BACKGROUND: Coagulase-negative staphylococci (CoNS) are a major cause of nosocomial blood stream infection, especially in critically ill and haematology patients. CoNS are usually multidrug-resistant and glycopeptide antibiotics have been to date considered the drugs of choice for treatment. The aim of this study was to characterize CoNS with reduced susceptibility to glycopeptides causing blood stream infection (BSI) in critically ill and haematology patients at the University Hospital Tor Vergata, Rome, Italy, in 2007.
METHODS: Hospital microbiology records for transplant haematology and IC…

Today, when the number of high-risk patients is increasing, special attention should be paid to polyresistant gram-positive microorganisms s … d, cerebrospinal fluid, urine) from high-risk patients increased 2-3 times during two recent years. Glycopeptides vancomycin and teicoplanine are drugs of choice for the treatment of infections caused by such microorganisms (sepsis, endocarditis, osteomyelitis, pneumonia, etc.). These drugs should be listed among obligatory antibiotics for resuscitation and int …

The simultaneous enrichment of glycopeptides and phosphopeptides from complex peptide mixtures remains challenging because boronate affinity and immobilized metal ion affinity chromatography (IMAC) typically require different pH conditions for efficient binding. We evaluated the previously reported dual boronate-Ti(IV) magnetic nanomaterial, Mag@DBA-Ti(IV), as a sample-preparation platform for the single-sorbent, single-loading-condition enrichment and sequential elution of glycopeptides and phosphopeptides. The material, prepared by co-immobilizing a low-pKₐ boronic acid ligand and alendronat…

Many therapeutic enzymes for lysosomal storage diseases require a high content of mannose-6-phosphate (M6P) glycan, which is important for cellular uptake and lysosomal targeting. We constructed glyco-engineered yeast harboring a high content of mannosylphosphorylated glycans, which can be converted to M6P glycans by uncapping of the outer mannose residue. In this study, the cell wall of this yeast was employed as a natural M6P glycan source for conjugation to therapeutic enzymes. The extracted cell wall mannoproteins were digested by pronase to generate short glycopeptides, which were further…

Protecting-group (PG)-free S-glycosylation offers an attractive strategy for glycopeptide synthesis, yet practical methods for complex peptides remain limited. Here, we report a non-photochemical S-glycosylation strategy that directly couples unprotected glycosyl donors with unprotected cysteine-containing peptides, enabling efficient access to β-S-glycopeptides. The method shows broad compatibility with glycosyl donors and peptide substrates and enables direct glycosylation of a structurally complex 37-mer precursor bearing one free cysteine, four protected cysteines, and oxidation-sensitive …

Glycopeptide antibiotics (GPAs) are the drug of choice for treating multidrug-resistant Gram-positive infections. GPAs possess a unique aromatic amino-acid-rich scaffold interconnected by a diverse set of biaryl cross-links, contributing to their rigid 3D structures for specific target binding on the cell wall. Disruption of the P450 genes responsible for biaryl linkage formation from the biosynthetic gene cluster of varsomycin identified a never-before-seen macrocyclic ring system in GPA, which greatly expands the chemical space of GPAs.

Up to 75% of bladder cancer patients suffer from recurrence due to postoperative tumor implantation. However, clinically used Bacillus Calmette-Guerin (BCG) treatment failed to inhibit the recurrence. Here, we report a bispecific glycopeptide (bsGP) that simultaneously targets CD206 on tumor-associated macrophages (TAMs) and CXCR4 on tumor cells. bsGP repolarizes protumoral M2-like TAMs to antitumor M1-like that mediated cytotoxicity and T cell recruitment. Meanwhile, bsGP is cleaved by the MMP-2 enzyme to form nanostructure for the long-term inhibition of CXCR4 downstream signaling, resulting

Our results show that the enhanced binding of oritavancin to PG over the binding of the other complexes studied is due to an increase in the hydrophobic effect, electrostatic and van der Waals interactions, and not the average number of hydrogen bo …

Glycopeptide antibiotics (GPAs) are last defense line drugs against multidrug-resistant Gram-positive pathogens. Natural GPAs teicoplanin and vancomycin, as well as semisynthetic oritavancin, telavancin, and dalbavancin, are currently approved for clinical use. Although these antibiotics remain efficient, emergence of novel GPA-resistant pathogens is a question of time. Therefore, it is important to investigate the natural variety of GPAs coming from so-called "rare" actinobacteria. Herein we describe a novel GPA producer-Nonomuraea coxensis DSM 45129. Its de novo sequenced and completely asse

Glycopeptides are a class of molecules that comprise two distinct families of biologically important scaffolds, peptides and oligosaccharides, each playing important roles in cellular communication and signaling. Rarely are small, endogenous secreted glycopeptides found that have significant impact on the progression of a specific disease state, but such is the case for the antiproliferative factor (APF) found in the urine and tissue of patients with the poorly understood bladder diseases collectively referred to as interstitial cystitis (IC). APF is a 9-mer peptide containing a sialylated O-l…

We report the cloning, expression, and characterization of a novel member of the mammalian UDP-GalNAc:polypeptide N-acetylgalactosaminyltransferase (ppGaNTase) family that transfers GalNAc to a GalNAc-containing glycopeptide. Northern blot analysis revealed that the gene encoding this enzyme, termed ppGaNTase-T6, is expressed in a highly tissue-specific manner. Significant levels of transcript were found in rat and mouse sublingual gland, stomach, small intestine, and colon; trace amounts were seen in the ovary, cervix, and uterus. Recombinant constructs were expressed transiently in COS7 cell

Patients with mannosidosis, an inherited deficiency of lysosomal alpha-mannosidase, accumulate large amounts of mannose-rich oligosaccharides (the "core" of the carbohydrate units of many glocoproteins) in brain and liver and excrete these partial degradation products in their urine. A profound alpha-mannosidase deficiency was demonstrated in fibroblasts cultured from a skin biopsy obtained from a child with mannosidosis. Further, abnormal glycopeptides rich in mannose and similar to oligosaccharides found in the patient's urine were isolated from fibroblast extracts by a variety of chromatogr…

In this work, poly(glycidyl methacrylate-ethyleneglycol dimethacrylate) monolith functionalized with copper tetra(N-carbonylacrylic) aminephthalocyanine and iminodiacetic acid was successfully synthesized. Owing to hydrogen bonding and hydrophilic interactions, the monolith exhibited good performance for glycopeptide enrichment. When the tryptic digests of horseradish peroxidase were enriched by the developed monolith, a total of 20 glycopeptides could be captured and identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry analysis with a detection limit as l…

One of the promising directions of the combined approach is the design of dual-acting antibiotics - heterodimeric structures on the basis of antimicrobial agents of different classes. In this study a novel series of azithromycin-glycopeptide conjugates were designed and synthesized. The structures of the obtained compounds were confirmed using NMR spectroscopy and mass spectrometry data including MS/MS analysis. All novel hybrid antibiotics were found to be either as active as azithromycin and vancomycin against Gram-positive bacterial strains or have superior activity in comparison with their

Strain D958, a methicillin-resistant Staphylococcus aureus strain with reduced susceptibility to vancomycin, was isolated from a 69-year-old Saudi male patient presenting with severe sepsis immediately after admission. Despite high serum levels of vancomycin, the same S. aureus strain was isolated from five blood culture sets during 1 week. Treatment failure under therapeutic levels of vancomycin prompted us to investigate the resistance profile of this strain in further detail. The MIC values for vancomycin as determined by Etest and microdilution were 3.0 and 2.0 mg/liter, respectively, and

The fate of viral glycopeptides as cytotoxic T lymphocyte (CTL) epitopes is unclear. We have dissected the mechanisms of antigen presentation and CTL recognition of the peptide GP392-400 (WLVTNGSYL) from the lymphocytic choriomeningitis virus (LCMV) and compared them with those of the previously reported GP92-101 antigen (CSANNSHHYI). Both GP392-400 and GP92-101 bear a glycosylation motif, are naturally N-glycosylated in the mature viral glycoproteins, bind to major histocompatibility complex H-2D(b) molecules, and are immunogenic. However, post-translational modifications differentially affec…

Aberrantly glycosylated membrane proteins represent promising targets for cancer immunotherapy. However, glycan diversity and heterogeneity pose a challenge to developing effective vaccines against tumors expressing different glycoantigens. To broaden the immune response and enhance the vaccine's efficacy by targeting a wide variety of glycosylation patterns on tumor cells, herein we developed a Mosaic glycopeptide vaccine by simultaneously presenting three different glycopeptides on the carrier protein tetanus toxoid (TT). Immunological evaluation revealed that the Mosaic vaccine elicited hig…
