-MS/MS method for EG isomers has been developed. EG isomers in several alcoholic beverages were analyzed. In sake, only αEG was observed in high concentrations. In wine and beer, both α and βEG were detected. Whisky, however, did not contain EG. EtOH and EG concentrations were analyzed in urine up to

Development of GLUT1-targeting alkyl glucoside-modified dihydroartemisinin liposomes for cancer therapy.

The antitumor activity of artemisinin derivatives has attracted much attention. However, lack of tumor targeting limits the anti-tumor activity of artemisinin derivatives. It is reported that tumor cells acquire energy through the glycolysis pathway. To meet their elevated glucose requirements, high expressions of glucose transporters (GLUTs) are observed in many malignant cells. On this basis, novel alkyl glycoside-modified dihydroartemisinin liposomes were successfully prepared with GLUT1 as the target and the glucose segment of an alkyl glycoside as the targeting head on the surface of lipo

Reconfigured Cyanogenic Glucoside Biosynthesis in Eucalyptus cladocalyx Involves a Cytochrome P450 CYP706C55.

Cyanogenic glucosides are a class of specialized metabolites widespread in the plant kingdom. Cyanogenic glucosides are α-hydroxynitriles, and their hydrolysis releases toxic hydrogen cyanide, providing an effective chemical defense against herbivores. Eucalyptus cladocalyx is a cyanogenic tree, allocating up to 20% of leaf nitrogen to the biosynthesis of the cyanogenic monoglucoside, prunasin. Here, mass spectrometry analyses of E. cladocalyx tissues revealed spatial and ontogenetic variations in prunasin content, as well as the presence of the cyanogenic diglucoside amygdalin in flower buds …

Intestinal absorption of luteolin and luteolin 7-O-beta-glucoside in rats and humans.

In this study, we investigated the intestinal absorption of luteolin and luteolin 7-O-beta-glucoside in rats by HPLC. The absorption analysis using rat everted small intestine demonstrated that luteolin was converted to glucuronides during passing through the intestinal mucosa and that luteolin 7-O-beta-glucoside was absorbed after hydrolysis to luteolin. Free luteolin, its conjugates and methylated conjugates were present in rat plasma after dosing. This suggests that some luteolin can escape the intestinal conjugation and the hepatic sulfation/methylation. LC/MS analysis showed that the main

The therapeutic potential of cyanidin-3-O-glucoside relating to female reproductive health.

Cyanidin-3-O-glucoside (C3G), a dietary flavonoid found in berries, exhibits strong antioxidant, anti-inflammatory, and anticancer properties. It plays a role in female reproductive health by protecting ovarian cells from oxidative stress while inhibiting tumour growth and inducing apoptosis in ovarian and cervical cancer cells. C3G can modulate estrogen receptors, growth factors, and apoptosis- and angiogenesis-related pathways. Its antioxidant and anti-inflammatory properties may be beneficial in hormone-related reproductive disorders and in oncological conditions of reproductive organs, suc
